    Abstract: Ticks are the most important obligate blood-feeding vectors of human pathogens. With the advance of high-throughput sequencing, more and more bacterial community and virome in tick has been reported, which seems to pose a great threat to people. Methods A total of 14 skin specimens collected from tick-bite patients with mild to severe symptoms were analyzed through meta-transcriptomic sequencings. Results Four bacteria genera were both detected in the skins and ticks, including Pseudomonas, Acinetobacter, Corynebacterium and Propionibacterium, and three tick-associated viruses, Jingmen tick virus (JMTV), Bole tick virus 4 (BLTV4) and Deer tick mononegavirales-like virus (DTMV) were identified in the skin samples. Except of known pathogens such as pathogenic rickettsia, Coxiella burnetii and JMTV, we suggest Roseomonas cervicalis and BLTV4 as potential new agents amplified in the skins and then disseminated into the blood. As early as 1 day after a tick-bite, these pathogens can transmit to skins and at most four ones can co-infect in skins. Discussion Advances in sequencing technologies have revealed that the diversity of tick microbiome and virome goes far beyond our previous understanding. This report not only identifies three new potential pathogens in humans but also shows that the skin barrier is vital in preventing horizontal transmissions of tick-associated bacteria or virus communities to the host. It is the first research on patients’ skin infectome after a tick bite and demonstrates that more attention should be paid to the cutaneous response to prevent tick-borne illness.

    Abstract: We launched this prospective phase II single-arm trial on the combination of moderately hypo-fractionated radiotherapy and S-1, to explore the safety and efficacy of the new potent regimen in inoperable locally advanced esophageal squamous carcinoma (LA-ESCC) patients. Methods Patients with unresectable stage II-IVB LA-ESCC (UICC 2002, IVB only with metastatic celiac or supraclavicular lymph nodes) were included. Moderately hypofractionated radiotherapy (60Gy in 24 fractions) concurrent with S-1 was delivered. Meanwhile, gastrostomy tube placement by percutaneous endoscopic gastrostomy (PEG) was performed to provide nutritional support. Nutritional supplements were prescribed to meet requirements. The study outcomes included objective response rate (ORR), progression-free survival (PFS), overall survival (OS), locoregional progression-free survival (LRPFS), distant metastasis-free survival (DMFS), failure pattern, toxicities, nutritional status and treatment compliance. Endoscopy was routinely performed during post-treatment follow-up. Results Fifty-eight patients were included with a median follow-up of 24.4 months. The median age was 63 years (range 49-83 years) and 42 patients (72.4%) had stage III or IV diseases. The ORR was 91.3% and the CR rate was 60.3%. The estimated 2-year PFS rate and 2-year OS rate was 44.2% (95% confidence interval (CI), 31.3-57.1%) and 71.4% (95% CI, 59.4-83.4%), respectively. Radiation-induced esophagitis was the most common non-hematologic toxicity and 5 patients (8.6%) developed grade≥3 esophagitis. While, with PEG nutrition support, the nutrition-related indicators presented a clear trend toward a gradual improvement. Treatment-related death was not observed. Conclusions The moderately hypo-fractionated radiotherapy combined with S-1 showed promising loco-regional disease control and survival benefit in inoperable LA-ESCC patients. Meanwhile, favorable nutritional status and low incidence of severe radiation-induced esophagitis were observed with PEG nutritional support. Moreover, endoscopy examination contributed to the early detection of recurrent esophageal lesions and timely salvage treatment. The efficacy and toxicity of the combined regimen deserved further evaluation. Trial registration Clinicaltrials.gov, identifier NCT03660449.

    Abstract: To evaluate longitudinal changes of concurrent chemoradiotherapy (CCRT) related lymphopenia and its association with survival in locally advanced non-small cell lung cancer (LA-NSCLC) patients. Methods Total lymphocyte count (TLC) at baseline, weekly intervals during CCRT and monthly intervals up to 12 months after CCRT were documented. The Common Terminology Criteria for Adverse Events version 5.0 was used to grade the severity of lymphopenia. Cox regression analysis was performed to evaluate the association between overall survival (OS) and CCRT related lymphopenia at different timepoints. Logistic regression model was used to determine the clinical factors associated with TLC level. Results 381 LA-NSCLC patients treated with definitive CCRT without consolidation therapy (NCT02573506/NCT02577341) between 2011 to 2020 were analyzed. With a median follow-up of 45.8 months, the median OS was 41.0 months for all patients. Univariable analysis demonstrated that the 3 weeks during CCRT Grade (G) 4 lymphopenia (P=0.018), 2 months after CCRT G1-4 lymphopenia (P=0.004), 6 months after CCRT (6m-post-CCRT) G1-4 lymphopenia (P=0.001), and TLC nadir (P=0.020) were significantly associated with poorer OS. Multivariable analysis suggested that 6m-post-CCRT G1-4 lymphopenia (HR 2.614; P=0.041) were one of the independent predictors of OS. Further analysis inferred that radiation dose (OR: 1.328; P=0.005), GTV volume (OR: 1.004; P=0.036), and baseline TLC (OR: 0.288; P=0.001) were associated with 6m-post-CCRT lymphopenia. Conclusion The persistent lymphopenia at 6 months after CCRT was an independent prognostic factor of OS in LA-NSCLC patients. Higher radiation dose, larger gross tumor volume and lower baseline TLC were significantly related to 6m-post-CCRT lymphopenia.

    Abstract: Driven by global demand for new energy, Li-ion batteries (LIBs) have developed rapidly due to their competitive performance. Although LIBs show the advantages of high capacity and good cycling stability, their disadvantages such as uneven distribution of lithium resources are gradually exposed. Therefore, with abundant reserves, Na-ion batteries (NIB) have become one of the most promising solutions to make up for the deficiency of Li-ion battery. The NIBs layered oxide cathodes have the most potential applications of cathode material due to their high specific capacity (167 mAh·g〈sup〉–1〈/sup〉 in 2.4–4.3 V) and simple synthesis method. However, improving the cycling stability of layered cathode materials is one of the keys to their large-scale industrialization. To develop high capacity and cycling stability cathode materials, the Mg〈sup〉2+〈/sup〉 is substituted for Ni〈sup〉2+〈/sup〉 in NaNi〈sub〉0.4〈/sub〉Cu〈sub〉0.1〈/sub〉Mn〈sub〉0.4〈/sub〉Ti〈sub〉0.1〈/sub〉O〈sub〉2〈/sub〉 (NCMT), thereby obtaining a NaNi〈sub〉0.35〈/sub〉Mg〈sub〉0.05〈/sub〉Cu〈sub〉0.1〈/sub〉Mn〈sub〉0.4〈/sub〉Ti〈sub〉0.1〈/sub〉O〈sub〉2〈/sub〉 (NCMT-Mg) cathode material. The NCMT-Mg has a high reversible specific capacity of 165 mAh·g〈sup〉–1〈/sup〉 in a voltage window of 2.4–4.3 V. The reversible specific capacity of about 110 mAh·g〈sup〉–1〈/sup〉 at 0.1 C after 350 cycles with a capacity retention of 67.3% is about 13% higher than the counterpart of NCMT. The irreversible reaction is suppressed from P'3 phase to X phase for NCMT. The ex-XRD spectrometers further prove that the NCMT-Mg shows a P3 and X mixed phase after being initially charged to 4.3 V, but the NCMT shows an X phase. The irreversible phase transition is suppressed to increase the cycling stability. The inactive Mg〈sup〉2+〈/sup〉 replaces Ni〈sup〉2+〈/sup〉, reducing the charge compensation and stabilizing the structure, the inactive Mg〈sup〉2+〈/sup〉 can activate the charge compensation of Ni〈sup〉2+〈/sup〉/Cu〈sup〉2+〈/sup〉. The electrochemical activity increases from 77% to 86%. The high capacity and excellent cycling stability prove that the NCMT-Mg structure remains intact after various current rates have been tested. The long cycling stability mechanism is further systematically studied by using various technologies. The present work will provide an important reference for developing high-performance Na-ion cathode materials.

    Abstract: Sensor node energy is the key to the long life cycle of a sensor network. This paper considers the correlation of sensor nodes in continuous time on-chip data transfer, and compares the derivative-based prediction, polynomial regression, back propagation and convolutional neural networks prediction algorithms for data transfer and energy consumption minimization. Experimental results show that convolutional neural networks are the optimal control solution.

    Abstract: We aimed to evaluate the efficacy and feasibility of concurrent weekly docetaxel-nedaplatin and hypo-fractionated radiotherapy (hypo-RT) in atypical histologic subtypes of primary and metastatic mediastinal malignancies. Methods Fifty-four patients diagnosed with atypical primary or metastatic mediastinal malignancies were retrospectively reviewed. 30 patients received concurrent weekly docetaxel and nedaplatin and hypo-RT (CChRT group) and 24 patients had hypo-RT alone (hRT group). Overall response rate (ORR), in-field locoregional progression-free survival (LPFS) and toxicities were analyzed. The radiobiological effect was evaluated by the LQRGC/TCP model, incorporating four “R”s of radiobiology, Gompertzian tumor growth and radio-sensitizing effect of chemotherapeutic agent. The biologically effective doses (BEDs) were calculated. Results The median follow-up time was 29.2 months for all patients. The ORR was 86.7% in CChRT group, compared with 62.5% in hRT group ( p =0.033). The 2-year in-field LPFS of CChRT and hRT group was 73.4% and 47.3%, respectively ( p =0.003). There was no significant difference of any & gt;=Grade 3 toxicities between the two groups ( p =0.754). The mean total dose and mean BED by the LQRGC/TCP model in CChRT group were 58.2Gy and 72.34Gy, versus 52.6Gy and 67.25Gy in hRT group. Conclusions Concurrent weekly docetaxel-nedaplatin and hypo-RT achieved promising in-field LPFS and tolerable toxicities compared with hypo-RT alone in different histologic subtypes of primary and metastatic mediastinal malignancies.
