    Abstract: Immunological mechanisms of susceptibility to nontuberculous mycobacterial (NTM) disease are poorly understood. To understand NTM pathogenesis, we evaluated innate and antigen-specific adaptive immune responses to Mycobacterium avium complex (MAC) in asymptomatic individuals with a previous history of MAC lung disease (MACDZ). We hypothesized that Mav-specific immune responses are associated with susceptibility to MAC lung disease. We measured MAC-, NTM-, or MAC/Mtb-specific T-cell responses by cytokine production, expression of surface markers, and analysis of global gene expression in 27 MACDZ individuals and 32 healthy controls. We also analyzed global gene expression in Mycobacterium avium -infected and uninfected peripheral blood monocytes from 17 MACDZ and 17 healthy controls. We were unable to detect increased T-cell responses against MAC-specific reagents in MACDZ compared to controls, while the responses to non-mycobacteria derived antigens were preserved. MACDZ individuals had a lower frequency of Th1 and Th1* T-cell populations. In addition, MACDZ subjects had lower transcriptional responses in PBMCs stimulated with a mycobacterial peptide pool (MTB300). By contrast, global gene expression analysis demonstrated upregulation of proinflammatory pathways in uninfected and M. avium -infected monocytes, i.e. a hyperinflammatory in vitro response, derived from MACDZ subjects compared to controls. Together, these data suggest a novel immunologic defect which underlies MAC pathogenesis and includes concurrent innate and adaptive dysregulation which persists years after completion of treatment.

    Abstract: To mitigate the risk of erosion and nutrient runoff, reduced tillage has become more prevalent in Norway. Within within recent decades, there have been some years with relatively high occurrence of Fusarium head blight and mycotoxins in Norwegian cereal grain. This is thought to have been caused by an increased inoculum potential (IP) of Fusarium spp. due to larger amount of crop residues remaining on the soil surface, in combination with weather conditions promoting fungal growth and infection of cereal plants. The objective of this work was to elucidate the influence of different tillage practices on the IP of Fusarium spp. and the subsequent Fusarium -infection and mycotoxin contamination of spring wheat grain at harvest. Tillage trials were conducted at two locations in southeast Norway (Solør and Toten) over three years, 2010-2012. Residues of wheat from the previous year were collected in spring. Fusarium avenaceum and Fusarium graminearum were the most common Fusarium species recorded on wheat straw residues. IP was calculated as the percentage of the residues infested with Fusarium spp. multiplied by the proportion of the soil surface covered with residues. The IP of Fusarium spp. was lower in ploughed plots compared to those tilled with harrowing only. Ploughing in spring resulted in a similarly low IP as autumn ploughing. In contrast, harrowing in autumn generally reduced IP more than did spring harrowing. The mycotoxin levels in the harvested wheat were generally low, except for deoxynivalenol at high levels in Solør 2011. Despite a lower IP of ploughed versus harrowed plots, this was not reflected in the content of Fusarium and mycotoxins in harvested grain. The Fusarium species that dominated in the residues examined in this study were the same as those detected in the harvested grain, supporting the finding that residues are an important source of inoculum.

    Abstract: A history of concussion is recognized as a risk factor for musculoskeletal injury, which is likely associated with physiological effects that warrant better understanding. This study aimed to assess the potential of measurements obtained from an immersive virtual reality (VR) test to identify a subtle perceptual–motor impairment that may be prospectively associated with the occurrence of a core or lower extremity sprain or strain. Methods A cohort of 68 high school athletes (41 female soccer players and 27 male football players) provided survey responses and completed an immersive VR test several days prior to the initiation of preseason practice sessions. Measurements of eye, neck, arm, and whole-body displacements were obtained during 40 successive lunging/reaching responses to visual stimuli moving horizontally across the VR headset display. Injury occurrences were electronically documented from the initial preseason practice session to the final game of the season. Results A statistically significant and intrinsically credible two-factor prediction model for core or lower extremity injury occurrence included an interaction between female sex and a self-reported history of two or more concussions, along with slow response time (RT) for arm reach (OR = 4.67; 95% CI, 1.51–14.43). Follow-up analyses identified sex-specific cut points for arm reach RT associated with elevated injury risk, which were ≥1.385 s for females and ≥1.257 s for males. Discussion High school female soccer players who have sustained more than one concussion appear to be highly vulnerable to core or lower extremity sprain or strain, with the risk of injury compounded by a slow arm reach RT. Male football players as a group demonstrated significantly faster arm reach RT than that of female soccer players, but slow perceptual–motor RT for arm reach was also identified as a potentially important injury risk factor for male players. Immersive VR appears to provide precise measurements of behavioral performance characteristics that depend on brain processing efficiency. Given that the speed, accuracy, and consistency of perceptual–motor responses may be modifiable, future research should explore the potential benefits of VR training for reducing the risk of sport-related injuries.
