    Abstract: In this paper, three species of the macrostomid free-living flatworm genus Macrostomum are described. Two species, Macrostomum littorale Wang & amp; Shi, sp. nov. and M. shekouense Wang & amp; Shi, sp. nov. , were collected from coastal water at Shenzhen, Guangdong Province, China. One species, M. brandi Wang & amp; Shi, sp. nov. , was collected from Perth, Western Australia and Queenscliff, Victoria, Australia. Macrostomum littorale sp. nov. differs from congeneric species within the genus in the length of the stylet, diameter of stylet opening, and the bend of the stylet. Macrostomum shekouense sp. nov. and M. brandi sp. nov. differ from similar species within the genus in the stylet morphology, position of the female antrum and developing eggs, or presence or absence of the false seminal vesicle. Phylogenetic analysis based on cytochrome c oxidase subunit I (COI) gene shows that M. littorale sp. nov. and M. hystrix are sister clades on two well-separated branch, M. shekouense sp. nov. and M. brandi sp. nov. are sister clades on two well-separated branches. Accordingly, both morphological and molecular evidence support M. littorale sp. nov. , M. shekouense sp. nov. , and M. brandi sp. nov. as three new species.

    Abstract: The circadian rhythm generated by circadian clock genes functions as an internal timing system. Since the circadian rhythm controls abundant physiological processes, the circadian rhythm evolved in organisms is salient for adaptation to environmental change. A disturbed circadian rhythm is a trigger for numerous pathological events. Recently, accumulated data have indicated that kidney stone disease (KSD) is related to circadian rhythm disturbance. However, the mechanism between them has not been fully elucidated. In this narrative review, we summarized existing evidence to illustrate the possible association between circadian rhythm disturbance and KSD based on the epidemiological studies and risk factors that are linked to circadian rhythm disturbance and discuss some chronotherapies for KSD. In summary, KSD is associated with systemic disorders. Metabolic syndrome, inflammatory bowel disease, and microbiome dysbiosis are the major risk factors supported by sufficient data to cause KSD in patients with circadian rhythm disturbance, while others including hypertension, vitamin D deficiency, parathyroid gland dysfunction, and renal tubular damage/dysfunction need further investigation. Then, some chronotherapies for KSD were confirmed to be effective, but the molecular mechanism is still unclear.

    Abstract: Based on butterfly microfluidic gene chip technology, a method for rapid, accurate and efficient detection of 19 common pathogenic microorganisms of nosocomial infection was established, and a butterfly microfluidic gene chip with high-throughput detection was designed and fabricated. Using constant temperature amplification technology, using the polymerase with chain replacement function to react at constant temperature (65?) and combined with microfluidic chip technology, primers were designed according to the target genes of 19 pathogenic microorganisms, and a butterfly microfluidic gene chip which can detect 19 common pathogenic microorganisms of nosocomial infection was made to simplify the inspection operation process and verify the sensitivity of the chip. The butterfly microfluidic gene chip can be used for the rapid and efficient detection of 19 common pathogenic microorganisms of nosocomial infection, and provides a new idea for the detection and auxiliary diagnosis of pathogenic microorganisms of nosocomial infection.

    Abstract: 〈sec〉 Moiré patterns formed by overlapping two circular gratings of slightly different pitches have been extensively used for measuring the two-dimensional (2D) and three-dimensional (3D) displacements. However, in the existing applications, Moiré patterns are analyzed based on geometric superposition, by which the 3D displacements cannot be instantaneously or simultaneously measured with a high accuracy. In this paper, radial shearing interferometry with double circular gratings of slightly different pitches is presented to realize the simultaneous measurement of 3D displacements. 〈/sec〉〈sec〉 The measurement is based on the principle that Moiré patterns produced by radial shearing interferometry are determined not only by the 2D in-plane displacements, but also by the out-of-plane displacement that brings about a phase shift between Moiré patterns of +1 and –1 diffraction orders. First, the production mechanism of Moiré patterns by radial shearing interferometry is studied based on the scalar diffraction theory and the intensity distribution of Moiré fringes of +1 and –1 orders is derived to establish the exact analytic relations between Moiré patterns and 3D displacements. Second, on the basis of spectrum characteristics of circular grating, a semicircular ring filter is proposed for spatial filtering to realize the simultaneous imaging of Moiré fringes of +1 and –1 orders. Then, the algorithm to quantitatively extract 3D displacements from Moiré patterns is proposed and demonstrated by numerical simulation. In the algorithm, Moiré patterns in the rectangular coordinate system are transformed into the polar coordinate system and skeletons are extracted to determine the feature points of the bright fringes. The in-plane displacements can be solved by feature points of +1 or –1 diffraction order, and the out-of-plane displacement can be computed by the feature points of +1 and –1 diffraction orders in the same bright fringe. Finally, experimental results prove that the maximum absolute error and mean error for in-plane displacements are 4.8 × 10〈sup〉–3〈/sup〉 mm and 2.0 × 10〈sup〉–4〈/sup〉 mm respectively, and 0.25 mm and 8.6 × 10〈sup〉–3〈/sup〉 mm for out-of-plane displacement. 〈/sec〉〈sec〉 In conclusion, by using the Moiré patterns of +1 and –1 diffraction orders imaged by radial shearing interferometer with double circular gratings of slightly different pitches, the 3D displacement can be simultaneously measured. The method has the advantages of simple device, high measurement accuracy, non-contact and instantaneous measurement, which provides an important guidance for practically measuring the 3D displacements. 〈/sec〉

    Abstract: Regulatory T cells (Tregs) have been found to play crucial roles in immune tolerance. However, the status of Tregs in refractory rheumatoid arthritis (RA) is still unclear. Moreover, low-dose interleukin-2 (IL-2) has been reported to selectively promote the expansion of Tregs. This study investigated the status of CD4 + Tregs and low-dose IL-2 therapy in patients with refractory RA. Methods The absolute number of CD4 + CD25 + FOXP3 + Treg (CD4 Treg), CD4 + IL17 + T (Th17), and other subsets in peripheral blood (PB) from 41 patients with refractory RA and 40 healthy donors was characterized by flow cytometry combined with an internal microsphere counting standard. Twenty-six patients with refractory RA were treated with daily subcutaneous injections of 0.5 million IU of human IL-2 for five consecutive days. Then, its effects on CD4 Treg and Th17 cells in PB were analyzed. Results A decrease in the absolute number of PB CD4 Tregs rather than the increase in the number of Th17 was found to contribute to an imbalance between Th17 and CD4 Tregs in these patients, suggesting an essential role of CD4 Tregs in sustained high disease activity. Low-dose IL-2 selectively increased the number of CD4 Tregs and rebalanced the ratio of Th17 and CD4 Tregs, leading to increased clinical symptom remission without the observed side effects. Conclusions An absolute decrease of PB CD4 Tregs in patients with refractory RA was associated with continuing disease activation but not the increase of Th17 cells. Low-dose IL-2, a potential therapeutic candidate, restored decreased CD4 Tregs and promoted the rapid remission of patients with refractory RA without overtreatment and the observed side effects.     Abstract: Lower-grade gliomas (LGGs) are characterized by remarkable genetic heterogeneity and different clinical outcomes. Classification of LGGs is improved by the development of molecular stratification markers including IDH mutation and 1p/19q chromosomal integrity, which are used as a hallmark of survival and therapy sensitivity of LGG patients. However, the reproducibility and sensitivity of the current classification remain ambiguous. This study aimed to construct more accurate risk-stratification approaches. Methods According to bioinformatics, the sequencing profiles of methylation and transcription and imaging data derived from LGG patients were analyzed and developed predictable risk score and radiomics score. Moreover, the performance of predictable models was further validated. Results In this study, we determined a cluster of 6 genes that were correlated with IDH mutation/1p19q co-deletion status. Risk score model was calculated based on 6 genes and showed gratifying sensitivity and specificity for survival prediction and therapy response of LGG patients. Furthermore, a radiomics risk score model was established to noninvasively assist judgment of risk score in pre-surgery. Taken together, a predictable nomogram that combined transcriptional signatures and clinical characteristics was established and validated to be preferable to the histopathological classification. Our novel multi-omics nomograms showed a satisfying performance. To establish a user-friendly application, the nomogram was further developed into a web-based platform: https://drw576223193.shinyapps.io/Nomo/ , which could be used as a supporting method in addition to the current histopathological-based classification of gliomas. Conclusions Our novel multi-omics nomograms showed the satisfying performance of LGG patients and assisted clinicians to draw up individualized clinical management.

    Abstract: It has been nearly 110 years since the discovery of superconductors, and more than 30 years since the discovery of high temperature superconductors (HTS). Great progress has been made in the application of superconducting electronics in the last two decades. HTS microwave devices have shown much higher perfomance than the traditional ones and have found their ways to the industry applications in mobile communication, radar, and special communication applications. Owing to the ultrahigh sensitivity to magnetic fields and currents, superconducting quantum interference devices (SQUIDs) have been used as the irresplacible sensors in geological surveying, magnetic resonanc imaging, biomagnetic imaging, and other areas. The sensitivity of superconducting radiation detectors such as superconducting SIS mixer, superconducting hot electron bolometer, superconducting transition edge sensor, superconducting nanowire single photon detector, and superconducting microwave kinetic inductance detector are near the quantum limitation. They are now key technology in geophysics, astrophysics, quantum information science, biomedicine, and so on. Superconducting Josephson parametric amplifier has become a key element for superconducting quantum computing. Superconducting integrated circuit has been included in the international roadmap for devices and systems, and shows that having the potential to become one of the mainstreams for post-Moore information processing technology. In metrology, superconducting Josephson effect and Josephson junction array devices have been widely used in the redefinition of quantum voltage reference and basic units of the International system of Units. Superconducting electronics plays an important role in the current quantum information technology boom, which in turn promotes the development of superconducting electronics. This review will brief introduce the research and application of superconducting electronics in China in recent years.

    Abstract: Tumor microenvironment (TME) status is closely related to breast cancer (BC) prognosis and systemic therapeutic effects. However, to date studies have not considered the interactions of immune and stromal cells at the gene expression level in BC as a whole. Herein, we constructed a predictive model, for adjuvant decision-making, by mining TME molecular expression information related to BC patient prognosis and drug treatment sensitivity. Methods Clinical information and gene expression profiles were extracted from The Cancer Genome Atlas (TCGA), with patients divided into high- and low-score groups according to immune/stromal scores. TME-related prognostic genes were identified using Kaplan-Meier analysis, functional enrichment analysis, and protein-protein interaction (PPI) networks, and validated in the Gene Expression Omnibus (GEO) database. Least absolute shrinkage and selection operator (LASSO) Cox regression analysis was used to construct and verify a prognostic model based on TME-related genes. In addition, the patients’ response to chemotherapy and immunotherapy was assessed by survival outcome and immunohistochemistry (IPS). Immunohistochemistry (IHC) staining laid a solid foundation for exploring the value of novel therapeutic target genes. Results By dividing patients into low- and high-risk groups, a significant distinction in overall survival was found (p & lt; 0.05). The risk model was independent of multiple clinicopathological parameters and accurately predicted prognosis in BC patients (p & lt; 0.05). The nomogram-integrated risk score had high prediction accuracy and applicability, when compared with simple clinicopathological features. As predicted by the risk model, regardless of the chemotherapy regimen, the survival advantage of the low-risk group was evident in those patients receiving chemotherapy (p & lt; 0.05). However, in patients receiving anthracycline (A) therapy, outcomes were not significantly different when compared with those receiving no-A therapy (p = 0.24), suggesting these patients may omit from A-containing adjuvant chemotherapy. Our risk model also effectively predicted tumor mutation burden (TMB) and immunotherapy efficacy in BC patients (p & lt; 0.05). Conclusion The prognostic score model based on TME-related genes effectively predicted prognosis and chemotherapy effects in BC patients. The model provides a theoretical basis for novel driver-gene discover in BC and guides the decision-making for the adjuvant treatment of early breast cancer (eBC).
