    Abstract: Stomata are pores found in the epidermis of stems or leaves that modulate both plant gas exchange and water/nutrient uptake. The development and function of plant stomata are regulated by a diverse range of environmental cues. However, how carbohydrate status in preexisting leaves might determine systemic stomatal formation within newly developing leaves has remained obscure. The glucose (Glc) sensor HEXOKINASE1 (HXK1) has been reported to decrease the stability of an ethylene/Glc signaling transcriptional regulator, EIN3 (ETHYLENE INSENSITIVE3). EIN3 in turn directly represses the expression of SUC2 ( sucrose transporter 2 ), encoding a master transporter of sucrose (Suc). Further, KIN10, a nuclear regulator involved in energy homeostasis, has been reported to repress the transcription factor SPCH (SPEECHLESS), a master regulator of stomatal development. Here, we demonstrate that the Glc status of preexisting leaves determines systemic stomatal development within newly developing leaves by the HXK1—¦EIN3—¦SUC2 module. Further, increasing Glc levels in preexisting leaves results in a HXK1-dependent decrease of EIN3 and increase of SUC2, triggering the perception, amplification and relay of HXK1-dependent Glc signaling and thereby triggering Suc transport from mature to newly developing leaves. The HXK1—¦EIN3—¦SUC2 molecular module thereby drives systemic Suc transport from preexisting leaves to newly developing leaves. Subsequently, increasing Suc levels within newly developing leaves promotes stomatal formation through the established KIN10⟶ SPCH module. Our findings thus show how a carbohydrate signal in preexisting leaves is sensed, amplified and relayed to determine the extent of systemic stomatal development within newly developing leaves.

    Abstract: Behçet’s disease (BD) is a chronic vasculitis characterized by systemic immune aberrations. However, a comprehensive understanding of immune disturbances in BD and how they contribute to BD pathogenesis is lacking. Here, we performed single-cell and bulk RNA sequencing to profile peripheral blood mononuclear cells (PBMCs) and isolated monocytes from BD patients and healthy donors. We observed prominent expansion and transcriptional changes in monocytes in PBMCs from BD patients. Deciphering the monocyte heterogeneity revealed the accumulation of C1q-high (C1q hi ) monocytes in BD. Pseudotime inference indicated that BD monocytes markedly shifted their differentiation toward inflammation-accompanied and C1q hi monocyte–ended trajectory. Further experiments showed that C1q hi monocytes enhanced phagocytosis and proinflammatory cytokine secretion, and multiplatform analyses revealed the significant clinical relevance of this subtype. Mechanistically, C1q hi monocytes were induced by activated interferon-γ (IFN-γ) signaling in BD patients and were decreased by tofacitinib treatment. Our study illustrates the BD immune landscape and the unrecognized contribution of C1q hi monocytes to BD hyperinflammation, showing their potential as therapeutic targets and clinical assessment indexes.

    Abstract: Child-directed speech is characterized by higher pitch, more expanded pitch contours, and more exaggerated phonetic contrasts, which was suggested to facilitate speech sound acquisition. This study examined the perceptual and acoustic differences of mothers' disyllabic Mandarin lexical tones directed to adults and children to determine whether mothers exaggerated the pitch targets of the four Mandarin tones when speaking to children. Twelve Mandarin-speaking mothers produced 700 child-directed (CD) and adult-directed (AD) disyllabic words in a picture naming task. Five Mandarin-native speakers identified the mothers' AD and CD tones in filtered speech. Overall, CD lexical tones were identified with significantly lower accuracy than AD lexical tones (89% vs. 94%, S = 25.5, p = 0.006, r = 0.927, Wilcoxon Signed Rank Test). Acoustic analysis showed that the mean fundamental frequency (f0) of the four tones in both syllables was significantly higher in CD than in AD productions. No difference was found between AD and CD in the distinctive pitch targets for the 4 tones, namely pitch shift for Tone1, F0 slope for Tone2, minimum F0 for Tone3 and F0 slope for Tone4. F0 plots showed mostly parallel contours in AD and CD productions without exaggeration of the phonetic contrasts of the tones. [Work supported by NIDCD F31 DC008470-01A1.]

    Abstract: Effective rehabilitation in Parkinson's disease (PD) is related to brain reorganization with restoration of cortico–subcortical networks and compensation of frontoparietal networks; however, further neural rehabilitation evidence from a multidimensional perspective is needed. To investigate how multidisciplinary intensive rehabilitation treatment affects neurovascular coupling, 31 PD patients (20 female) before and after treatment and 30 healthy controls (17 female) underwent blood oxygenation level-dependent functional magnetic resonance imaging and arterial spin labeling scans. Cerebral blood flow (CBF) was used to measure perfusion, and fractional amplitude of low-frequency fluctuation (fALFF) was used to measure neural activity. The global CBF–fALFF correlation and regional CBF/fALFF ratio were calculated as neurovascular coupling. Dynamic causal modeling (DCM) was used to evaluate treatment-related alterations in the strength and directionality of information flow. Treatment reduced CBF–fALFF correlations. The altered CBF/fALFF exhibited increases in the left angular gyrus and the right inferior parietal gyrus and decreases in the bilateral thalamus and the right superior frontal gyrus. The CBF/fALFF alteration in right superior frontal gyrus showed correlations with motor improvement. Further, DCM indicated increases in connectivity from the superior frontal gyrus and decreases from the thalamus to the inferior parietal gyrus. The benefits of rehabilitation were reflected in the dual mechanism, with restoration of executive control occurring in the initial phase of motor learning and compensation of information integration occurring in the latter phase. These findings may yield multimodal insights into the role of rehabilitation in disease modification and identify the dorsolateral superior frontal gyrus as a potential target for noninvasive neuromodulation in PD. SIGNIFICANCE STATEMENT Although rehabilitation has been proposed as a promising supplemental treatment for PD as it results in brain reorganization, restoring cortico–subcortical networks and eliciting compensatory activation of frontoparietal networks, further multimodal evidence of the neural mechanisms underlying rehabilitation is needed. We measured the ratio of perfusion and neural activity derived from arterial spin labeling and blood oxygenation level-dependent fMRI data and found that benefits of rehabilitation seem to be related to the dual mechanism, restoring executive control in the initial phase of motor learning and compensating for information integration in the latter phase. We also identified the dorsolateral superior frontal gyrus as a potential target for noninvasive neuromodulation in PD patients.

    Abstract: Impaired oxygen and cellular metabolism is a hallmark of ischaemic injury in acute stroke. Magnetic resonance spectroscopic imaging (MRSI) has long been recognized as a potentially powerful tool for non-invasive metabolic imaging. Nonetheless, long acquisition time, poor spatial resolution, and narrow coverage have limited its clinical application. Here we investigated the feasibility and potential clinical utility of rapid, high spatial resolution, near whole-brain 3D metabolic imaging based on a novel MRSI technology. In an 8-min scan, we simultaneously obtained 3D maps of N-acetylaspartate and lactate at a nominal spatial resolution of 2.0 × 3.0 × 3.0 mm3 with near whole-brain coverage from a cohort of 18 patients with acute ischaemic stroke. Serial structural and perfusion MRI was used to define detailed spatial maps of tissue-level outcomes against which high-resolution metabolic changes were evaluated. Within hypoperfused tissue, the lactate signal was higher in areas that ultimately infarcted compared with those that recovered (P  & lt; 0.0001). Both lactate (P  & lt; 0.0001) and N-acetylaspartate (P  & lt; 0.001) differed between infarcted and other regions. Within the areas of diffusion-weighted abnormality, lactate was lower where recovery was observed compared with elsewhere (P  & lt; 0.001). This feasibility study supports further investigation of fast high-resolution MRSI in acute stroke.

    Abstract: Factors that may/may not influence inter-rater reliability in assessing the accuracy of monosyllabic Mandarin tones produced by children and adults were examined in three experiments. Experiment 1 investigated inter-judge reliability in two groups of Mandarin-speaking adults—one group from China and the other from Taiwan—on their categorization of filtered tones produced by adults and children. The results showed that the magnitude of inter-rater agreement was associated with the production accuracy of the speakers; the judges attained lower agreement in categorizing children’s tones than adults’ tones. All judges who indicated that Mandarin was their strongest language and that they had learned and used Mandarin since birth performed similarly in their tone categorization despite the fact that they came from and were residing in different countries. Similar results was found in experiment 2, in which one group of the judges in experiment 1 categorized tones produced by a new and larger group of adults and children, and in experiment 3, in which a different group of adults categorized another new set of tones produced by a different group of speakers.     Abstract: Photosynthesis of hydrogen peroxide (H 2 O 2 ) in ambient conditions remains neither cost effective nor environmentally friendly enough because of the rapid charge recombination. Here, a photocatalytic rate of as high as 114 μmol⋅g −1 ⋅h −1 for the production of H 2 O 2 in pure water and open air is achieved by using a Z-scheme heterojunction, which outperforms almost all reported photocatalysts under the same conditions. An extensive study at the atomic level demonstrates that Z-scheme electron transfer is realized by improving the photoresponse of the oxidation semiconductor under visible light, when the difference between the Fermi levels of the two constituent semiconductors is not sufficiently large. Moreover, it is verified that a type II electron transfer pathway can be converted to the desired Z-scheme pathway by tuning the excitation wavelengths. This study demonstrates a feasible strategy for developing efficient Z-scheme photocatalysts by regulating photoresponses.
