    Abstract: The present study investigated reliable global measures that can be used to screen/assess infants at-risk for speech problems due to cerebral palsy (CP). Three measures were used in this study: mean duration of utterances, mean duration of syllables, and vocalization rate, which are suggested as likely points of divergence from typical development in infants with CP. Recordings of 9 months and 21 months of ten infants who were at risk of CP and seventeen 9-month-old typically-developing (TD) infants were coded. Naïve listeners filled out a questionnaire about a perceptual evaluation of speech samples of these infants after listening to the first 50 utterances with little background noise in each recording. The questions included, for example, resonance, hoarseness, breathiness, loudness, and tremor. Results of the three measures did not show statistically significant differences between the groups. However, descriptive analyses showed that infants with CP at 9 months produced longer utterances and syllables than when at 21 months. Infants with CP also produced shorter utterances and syllables and fewer vocalizations per minute than TD infants. Furthermore, the correlation between the results of the three measures and perceptual ratings was examined in search for other global measures and to refine the questionnaire questions.

    Abstract: The age means between cerebral palsy children (CPs) and typically developing children (TDs) were not ideally matched in previous studies targeting the fricative productions of Mandarin-acquiring CPs (Liu et al., 2017, 2016). Therefore, in this study, age-matched CPs (N = 8, Mean = 5;6) and TDs (N = 8, Mean = 5;6) were recruited. Six acoustical parameters (frication duration, rise time, and first to fourth spectral moments) of the five Mandarin Chinese voiceless fricatives (labiodental, alveolar, alveolo-palatal, velar, and retroflex) produced by the participants were compared. The results from Mann-Whitney U tests indicated that, for the labiodental, alveolar, alveolo-palatal, and retroflex fricatives, the values from frication duration, rise time, and first and second spectral moments produced by CPs were generally significantly lower than those produced by TDs. The current results from age-matched groups were different from previous studies and suggested that: 1) CPs tended to have a more posterior constriction site and a more restricted lingual constriction area with the passive articulator; 2) the third and fourth spectral moments were less effective in quantifying the differences between the two groups. Future studies are suggested to observe the fricative productions from older or younger age-matched groups so that the fricative developmental trajectory of Mandarin-acquiring CPs can be faithfully depicted.

    Abstract: Most eukaryotes possess two RecA-like recombinases (ubiquitous Rad51 and meiosis-specific Dmc1) to promote interhomolog recombination during meiosis. However, some eukaryotes have lost Dmc1. Given that mammalian and yeast Saccharomyces cerevisiae ( Sc ) Dmc1 have been shown to stabilize recombination intermediates containing mismatches better than Rad51, we used the Pezizomycotina filamentous fungus Trichoderma reesei to address if and how Rad51-only eukaryotes conduct interhomolog recombination in zygotes with high sequence heterogeneity. We applied multidisciplinary approaches (next- and third-generation sequencing technology, genetics, cytology, bioinformatics, biochemistry, and single-molecule biophysics) to show that T. reesei Rad51 ( Tr Rad51) is indispensable for interhomolog recombination during meiosis and, like Sc Dmc1, Tr Rad51 possesses better mismatch tolerance than Sc Rad51 during homologous recombination. Our results also indicate that the ancestral Tr Rad51 evolved to acquire Sc Dmc1-like properties by creating multiple structural variations, including via amino acid residues in the L1 and L2 DNA-binding loops.

    Abstract: Advancing new ideas of rechargeable batteries represents an important path to meeting the ever-increasing energy storage needs. Recently, we showed rechargeable sodium/chlorine (Na/Cl 2 ) (or lithium/chlorine Li/Cl 2 ) batteries that used a Na (or Li) metal negative electrode, a microporous amorphous carbon nanosphere (aCNS) positive electrode, and an electrolyte containing dissolved aluminum chloride and fluoride additives in thionyl chloride [G. Zhu et al. , Nature 596 , 525–530 (2021) and G. Zhu et al. , J. Am. Chem. Soc. 144 , 22505–22513 (2022)]. The main battery redox reaction involved conversion between NaCl and Cl 2 trapped in the carbon positive electrode, delivering a cyclable capacity of up to 1,200 mAh g −1 (based on positive electrode mass) at a ~3.5 V discharge voltage [G. Zhu et al. , Nature 596 , 525–530 (2021) and G. Zhu et al. , J. Am. Chem. Soc. 144 , 22505–22513 (2022)]. Here, we identified by X-ray photoelectron spectroscopy (XPS) that upon charging a Na/Cl 2 battery, chlorination of carbon in the positive electrode occurred to form carbon-chlorine (C-Cl) accompanied by molecular Cl 2 infiltrating the porous aCNS, consistent with Cl 2 probed by mass spectrometry. Synchrotron X-ray diffraction observed the development of graphitic ordering in the initially amorphous aCNS under battery charging when the carbon matrix was oxidized/chlorinated and infiltrated with Cl 2 . The C-Cl, Cl 2 species and graphitic ordering were reversible upon discharge, accompanied by NaCl formation. The results revealed redox conversion between NaCl and Cl 2 , reversible graphitic ordering/amorphourization of carbon through battery charge/discharge, and probed trapped Cl 2 in porous carbon by XPS.

    Abstract: What kind of genetic variation contributes the most to adaptation is a fundamental question in evolutionary biology. By resequencing genomes of 80 individuals, we inferred the origin of genomic variants associated with a complex adaptive syndrome involving multiple quantitative traits, namely, adaptation between high and low altitudes, in the vinous-throated parrotbill ( Sinosuthora webbiana ) in Taiwan. By comparing these variants with those in the Asian mainland population, we revealed standing variation in 24 noncoding genomic regions to be the predominant genetic source of adaptation. Parrotbills at both high and low altitudes exhibited signatures of recent selection, suggesting that not only the front but also the trailing edges of postglacial expanding populations could be subjected to environmental stresses. This study verifies and quantifies the importance of standing variation in adaptation in a cohort of genes, illustrating that the evolutionary potential of a population depends significantly on its preexisting genetic diversity. These findings provide important context for understanding adaptation and conservation of species in the Anthropocene.

    Abstract: The high degree of similarity between the mouse and human genomes is demonstrated through analysis of the sequence of mouse chromosome 16 (Mmu 16), which was obtained as part of a whole-genome shotgun assembly of the mouse genome. The mouse genome is about 10% smaller than the human genome, owing to a lower repetitive DNA content. Comparison of the structure and protein-coding potential of Mmu 16 with that of the homologous segments of the human genome identifies regions of conserved synteny with human chromosomes (Hsa) 3, 8, 12, 16, 21, and 22. Gene content and order are highly conserved between Mmu 16 and the syntenic blocks of the human genome. Of the 731 predicted genes on Mmu 16, 509 align with orthologs on the corresponding portions of the human genome, 44 are likely paralogous to these genes, and 164 genes have homologs elsewhere in the human genome; there are 14 genes for which we could find no human counterpart.

    Abstract: Two-dimensional transition metal dichalcogenides (TMDCs) such as molybdenum sulfide MoS 2 and tungsten sulfide WSe 2 have potential applications in electronics because they exhibit high on-off current ratios and distinctive electro-optical properties. Spatially connected TMDC lateral heterojunctions are key components for constructing monolayer p-n rectifying diodes, light-emitting diodes, photovoltaic devices, and bipolar junction transistors. However, such structures are not readily prepared via the layer-stacking techniques, and direct growth favors the thermodynamically preferred TMDC alloys. We report the two-step epitaxial growth of lateral WSe 2 -MoS 2 heterojunction, where the edge of WSe 2 induces the epitaxial MoS 2 growth despite a large lattice mismatch. The epitaxial growth process offers a controllable method to obtain lateral heterojunction with an atomically sharp interface.

    Abstract: Bacterial transpeptidase and transglycosylase on the surface are essential for cell wall synthesis, and many antibiotics have been developed to target the transpeptidase; however, the problem of antibiotic resistance has arisen and caused a major threat in bacterial infection. The transglycosylase has been considered to be another excellent target, but no antibiotics have been developed to target this enzyme. Here, we determined the crystal structure of the Staphylococcus aureus membrane-bound transglycosylase, monofunctional glycosyltransferase, in complex with a lipid II analog to 2.3 Å resolution. Our results showed that the lipid II-contacting residues are not only conserved in WT and drug-resistant bacteria but also significant in enzymatic activity. Mechanistically, we proposed that K140 and R148 in the donor site, instead of the previously proposed E156, are used to stabilize the pyrophosphate-leaving group of lipid II, and E100 in the acceptor site acts as general base for the 4-OH of GlcNAc to facilitate the transglycosylation reaction. This mechanism, further supported by mutagenesis study and the structure of monofunctional glycosyltransferase in complex with moenomycin in the donor site, provides a direction for antibacterial drugs design.
