    Abstract: In the present study, urban soundscape has been investigated by social survey and soundwalking. An intensive social survey using 11-point numerical scale was carried out with narrative interviews; the dominant contexts affecting the perception of urban soundscape were derived from the survey: visual image, acoustic comfort, and natural brightness. Thus, the effects of contexts on the perception of soundscape were also confirmed by soundwalking. In addition, the effects of water sounds on the enhancement of urban soundscape were examined; auditory experiments were performed to enhance the perception of urban spaces with various kinds of water sounds. Water sound, which had 3 dB less than urban noise, was found as a preferable masker. Visual images, as well as acoustical characteristics of water sounds, had an effect on the perception of urban soundscape.

    Abstract: The goal of the present study is to characterize water sounds that can be used in urban open spaces to mask road traffic noise. Sounds and visual images of a number of water features located in urban open places were obtained and subsequently analyzed in terms of psychoacoustical metrics and acoustical measures. Laboratory experiments were then conducted to investigate which water sound is appropriate for masking urban noise. The experiments consisted of two sessions: (1) Audio-only condition and (2) combined audio-visual condition. Subjective responses to stimuli were rated through the use of preference scores and 15 adjectives. The results of the experiments revealed that preference scores for the urban soundscape were affected by the acoustical characteristics of water sounds and visual images of water features; Sharpness that was used to explain the spectral envelopes of water sounds was proved to be a dominant factor for urban soundscape perception; and preferences regarding the urban soundscape were significantly related to adjectives describing “freshness” and “calmness.”

    Abstract: In this study, urban soundscapes containing combined noise sources were evaluated through field surveys and laboratory experiments. The effect of water sounds on masking urban noises was then examined in order to enhance the soundscape perception. Field surveys in 16 urban spaces were conducted through soundwalking to evaluate the annoyance of combined noise sources. Synthesis curves were derived for the relationships between noise levels and the percentage of highly annoyed (%HA) and the percentage of annoyed (%A) for the combined noise sources. Qualitative analysis was also made using semantic scales for evaluating the quality of the soundscape, and it was shown that the perception of acoustic comfort and loudness was strongly related to the annoyance. A laboratory auditory experiment was then conducted in order to quantify the total annoyance caused by road traffic noise and four types of construction noise. It was shown that the annoyance ratings were related to the types of construction noise in combination with road traffic noise and the level of the road traffic noise. Finally, water sounds were determined to be the best sounds to use for enhancing the urban soundscape. The level of the water sounds should be similar to or not less than 3 dB below the level of the urban noises.

    Abstract: Wnt signaling through both canonical and noncanonical pathways plays a core role in development. Dysregulation of these pathways often causes cancer development and progression. Although the pathways independently contribute to the core processes, a regulatory molecule that commonly activates both of them has not yet been reported. Here, we describe a long noncoding RNA (lncRNA), HERES, that epigenetically regulates both canonical and noncanonical Wnt signaling pathways in esophageal squamous cell carcinoma (ESCC). For this study, we performed RNA-seq analysis on Korean ESCC patients and validated these results on a larger ESCC cohort to identify lncRNAs commonly dysregulated in ESCCs. Six of the dysregulated lncRNAs were significantly associated with the clinical outcomes of ESCC patients and defined 4 ESCC subclasses with different prognoses. HERES reduction repressed cell proliferation, migration, invasion, and colony formation in ESCC cell lines and tumor growth in xenograft models. HERES appears to be a transacting factor that regulates CACNA2D3 , SFRP2 , and CXXC4 simultaneously to activate Wnt signaling pathways through an interaction with EZH2 via its G-quadruple structure-like motif. Our results suggest that HERES holds substantial potential as a therapeutic target for ESCC and probably other cancers caused by defects in Wnt signaling pathways.

    Abstract: As the clinical failure of glioblastoma treatment is attributed by multiple components, including myelin-associated infiltration, assessment of the molecular mechanisms underlying such process and identification of the infiltrating cells have been the primary objectives in glioblastoma research. Here, we adopted radiogenomic analysis to screen for functionally relevant genes that orchestrate the process of glioma cell infiltration through myelin and promote glioblastoma aggressiveness. The receptor of the Nogo ligand (NgR1) was selected as the top candidate through Differentially Expressed Genes (DEG) and Gene Ontology (GO) enrichment analysis. Gain and loss of function studies on NgR1 elucidated its underlying molecular importance in suppressing myelin-associated infiltration in vitro and in vivo. The migratory ability of glioblastoma cells on myelin is reversibly modulated by NgR1 during differentiation and dedifferentiation process through deubiquitinating activity of USP1, which inhibits the degradation of ID1 to downregulate NgR1 expression. Furthermore, pimozide, a well-known antipsychotic drug, upregulates NgR1 by post-translational targeting of USP1, which sensitizes glioma stem cells to myelin inhibition and suppresses myelin-associated infiltration in vivo. In primary human glioblastoma, downregulation of NgR1 expression is associated with highly infiltrative characteristics and poor survival. Together, our findings reveal that loss of NgR1 drives myelin-associated infiltration of glioblastoma and suggest that novel therapeutic strategies aimed at reactivating expression of NgR1 will improve the clinical outcome of glioblastoma patients.

    Abstract: Iron is an essential biometal, but is toxic if it exists in excess. Therefore, iron content is tightly regulated at cellular and systemic levels to meet metabolic demands but to avoid toxicity. We have recently reported that adaptive thermogenesis, a critical metabolic pathway to maintain whole-body energy homeostasis, is an iron-demanding process for rapid biogenesis of mitochondria. However, little information is available on iron mobilization from storage sites to thermogenic fat. This study aimed to determine the iron-regulatory network that underlies beige adipogenesis. We hypothesized that thermogenic stimulus initiates the signaling interplay between adipocyte iron demands and systemic iron liberation, resulting in iron redistribution into beige fat. To test this hypothesis, we induced reversible activation of beige adipogenesis in C57BL/6 mice by administering a β3-adrenoreceptor agonist CL 316,243 (CL). Our results revealed that CL stimulation induced the iron-regulatory protein–mediated iron import into adipocytes, suppressed hepcidin transcription, and mobilized iron from the spleen. Mechanistically, CL stimulation induced an acute activation of hypoxia-inducible factor 2-α (HIF2-α), erythropoietin production, and splenic erythroid maturation, leading to hepcidin suppression. Disruption of systemic iron homeostasis by pharmacological HIF2-α inhibitor PT2385 or exogenous administration of hepcidin-25 significantly impaired beige fat development. Our findings suggest that securing iron availability via coordinated interplay between renal hypoxia and hepcidin down-regulation is a fundamental mechanism to activate adaptive thermogenesis. It also provides an insight into the effects of adaptive thermogenesis on systemic iron mobilization and redistribution.

    Abstract: Although aminoacyl-tRNA synthetases (ARSs) are essential for protein synthesis, they also function as regulators and signaling molecules in diverse biological processes. Here, we screened 11 different human ARSs to identify the enzyme that is secreted as a signaling molecule. Among them, we found that lysyl-tRNA synthetase (KRS) was secreted from intact human cells, and its secretion was induced by TNF-α. The secreted KRS bound to macrophages and peripheral blood mononuclear cells to enhance the TNF-α production and their migration. The mitogen-activated protein kinases, extracellular signal-regulated kinase and p38 mitogen-activated protein kinase, and Gαi were determined to be involved in the signal transduction triggered by KRS. All of these activities demonstrate that human KRS may work as a previously uncharacterized signaling molecule, inducing immune response through the activation of monocyte/macrophages.
