    Abstract: This study conducts an ERP experiment to explore the online processing mechanism of Chinese xiehouyu, a subcategory of Chinese idiomatic expressions with a metaphorical two-part allegorical saying, regarded as a non-literal language construct. Using a 2 × 2 design, (high familiarity (HF)/low familiarity (LF)) × (literally-biasing context (LC)/metaphorically-biasing context (MC)), the researchers have obtained the following findings: (1) familiarity plays an important role in Chinese xiehouyu processing, i.e. the metaphorical meaning of a HF Chinese xiehouyu can be directly activated while that of a LF one has to be derived from its literal meaning first; (2) contextual information also weighs in the process, i.e. the metaphorical meaning of a Chinese xiehouyu can be promoted in MC condition but suppressed in LC condition; (3) the interactive effect of familiarity and contextual information can be explained by the career of metaphor hypothesis; and (4) the Standard Pragmatic Model (SPM) of non-literal languages can explain the processing of LF xiehouyu , and the Direct Access Model (DAM) may to some extent account for the mechanism of HF one but fails to explain the case of LF one, while the Graded Salience Hypothesis (GSH) can provide an acceptable explanation for the processing mechanism of Chinese xiehouyus of varied familiarity.

    Abstract: This study investigated the categorical perception of Mandarin tones and consonant aspiration contrasts in babble noise among adults and adolescents aged 12–14 years, and explored the association between working memory and categorical perception. Method: Twenty-four adults and 20 adolescents with Mandarin as their native language were recruited. Their performances of phonemic identification and discrimination in babble noise and quiet conditions, digit span tasks, and nonword repetition were assessed. Results: Results indicated that, firstly, in the noise condition, both adults and adolescents showed wider boundary widths and lower between-category accuracies when perceiving aspiration of consonants than in the quiet condition, and the categorical boundary of tone perception in adolescents showed a transitional tendency toward Tone 1. Secondly, discrimination of consonant aspiration in adolescents needed to be further developed. Lastly, the accuracy of nonword repetition in adolescents was lower than that in adults, and adults with better auditory verbal working memory had better performance on tone perception. Conclusions: Our results provided evidence that tone perception is acquired easier than consonant aspiration perception, and tone perception is more robust and less susceptible to noise interference. Categorical perception performance relates to the capacity and utilization of auditory verbal working memory in some ways.

    Abstract: Information processing during human cognitive and emotional operations is thought to involve the dynamic interplay of several large-scale neural networks, including the fronto-parietal central executive network (CEN), cingulo-opercular salience network (SN), and the medial prefrontal-medial parietal default mode networks (DMN). It has been theorized that there is a causal neural mechanism by which the CEN/SN negatively regulate the DMN. Support for this idea has come from correlational neuroimaging studies; however, direct evidence for this neural mechanism is lacking. Here we undertook a direct test of this mechanism by combining transcranial magnetic stimulation (TMS) with functional MRI to causally excite or inhibit TMS-accessible prefrontal nodes within the CEN or SN and determine consequent effects on the DMN. Single-pulse excitatory stimulations delivered to only the CEN node induced negative DMN connectivity with the CEN and SN, consistent with the CEN/SN’s hypothesized negative regulation of the DMN. Conversely, low-frequency inhibitory repetitive TMS to the CEN node resulted in a shift of DMN signal from its normally low-frequency range to a higher frequency, suggesting disinhibition of DMN activity. Moreover, the CEN node exhibited this causal regulatory relationship primarily with the medial prefrontal portion of the DMN. These findings significantly advance our understanding of the causal mechanisms by which major brain networks normally coordinate information processing. Given that poorly regulated information processing is a hallmark of most neuropsychiatric disorders, these findings provide a foundation for ways to study network dysregulation and develop brain stimulation treatments for these disorders.

    Abstract: The migratory locust is one of the most notorious agricultural pests that undergo a well known reversible, density-dependent phase transition from the solitary to the gregarious. To demonstrate the underlying molecular mechanisms of the phase change, we generated 76,012 ESTs from the whole body and dissected organs in the two phases. Comparing 12,161 unigene clusters, we identified 532 genes as phase-related ( P 〈 0.01). Comprehensive assessment of the phase-related expression revealed that, whereas most of the genes in various categories from hind legs and the midgut are down-regulated in the gregarious phase, several gene classes in the head are impressively up-regulated, including those with peptidase, receptor, and oxygen-binding activities and those related to development, cell growth, and responses to external stimuli. Among them, a superfamily of proteins, the JHPH super-family, which includes j uvenile hormone-binding protein, h examerins, p rophenoloxidase, and h emocyanins, were highly expressed in the heads of the gregarious hoppers and hind legs of the solitary hoppers. Quantitative PCR experiments confirmed in part the EST results. These differentially regulated genes have strong functional implications that numerous molecular activities are involved in phase plasticity. This study provides ample molecular markers and genomic information on hemimetabolous insects and insights into the genetic and molecular mechanisms of phase changes in locusts.

    Abstract: G-quadruplexes (G4s) formed by guanine-rich nucleic acids play a role in essential biological processes such as transcription and replication. Besides the 〉 1.5 million putative G-4–forming sequences (PQSs), the human genome features 〉 640 million single-nucleotide variations (SNVs), the most common type of genetic variation among people or populations. An SNV may alter a G4 structure when it falls within a PQS motif. To date, genome-wide PQS–SNV interactions and their impact have not been investigated. Herein, we present a study on the PQS–SNV interactions and the impact they can bring to G4 structures and, subsequently, gene expressions. Based on build 154 of the Single Nucleotide Polymorphism Database (dbSNP), we identified 5 million gains/losses or structural conversions of G4s that can be caused by the SNVs. Of these G4 variations (G4Vs), 3.4 million are within genes, resulting in an average load of 〉 120 G4Vs per gene, preferentially enriched near the transcription start site. Moreover, 〉 80% of the G4Vs overlap with transcription factor–binding sites and 〉 14% with enhancers, giving an average load of 3 and 7.5 for the two regulatory elements, respectively. Our experiments show that such G4Vs can significantly influence the expression of their host genes. These results reveal genome-wide G4Vs and their impact on gene activity, emphasizing an understanding of genetic variation, from a structural perspective, of their physiological function and pathological implications. The G4Vs may also provide a unique category of drug targets for individualized therapeutics, health risk assessment, and drug development.
