    Abstract: This study was conducted to investigate the effects of partial substitution of dietary fishmeal (FM) by soy protein isolate (SPI) on lipid metabolism and gut microbiota of spotted seabass. A diet containing 30% FM formed the basal diet (FM), and two SPI diets were formulated in which 25% (SPI25) and 50% (SPI50) of FM were replaced by SPI. Each diet was fed to triplicates of fish for 56 days. The results showed that replacing dietary FM with SPI reduced triglyceride and cholesterol contents in the liver and serum, and the hepatic lipid droplets area was also decreased by dietary SPI inclusion. Furthermore, replacement of dietary FM with SPI markedly down-regulated the mRNA expression of genes involved in lipogenesis (srebpc-1c, fas, acc1, hmgcr, pparγ and chrebp) and lipolysis (atgl, hsl, pparα, and cpt1). Moreover, high-throughput sequencing analyses of gut microbiota revealed that dietary SPI inclusion dramatically decreased the Firmicutes/Bacteroidetes ratio. Overall, this study indicated that replacing 25-50% of dietary FM with SPI could reduce lipid accumulation in serum and liver of spotted seabass, which was associated with the suppressed hepatic lipogenesis and the remodeled gut microbiota.

    Abstract: Soy protein isolate (SPI) is a promising plant protein source to replace fish meal (FM) in aquatic feeds. This study investigated the growth performance, digestive enzyme activity, and intestinal morphology of spotted seabass (Lateolabrax maculatus) fed diets with FM partially substituted by SPI. Three iso-nitrogenous and iso-lipidic diets were formulated to replace 0, 25, and 50% of FM with SPI, being abbreviated as FM, SPI25, and SPI50, respectively. Each diet was allocated to triplicates of fish for 8 weeks. The results indicated that the weight gain was gradually reduced with increasing dietary SPI levels, as did intestinal trypsin and lipase activities. However, these traits were not significantly decreased until 50% of FM was substituted by SPI. Furthermore, feed intake and lipid digestibility significantly decreased with dietary SPI inclusion. Moreover, replacing 50% of dietary FM with SPI significantly reduced the intestinal villus height and perimeter ratio, whereas these traits did not markedly differ between FM and SPI25 groups. In conclusion, this study indicated that up to 25% of dietary FM could be substituted with SPI without significantly affecting the growth, digestive enzyme activity, or intestinal morphology in spotted seabass.

    Abstract: Immunoglobulin (Ig) is a type of globulin produced by B lymphocytes during pathogenic infection of vertebrates. It has immune functions and can realize specific recognition and neutralization of corresponding antigens. As IgM is reported first in fish, IgM is the first antibody produced during immune responses and plays a vital role in systemic and mucosal immune tissues. IgM molecules have two forms: membrane-bound IgM (mIgM) and secreted IgM (sIgM). The latter is produced by plasmacytes and secreted into body fluid, existing as immunological effect molecules. The former embeds into B cytomembrane and exists as an antigen receptor. It binds with assistant molecules to form cell receptor compounds. This study reviews research progress on the structures and production processes of IgM genes in different fish species and the distribution characteristics of IgM on B cells, mediated signal pathways, and functions. It aims to enrich basic theoretical knowledge of fish immunology and provide some scientific references for disease control in fishes.

    Abstract: The purpose of this study was to develop a loop-mediated isothermal amplification (LAMP) method for rapid, sensitive, and simple detection of Streptococcus agalactiae in farmed fish. A set of four primers, two outer and two inner, was designed from the fibrinogen-binding (fbsB) proteins gene sequence of S. agalactiae, and conditions for LAMP were optimized as incubation of all reagents for 60 min at 64°C. LAMP products were judged by agar gel electrophoresis as well as the naked eye after development by Synergy Brands Green I (SYBR Green I). Sensitivity of the assay was 2.8 × 103 CFU/ml (2.8 CFU per reaction) and no positive amplification was detected for other related bacteria. This LAMP method can be used to detect S. agalactiae infection in the laboratory and the field.

    Abstract: Complement (C) is a serum protein with immunity-related functions and enzymatic activities. It can induce inflammatory responses and antibody formation following activation by substances such as pathogens. Moreover, it is a crucial molecule in the congenital immune defense system. Complement manifests earlier than immunoglobulin in the evolutionary process of fish’s immune system. Therefore, the complement system of fish is vital for congenital and acquired immunities. A complement system is a polymolecule system comprising more than 30 soluble proteins, membrane-bound proteins, and complement receptors. It can be activated through three pathways for immunoregulation, the clearance of the immune complex, etc. The liver primarily produces the fish complement C3, which is a significant component of the complement system of fishes. C3 is the hub of three activation pathways. Research works concerning the structure, generation process, expression, and functions of C3 in fishes are reviewed in this study.

    Abstract: This study was conducted to investigate the effect of Candida utilis and Aspergillus niger, combined with fermented Ginkgo biloba leaves (FGB) on immunity, antioxidant capability, lipid metabolism, and disease resistance against Aeromonas hydrophila infection in juvenile blunt snout bream (Megalobrama amblycephala). Fish were fed basal diets supplemented with fermented Ginkgo biloba leaves FGB1(0.125%), FGB2(0.25%), FGB3(0.5%), FGB4(1.0%), or Control (0.00%) respectively. At the end of the 60-day feeding trial, fish were challenged by A. hydrophila and mortality rate was recorded for the next 7 days. Results showed that, compared with the control, serum myeloperoxidase (MPO) activity of groups FGB1 and FGB2, alkaline phosphatase (AKP) and lysozyme (LZM) activity of group FGB2 increased significantly (P 〈 0.05), while malondialdehyde (MDA) and low density lipoprotein cholesterol (LDL-C) contents of group FGB2, immunoglobulin M (IgM) activity of group FGB4 decreased significantly (P 〈 0.05). Furthermore, Acid phosphatase (ACP), superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSH) and triglyceride (TG) activities were not affected (P 〉 0.05). After A. hydrophila challenge, the relative mortality of group FGB1 and FGB2 were significantly lower than the control (P 〈 0.05), however the highest rate was in group FGB4. Results of this study indicate that dietary supplementation of 0.125%~0.25% fermented G. biloba leaves can significantly enhance immunity, antioxidant capability, and lipid metabolism of blunt snout bream, as well as improve its disease resistance.

    Abstract: In the present study, a pathogenic strain, JL, was isolated from Pengze crucian carp (Carassius auratus) eggs suffering from saprolegniosis. It was initially determined as Saprolegnia sp. strain JL. Saprolegnia species have been implicated for significant fungal contamination, involving both living and dead fish and their eggs. A phylogenetic tree was constructed using the maximum parsimony method. The tree shows that the JL strain was closely related to Saprolegnia parasitica isolate SAP171, isolated from Salmo trutta suffering from saprolegniasis in Laukaa, Finland. The minimum inhibitory concentration (MIC) of 20 Chinese herbs was screened. Pseudolarix kaempferi Gord. (Pinaceae) was the most effective in inhibiting growth of the bacteria and was chosen for further trial. Significant protective efficacy of 52.63% and 73.68% was obtained against the JL strain in Pengze crucian carp eggs at P. kaempferi concentrations of 12.5 mg/ml and 25.0 mg/ml, respectively.

    Abstract: To assess the genetic background of the germplasm resource of rainbow clam Moerella iridescens along China coasts, population genetic structure and genetic diversity of rainbow clam from five geographical populations in the Yellow Sea and the East China Sea (Lianyungang, Chongming, Cixi, Zhoushan, and Ningde) were analyzed based on rDNA (ITS1-5.8S-ITS2) sequences. Forty-Seven rDNA haplotypes were identified among 109 rainbow clam individuals of the five populations. Gen4 and Gen21 were the dominant haplotypes, accounting for 49.5% of rDNA whole gene sequences, and shared by the five populations. Six to eight exclusive haplotypes were found in each population. The average haplotype diversity index (HD) of the five populations was 0.8733. The genetic distance among populations ranged from 0.002 to 0.004, and haplotypes from the same population were not clustered in a single clade. Molecular variance analysis (AMOVA) demonstrated that the main genetic differentiation mainly occurred within population, and the genetic differentiation coefficient (FST) among five populations was between -0.02836 and 0.01896, which meant that no genetic differentiation among populations occurred. Overall, the five geographical populations showed a high genetic diversity, but no evident genetic structure or genetic differentiation.
