    Abstract: β-glucan is an effective, available immunostimulant that has been successfully used to improve the immune defense of shrimp against pathogenic infection. However, the long-term continuous use of β-glucan can lead to immune fatigue, and intermittent feeding may be an effective way of avoiding this phenomenon. A 60-day growth trial was conducted to compare the effects of different feeding strategies of diets containing 200 mg/kg β-glucan on the growth and immune system of the Pacific white shrimp Litopenaeus vannamei. The results showed that either continuous feeding or intermittent feeding of diets containing 200 mg/kg β-glucan did not promote the growth of L. vannamei. However, the levels of superoxide dismutase, alkaline phosphatase, and acid phosphatase in the hepatopancreas of L. vannamei in the intermittent feeding group were significantly higher than those in the control and continuous feeding groups. Expression levels of the genes encoding LPS/β-glucan binding protein, superoxide dismutase, lysozyme, penaeid in-3a, and catalase were also significantly upregulated in the intermittent feeding group, whereas most of the immune parameters in the continuous feeding group were not significantly different from those of the control group. After artificial infection with Vibrio parahaemolyticus, the average mortality rates in the control group, the continuous feeding group, and the intermittent feeding group were 76.67%, 78.89%, and 45.56%, respectively. The results indicated that the addition of 200 mg/kg β-glucan to the diet did not promote the growth of L. vannamei, but the strategy of intermittent feeding effectively prevented immune fatigue and enhanced disease resistance, perhaps by increasing nonspecific immunity in L. vannamei.

    Abstract: Portunus trituberculatus is an important aquaculture species in China. Presently, three main feeding models are used for P. trituberculatus: complete feeding with commercial formulated feed (M1), mixed feeding with commercial formulated feed and trash fish (M2) and complete feeding with trash fish (M3). This study assessed the gonad development and nutritional quality of female P. trituberculatus under the three feeding models and provides a scientific basis for optimizing the feeding models of P. trituberculatus in the future. Overall, this study concluded that hypoplasia of gonads, and the biochemical, fatty acid, and amino acid composition in the gonads, hepatopancreas, and muscle of P. trituberculatus fed with the commercial formulated diet were poor. The mixed feeding with the commercial formulated diet and trash fish was an ideal feeding mode. This mode not only alleviates the dependence of the P. trituberculatus breeding industry on miscellaneous fish resources but also ensures the excellent quality of P. trituberculatus.

    Abstract: In this study, the effects of salinity fluctuation on gene expression patterns in ovary and hepatopancreas of Litopenaeus vannamei were analyzed. The expression level of vitellogenin in the ovaries were significantly, markedly, and slightly, inhibited respectively in the hepatopancreas after brooders were transferred from 30‰ to 20‰ or 40‰ salinity for 4 days. The expression levels of the development of germ cell related genes Dmc1, proliferating cell nuclear antigen (PCNA) and cyclophilin A were significantly reduced at 20‰ or 40‰ salinity. The transcript level of shrimp ovarian peritrophin (SOP) was significantly reduced at 20‰ salinity, and the expression level of ovary specific gene mucin-5 AC was significantly higher at 40‰ salinity. Four stress related genes, cytoplasmic carbonic anhydrase (LvCAc), Na+/K+-ATPase alpha subunit (LvNK), V-H+-ATPase subunit A (LvVH), and heat shock protein 90 (LvHSP90), were significantly up-regulated at 40‰ salinity in the ovaries. In the hepatopancreas, the transcript level of fatty acid synthase (LvFAS) was significantly inhibited under 40‰ salinity. The mRNA levels of stress related genes LvNK, LvVH, and LvHSP90, were significantly lower at 40‰ salinity. Results indicated that changes in salinity caused transcription inhibition of germ cell and ovarian development related genes, and the expression patterns of stress related genes in the ovaries and hepatopancreas were quite different.

    Abstract: Clarias fuscus, Silurus asotus and Silurus meridionalis are economically important species in the aquaculture industry in China. However, comparative analysis of immune indices of C. fuscus, S. asotus, and S. eridionalis has been rarely been conducted. In the present study, head kidney and spleen indices and blood serum immunologically related enzyme activity of C. fuscus, S. asotus, and S. meridionalis were detected. Both head kidney and spleen body ratios were highest in S. meridionalis, followed by S. asotus, and lowest in C. fuscus. This suggests that the degree of lymphocyte proliferation and immune function of S. eridionalis might be strongest compared to S. asotus and C. fuscus. Blood serum lysozyme activity, superoxide dismutase (SOD) activity, and alkaline phosphatase activity, were highest in S. meridionalis, followed by S. asotus, but lowest in C. fuscus. Compared to S. asotus, acid phosphatase activity and catalase activity was higher in C. fuscus. Both acid phosphatase and catalase activities were highest in S. meridionalis, which was in line with lysozyme, superoxide dismutase (SOD), alkaline phosphatase, head kidney and spleen body ratios. This indicated that compared to S. asotus and C. fuscus, the immune function of S. meridionalis was strongest, and might have the greatest resistance to invaders like parasites, bacteria, and viruses. These results provide a theoretical basis for selection of varieties and optimization of germplasm in fish culture production and may be very valuable in future research.

    Abstract: Ammonia-N, the principal end-product of protein catabolism, is an important environmental toxic factor in ponds. Elevated environmental ammonia-N is very toxic to aquatic animals and has a deleterious effect on their immune system. However, its effect on their immune system remains unclear. In this study, the effects of ammonia-N (0, 5, 10 and 30 mg/L) on immune responses of Fresh Water Pearl Mussel Hyriopsis cumingii were determined. When exposed to 5 mg/L ammonia-N lysozyme activity increased significantly. Superoxide dismutase activity was highest in the 5 mg/L group followed by the 10 mg/L group. The marked decrease of Lysozyme activity of Fresh Water Pearl Mussel Hyriopsis cumingii in 10 mg/L and 30 mg/L ammonia-N groups suggests that the higher concentrations of ammonia-N reduces or inhibits their non-specific immunity. Compared to the control group, superoxide dismutase activity in 30 mg/L ammonia also decreased significantly. When exposed to 5mg/L ammonia-N, catalase, acid phosphatase and alkaline phosphatase activity enhanced. In the 10 mg/L and 30 mg/L group, ammonia-N decreased significantly The lysozyme, catalase, acid phosphatase and alkaline phosphatase activity showed a similar tendency. This suggests that in H. cumingii immunostimulatory response is enhanced at low ammoniaN concentrations. The results of this study provide a theoretical basis for disease prevention in the freshwater pearl mussel.

    Abstract: In May 2016, there was a disease outbreak among pufferfish (Takifugu rubripes) in a marine farm in Tianjin municipality, with the cumulative mortality rate reaching 45% within 7 days from the beginning of the outbreak. The main symptoms of the disease were abdominal and anal swelling of the diseased fish. Significant damage was also observed histopathologically in the intestine, liver, and kidney. The strain H008 was isolated from diseased kidneys, and the challenge test revealed that the same disease was diagnosed in pufferfish which suffered similarly high mortality rates. Using physiological biochemical tests combined with 16S rDNA sequence analysis, the strain was identified as Listonella anguillarum. Antimicrobial susceptibility tests showed that H008 was resistant to 10 of 15 antimicrobial agents tested. To the best of our knowledge, this is the first report of L. anguillarum causing disease in pufferfish.

    Abstract: Immunoglobulins are globulins with antibody activity or antibody-like structures, and fish were the first vertebrates to develop immunoglobulins. There are three major types of immunoglobulins (IgM, IgD and IgZ) in teleost fish, which play an important role in resisting pathogen infection. For fish, the intestinal tract is not only the place for nutrient digestion but also the largest endocrine organ. The intestinal mucosal barrier system can block pathogenic microorganisms and toxins and is important for immunologic defense. Therefore, the study of the molecular mechanism of fish immunoglobulin in intestinal mucosa is of great significance to the selection of immune prevention strategies for fish diseases. At present, information on the immune system of intestinal mucosa in fish is scarce, and there are still many problems in the function of newly identified antibodies, which need further research. In this study, we have summarized current knowledge about the types, gene structure, biological function, and immune response of immunoglobulins in teleost fish, and intestinal mucosal immune tissue of fish. The scientific questions in this field were also presented according to the progress from the latest research in fish immunology. The aim of present study is to provide a theoretical basis for the study of fish immunology.

    Abstract: Vibrio species frequently infect Penaeus vannamei causing great economic losses to the Whiteleg shrimp industry. Rapid detection of pathogenic Vibrio infection would improve the fight against these diseases. In this study, single and multiple polymerase chain reaction (PCR) methods were developed to detect three species of pathogenic Vibrio: Vibrio fluvialis, Vibrio anguillarum, and Vibrio alginolyticus. Specific primers were designed for the toxR gene of V. fluvialis, the flaA gene of V. anguillarum, and the pyrH gene of V. alginolyticus. The bacteria were used as templates to establish a 25 μL reaction system for PCR amplification. The results showed that single and specific PCR amplification products of expected sizes were obtained (228bp, 1665bp, and 383bp, respectively). The lowest concentration detected for the three Vibrio species were 5.21×102 , 2.70×104 , and 2.48×102 colony forming units (cfu)/mL, respectively. We also developed a multiplex PCR method to identify the three Vibrio species accurately, and with improved identification efficiency. In addition, quantitative real-time PCR (qPCR) was used to identify the minimum detectable DNA concentration for the three Vibrio species (1.0×10-6 nmol/L for V. fluvialis, 1.0×10-7 nmol/L for V. anguillarum, and 1.0×10- 8 nmol/L for V. alginolyticus). Technical requirements for ordinary PCR are low, therefore PCR is a feasible technique to detect and diagnose Penaeus vannamei bacterial disease.

    Abstract: Mandarin fish Siniperca chuatsi (Basilewsky) is a unique species in East Asia and has very important ecological and economic value. There are significant differences in immune function between different populations of Mandarin fish. However, up to now, the comparative study of immune indices of different populations of wild mandarin fish has been scarce. In the present study, the transcription level of Immunoglobulin M (IgM) and blood serum immunologically related enzyme activity of three wild Mandarin fish populations from Wuqiangxi reservoir, Lingjintan reservoir, and Taoyuan reservoir of Yuanjiang river basin were detected. In head kidney, spleen and in the liver of Mandarin fish, the transcription level of IgM was highest in Taoyuan reservoir followed by Lingjintan reservoir, and was lowest in Wuqiangxi reservoir. The catalase activity of Mandarin fish was highest in Taoyuan reservoir followed by Wuqiangxi reservoir, and was lowest in Lingjintan reservoir. Total number of white blood cells, lysozyme activity, acid phosphatase activity, superoxide dismutase (SOD) activity, and alkaline phosphatase activity of Mandarin fish were in line with immunoglobulin M gene expression, which were highest in Taoyuan reservoir followed by Lingjintan reservoir, and lowest in Wuqiangxi reservoir. This indicated that the immune function of Mandarin fish from Taoyuan reservoir was strongest, and these fish had the greatest resistance to harmful invaders such as parasite, bacteria, and virus compared to Lingjintan reservoir group and Wuqiangxi reservoir group. The results of this study provide a theoretical basis for disease-resistant breeding and germplasm optimization in mandarin fish breeding production, and may be valuable in future research
