    Abstract: The present experiment evaluated the effects of dietary copper (Cu) on growth, antioxidant activities, and susceptibility to Aeromonas hydrophila infection of juvenile Oriental river prawn Macrobrachium nipponense, as well as determining the optimal dietary copper requirement. Semi-purified diets containing seven graded levels of copper (2.8, 12.2, 20.9, 29.8, 43.1, 78.9 and 157.1 mg/kg diet) from CuSO4⋅5H2O were fed to juvenile prawn (initial weight 0.101±0.002 g). The weight gain of prawns fed with 2.8-78.9 mg/kg Cu was higher and they had lower feed conversion ratios than the group fed 157.1 mg/kg Cu. Cu concentrations in the hepatopancreas, muscle, and whole body in prawns tended to increase with increased dietary Cu concentration. Hepatopancreas Cu-Zn superoxide dismutase (Cu-Zn SOD) activity, glutathione peroxidase (GPx), and total antioxidant competence (T-AOC) were highest (P 〈 0.05) in the 43.1 mg/kg Cu group. The hepatopancreas malondialdehyde (MDA) content was lower (P 〈 0.05) in the prawns fed 29.8 mg/kg Cu than that in prawns fed 2.8, 12.2, 78.9 and 157.1 mg/kg Cu. After the feeding experiment, prawns were injected with A. hydrophila, and the cumulative mortality rate of the prawns fed with 20.9-43.1 mg/kg Cu was lower than the prawns fed with 2.8 and 157.1 mg/kg Cu. The optimum requirement of dietary Cu in juvenile prawns was estimated at 26.9-27.8 mg/kg diet based on cumulative mortality rate and whole-body Cu retention.

    Abstract: The experiment simulated the logistical transportation of Penaeus monodon . It set four distinct gradient Vitamin C (Vc) addition levels, including 0mg/L, 111mg/L, 222mg/L, and 333mg/L, to investigate the impact of Vc on the anti-stress ability of P. monodon during transportation. And the survival rate following transportation and a 15-day interim rearing period were recorded. Moreover, the alterations in tissue structure and activity of the enzymes alkaline phosphatase (ACP), alkaline phosphatase (AKP), total superoxide dismutase (T-SOD), and total superoxide dismutase (T-AOC) were checked. The results showed that with the increase of Vc supplemental level, the survival rate of P. monodon after transportation and after 15 days of temporary rearing increased to varying degrees. ACP and AKP in the hepatopancreas increased first and then decreased. T-SOD activity in the 0mg/L group was the lowest and then decreased gradually. The activity of total antioxidant capacity (T-AOC) in the 0mg/L group was the lowest and then stabilized. The branchial tissue structure also changed. The branchial tissue blood cell disorder decreased, the diaphragm gradually narrowed, the cornered cortex gradually recovered, and the swelling decreased. In conclusion, Vc positively affects the survival rate of P. monodon after transportation and temporary cultivation and alleviates the stress of P. monodon . The amount of Vc added at about 333mg/Lin, the transportation of P. monodon could play a positive role. The experimental results provide primary data for the transportation of P. monodon .

    Abstract: Shewanella infection of fish has become a significant problem in aquaculture. In September 2014, a disease was seen in cultured loach (Misgurnus anguillicaudatus) in Xuzhou, central China. A gram-negative bacillus was isolated from the diseased loaches and was tentatively named strain MS1, which was then identified as Shewanella sp. by physiological and biochemical characteristics analysis. The strain MS1 showed highest 16S rRNA sequence identities (98.93%, 98.87%) with the latest two species listed (Shewanella sp. MR7, Shewanella sp. MR4). The phylogenetic tree constructed on the basis of 16S rRNA gene sequences strongly indicated that the strain MS1 is most closely related to the new Shewanella strains MR7 and MR4. The isolate MS1 was confirmed as the pathogen of the infected loaches by experimental reinoculation. The strain was susceptible to most antimicrobial agents tested, but resistant to glycopeptides (vancomycin, teicoplanin) and lincosamide (lincomycin, clindamycin). This is the second report on Shewanella sp. isolated from the diseased loach.

    Abstract: Pond water with algae blooms was collected, and maintained in the dark over a 2-day period without nutritional supplements. The physico-chemical parameters and the density of algae were measured. The concentration of NH 4+-N, DIN and PO4–P in water in darkness significantly increased, and the density of algae decreased significantly. Isolated water was exposed to three bio-remediation water treatments: addition of green algae, Bacillus subtilis and photosynthetic bacteria. After remediation, the concentrations of NH4+ -N, NO3- -N, DIN and PO4- -P in the three treatment groups were significantly lower than the control group. No difference was observed in the concentration of NO2–N between the control group and the photosynthetic bacteria group. Results from all treatments showed water quality deterioration along with increase in nitrogen and phosphorus concentrations. Bio-remediation methods can be effectively adapted in shrimp culture after algae decay, and one of the most promising biological remediation strategies is the use of green algae.

    Abstract: Aeromonas salmonicida is a fish pathogen that has resulted in significant losses to the aquaculture industry worldwide. In the winter of 2018/2019, a serious outbreak of A. salmonicida infection of grass carp (Ctenopharyngodon idella) occurred in China. Since C. idella do not feed at low water temperatures, antibiotics cannot be delivered orally, thus the use of disinfectants is the only viable option to control the disease in the winter months. Therefore, the aim of this study was to elucidate the inhibitory effects of different disinfectants against A. salmonicida at various temperatures and pH values. The microdilution method was used to determine the minimum inhibitory concentration (MIC) of five disinfectants against A. salmonicida at different temperatures. The results showed that A. salmonicida was highly adaptable to temperatures ranging from 8 to 35 °C and pH values from 5 to 9. Moreover, based on the results, benzalkonium bromide (BB) had the strongest inhibitory effect at every temperature. Since A. salmonicida has strong environment adaptability, BB should be chosen to control the disease at low temperatures.

    Abstract: This trial investigated the effect of dietary Ginkgo biloba leaf (GBL) on the growth performance and nonspecific immunity of red swamp crayfish Procambarus clarkii. 180 Crayfishes were randomly divided into three groups. One group was fed with basic diet, whereas the other two groups were fed with diets containing 1% and 3% GBL. After 32 days of feeding, GBL addition tended to increase the body weight gain rate compared with control. In 3% GBL group, the bodyweight gain rate of male crayfish was higher than that of female crayfish. While female crayfish were advantageous in terms of meat yield. Liver-related indexes were influenced by GBL addition and 3% GBL could reduce glutamic pyruvic transaminase and glutamic oxaloacetic transaminase as well as total cholesterol in male crayfish, showing its function in liver protection. Moreover, GBL addition effects on liver protection was better in male crayfish than female crayfish.

    Abstract: We used pseudo‐triple‐stage tandem MS to rapidly differentiate terminal α(2,3)‐ and α(2,6)‐linked sialic acids of glycopeptides and native glycans. The precursor ions generate important diagnostic fragments to distinguish different terminal linked isomeric glycans and glycopeptides based on collision‐induced dissociation (CID). The diagnostic ions include hexose linked with N ‐acetylhexosamine residues, sialic acid with the loss of water, and sialic acid residues. Their values of mass‐to‐charge ratio ( m / z ) are at 366.14, 274.09, and 292.10, respectively. The relative intensity ratios (RIR) were calculated by the intensity of signals at m / z 274.09 and 292.10 divided by the intensity of the signal at m / z 366.14. Under optimal conditions of an in‐source fragmentation voltage of 70 V and a normalized collision energy of 30 %, the difference in RIR of diagnostic ions between terminal α(2,3)‐linked sialic acid of glycopeptide (1.3) and terminal α(2,6)‐linked sialic acid of glycopeptide ( 〈 0.18) is the largest. Moreover, quantitative calibration curves with different percentages of α(2,3)‐ and α(2,6)‐linked sialoglycopeptides demonstrated that this method applied not only to biantennary N ‐linked glycan, but also to complex and hybrid N ‐linked glycans and O ‐linked glycans. From measurements of the RIR of diagnostic ions, erythropoietin, fetuin, and herceptin have 98, 70, and 100 %, respectively, α(2,3)‐linked N ‐sialic acid of total terminal sugar moieties. N ‐Glycosylation of the antibody‐binding fragment (Fab) moiety of cetuximab has 100 % α(2,6)‐linked N ‐glycolylneuraminic acid terminal glycan linkages. This method can be applied to identify compounds from chemical synthesis. It can also be used for quality evaluation of bio‐similar protein drugs.

    Abstract: Interferon regulatory factors (IRFs) have been confirmed as vital transcription factors for the immune system, which play a certain role in anti-infection defense, immune regulation, hematopoietic cell development, and maturation of the immune system. In this study, the cDNA of Interferon regulatory factor 5 (IRF5) was cloned from black-spotted frog, Pelophylax nigromaculatus , and termed PnIRF5. The results indicated, the full-length cDNA of PnIRF5 was 2090 bp, and a putative protein of 504 amino acids was encoded. The results of protein domain prediction suggested that PnIRF5 exhibited a DNA-binding domain (DBD), a middle region (MR), an IRF association domain (IAD), a virus activated domain (VAD), as well as two nuclear localization signals (NLSs). Phylogenetic analysis clustered PnIRF5 into the amphibian IRF5 subgroup in vertebrate IRF5 group. The results of qPCR show that PnIRF5 mRNA was expressed in all examined tissues, with higher levels were identified in the kidney, intestine, and lung. The gene expression of PnIRF5 was analyzed in kidney, spleen, and liver of black-spotted frog after challenged with Elizabethkingia miricola , and the maximum expression level of PnIRF5 in liver, spleen, and kidney were 1.80, 2.33 and 2.88 times that at 0 days post challenged, respectively. As indicated by the results, the PnIRF5 protein may take on critical significance in protecting black-spotted frog from E. miricola infection, and they may play a certain role in the immune response of black-spotted frog against bacteria.

    Abstract: Pond soils play a fundamental role determining nutrient availability and regulate the characteristics of phytoplankton communities. This study investigated the change of phytoplankton communities in three types (alkaline, neutral, and acid) of purple soil. There were 107 species of phytoplankton detected in three purple soils belonging to 52 genera of 6 phyla (68 species in acid group, 69 species in neutral group and 64 species in alkaline group). The highest percentage of phytoplankton in three purple soil groups is Chlorophyta, followed by Bacillariophyta, and Euglenophyta. There were 23 species of 5 phyla in three purple soil ponds as the dominant species of phytoplankton, and the dominance varied from 0.05 to 0.67. The dominant species in neutral and alkaline purple soil groups were more than those in acid purple soil. Phytoplankton density varied from 43 to 7360 × 106 ind/L in the acid purple soil group, and the ratio of the highest value to the lowest one was 170. The variation range of phytoplankton density in neutral and alkaline purple soil ponds was 83 to 565 × 106 and 56 to 437 × 106 ind/L, and the ratio of the highest value to the lowest one was 6.8 and 7.8 respectively. The variation of biodiversity indices (Shannon—Wiener index, Pielous evenness index and Margalef richness index), were higher in the acid purple soil group than in the neutral and alkaline purple soil groups, similar to the result of phytoplankton density. Our results indicate that the phytoplankton community was more stable in the neutral and alkaline purple soil groups than in the acid soil group, and these two soil types are recommended for site-selection to build aquaculture ponds.
