    Abstract: We investigated the effects of ammonia and nitrite stress on genes involved in metabolism and disease resistance in Japanese sea bass ( Lateolabrax japonicus ). Fish were exposed to pure seawater (control), 65.4 mg/L NH4+ -N, or 60.9 mg/L NO2–N for 7 days. Muscle, head-kidney, gill, spleen, and liver tissues were sampled regularly and the expression of immune and metabolism-related genes was monitored. The results indicated that the expression of immune-related genes such as heat shock proteins 70 and 90 were upregulated in the head-kidney, spleen, gill, liver, and muscle tissues under ammonia and nitrite stress. Glutamine synthetase (GS) expression increased to different degrees under ammonia and nitrite stress, while glutamate dehydrogenase (GDH) in the stress group upregulated in the gills. The expression of carbonic anhydrase (CA) was upregulated in the spleen and gill tissue under ammonia or nitrite stress. Expression of leptin (LEP) was upregulated in the muscle, gills and liver under nitrite stress; under ammonia stress, LEP expression did not fluctuate significantly in the spleen or gill. Insulin-like growth factor-1 (IGF-1) was downregulated in the spleen, while did not fluctuate obviously in the muscle and head-kidney under ammonia stress. These results indicate that ammonia and nitrite stress led to an upregulated immune related indexes response and increased metabolic activities in L. japonicus , and provides reference suggestions for response status of these health markers answering the outside environment intimidation.

    Abstract: In order to explore the effects of acute low salinity stress on the liver structure, physiology and biochemistry of the larvae of Lateolabrax maculatus , the experiment was carried out to transfer the juveniles direct from salinity 30‰ (control group) to salinity 20‰, 10‰, and 0‰, respectively. Changes in liver microstructure and related physiological and biochemical indexes at different time points were observed. The results showed that no fish died during the whole experiment and all of them returned to normal behavior within 15 minutes. The liver cells of juvenile Chinese sea bass showed pathological changes such as swelling, vacuolation and nuclear pyknosis after low salinity stress, and the lower the salinity, the earlier the abnormal phenomenon appeared. The total antioxidant capacity (T-AOC) and superoxide dismutase (SOD) of liver showed a rapid increase and then decrease; The level of malondialdehyde (MDA) returned to normal after high fluctuations. The activity of lysozyme (LZM) in experimental groups were higher than that in control group between 12h and 48h, while there was not significantly different before 6h in each group (P 〉 0.05), all of them returned to normal in 96 h. The activity of alkaline phosphatase (ALP) and acid phosphatase (ACP) increased firstly and then decreased during the time of stress, and there was no significant difference between each group at the same time point (P 〉 0.05). The comprehensive analysis showed that the juvenile Chinese sea bass had strong osmotic pressure regulation ability, and the acute low salinity stress had little effect on the liver structure and physiological and biochemical indexes.

    Abstract: Truncated mucin‐type O‐glycans, such as Tn‐associated antigens, are aberrantly expressed biomarkers of cancer, but remain challenging to target. Reactive antibodies to these antigens either lack high affinity or are prone to antigen escape. Here, we have developed a robust chemoenzymatic strategy for the global labeling of Tn‐associated antigens, i. e. Tn (GalNAcα‐O‐Ser/Thr), Thomsen‐Friedenreich (Galβ1‐3GalNAcα‐O‐Ser/Thr, TF) and STF (Neu5Acα2‐3Galβ1‐3GalNAcα‐O‐Ser/Thr, STF) antigens, in human whole blood with high efficiency and selectivity. This method relies on the use of the O‐glycan sialyltransferase ST6GalNAc1 to transfer a sialic acid‐functionalized adaptor to the GalNAc residue of these antigens. By tagging, the adaptor functionalized antigens can be easily targeted by customized strategies such as, but not limited to, chimeric antigen receptor T‐Cells (CAR‐T). We expect this tagging system to find broad applications in cancer diagnostics and targeting in combination with established strategies.

    Abstract: Streptococcus agalactiae (GBS) is an important Gram-positive pathogen of fish aquaculture worldwide. In this study, we performed a comparative analysis of GBS S03 (serotype III) and S07 (serotype Ia) isolated from Schizothorax spp.. We explored the association between phenotypic antibiotic resistance, virulence, and genomic characteristics. Antimicrobial sensitivity tests on 12 common antibiotics using the disc diffusion method revealed that the GBS S03 resisted seven antibiotics, while GBS S07 showed sensitivity to eleven antibiotics. Pathogenicity analysis demonstrated greater virulence of GBS S07 than S03. Then, the occurrence of antibiotic resistance and virulence genes were identified using the whole-genome sequence (WGS) of GBS S03 and GBS S07. Besides just GBS S03 had gyrA and parC gene mutations, there were mefE , tetO , lnuB , lsaE , APH3’ , and sat-4 resistance genes present only in GBS S03 genome. In addition to 51 virulence genes in both GBS S03 and S07 genomes, GBS S07 carried virulence genes associated with invasion, such as SAN_1519 , rfbA and cylE genes. There was complete concordance between genotypic evidence and phenotypic characteristics. Virulence factors and phylogenetic analysis showed that GBS S03 and human sources shared an extremely close evolutionary relationship. Our findings also provide important proof for using WGS as an effective tool for phenotypic predictions of GBS.
