image The IP 3 receptor IP 3 R1 controls the ability of immature dendritic cells to explore their environment by regulating calcium release from the endoplasmic reticulum and by reinforcing the activity of the myosin light chain ( MLC ) kinase. Immature dendritic cells migrating in confinement exhibit peaks of calcium release during phases of fast locomotion. The migration of immature dendritic cells does not require extracellular calcium. Silencing of IP 3 R isoforms reduces the migration speed of immature dendritic cells. IP 3 R1 silencing specifically compromises the migration persistence of immature dendritic cells in 1‐, 2‐, and 3‐dimensional‐confined environments. By activating the MLC kinase, IP 3 R1 maintains the phosphorylation levels of MLC and the back/front polarization of myosin II in migrating immature dendritic cells.
