    Notizen: Abstract— Partially purified myelin from the brains of 17-day-old rats was separated into 4 subfractions on a three-step sucrose gradient by virtue of heterogeneity in density and particle size. Precursor-product relationships between different membrane fractions were investigated by determining the specific radioactivity of individual lipids in each subcellular fraction 15 min after intracranial injection of an appropriate precursor. Rats were injected with [2-3H]glycerol. myelin subfractions prepared, and individual lipids separated by TLC. For choline and ethanolamine phospholipids, specific radioactivity was highest in the densest fraction (D), intermediate in the next densest fraction (C), and lowest in the lighter fractions (B and A). Similar results were observed for cerebroside and sulphatide when [3H]galactose was the precursor. These data are consistent with (but do not prove) a precursor-product relationship for individual lipids from the densest to the lightest subfraction.Another experimental design involving time staggered injections of [3H] and [14C] precursors was developed which enables a more definitive result with regard to precursor-product relationships to be obtained. A precursor-product relationship between a given lipid in a dense myelin membrane fraction, and the same lipid in a lighter subfraction, would be indicated by a change in isotope ratio. If there is no precursor-product relationship. Ihe isotope ratio should be constant. Such experiments were done with [3H] and [14C]glycerol. The data indicated that phosphatidyl ethanolamine and its plasmalogen analog were added first to the densest subfraction and then in turn to the lighter subfractions. In contrast, phosphatidyl choline and its plasmalogen analog were added “simultaneously” (i.e. with delays of much less than 15min) to each of the subfractions. Similar experiments with [3H] and [14C]galactose showed that cerebroside, sulphatide and galactosyl diglyceride also entered the subfractions simultaneously rather than in sequential order. Thus the assembly of the myelin sheath involves an obligate order of addition of certain lipids. while other lipids are probably added in a random order.

    Notizen: Abstract— This paper specifies methodology for the sequential determination of manganese and catecholamines in selfsame brain samples and shows correlations between them. Small samples were obtained from five regions of brain of cats that had received either saline or levodopa. The doses of levodopa were varied so that although all animals reacted, some developed dyskinesia while others did not. Each sample was first analyzed nondestructively for manganese and then destructively for dopa and dopamine; thus errors inherent in analysing separate samples, due to the structural heterogeneity of the brain, were avoided. Statistically significant correlations were found (1) between levodopa-induced dyskinesia and the concentrations of dopamine and manganese in some of the regions analysed, and (2) between the concentrations of dopamine and of manganese in the caudates of the cats receiving the highest doses of levodopa.

    Notizen: The effect of neonatal thymectomy on the development of splenic and bone marrow natural cell-mediated cytotoxicity and on genetic resistance to bone marrow transplantation was examined in mice. Natural cytotoxicity was measured by a 51Cr release assay; the ability to engraft foreign bone marrow was assayed by the spleen colony method. The natural cytolytic response of spleen cells increased progressively from youth to early adulthood, whereas that of the bone marrow declined during the same age period. Neonatal thymectomy significantly elevated the natural killer cell response of young mice only (4 weeks, spleen; 6 weeks, bone marrow). In other experiments, neonatally thymectomized and sham-operated mice were lethally irradiated at 4 or 6 weeks of age and injected with 2.5, 5.0 or 10 million rat marrow cells. Six days later spleen colonies were markedly reduced in both 4- and 6-week-old neonatally thymectomized mice with all rat marrow cell doses tested. Neonatal thymectomy did not alter the percentage of erythroid versus other colonies at either 4 or 6 weeks. In both thymectomized and sham-operated mice the number of colonies increased with increases in marrow cell dose. The data are suggestive of a production and dissemination to the spleen of cells involved in the natural cytotoxic response from the bone marrow.

    Notizen: Abstract. Root nodule organogenesis is described. Plant regulated aspects of nodulation and N2 fixation are reviewed and discussed. Since the effective N2 fixing symbiosis requires the interaction of the host plant and bacterium in an appropriate environment (the rhizosphere and the root nodule) it is essential that research aimed at improving N2 fixation involve a knowledge and understanding of the plant genes that affect nodule development, growth, and function. Current knowledge of host plant genes involved in N2 fixation is summarized. Various experimental approaches to the study of the host plant's contribution to nodulation are noted. The functions of nodule specific proteins (nodulins) in symbiosis are delineated. Future areas of research are suggested.

Post harvest soil samples taken during the autumn of 1985 and 1986 were split and estimates made of the weed seedbank using two methods: (1) a physical separation of seed from the soil mineral fraction by a sieving/flotation procedure and (2) by placing soil in shallow containers in a greenhouse where seeds could germinate and be periodically counted over a period of eight months. Seedbank estimates derived from each procedure were analysed to determine the suitability of each technique for detecting treatment differences from an experiment evaluating tillage/herbicide effects on weed populations. Both techniques were suitable for determination of seedbank changes due to different tillage treatments and herbicide inputs. The two techniques also proved effective for detection of individual species in the seedbank and the two techniques provided comparable estimates of the relative density of individual weed species in the seedbank. Weed seedbank estimates obtained by the physical extraction procedure from the autumn 1985 soil samples were correlated with weed seedling counts made in the spring of 1986. In most cases, weed seedlings represented less than 10% of the number of seeds estimated by physical extraction the preceding autumn. Individual species seed estimates and subsequent weed counts were poorly correlated which indicated that the seed count estimates alone were poor predictors of weed flora.

    Notizen: The regulation of gene expression by the two-component regulatory system PhoP/PhoQ is necessary for Salmonella typhimurium survival within macro-phages, defensin resistance, acid resistance, and murine typhoid fever pathogenesis. Salmonella experience multiple environments during mammalian infection and survival requires tightly regulated gene expression. Alter phagocytosis by macrophages, signal transduction by PhoQ results in the transcription of QhoP-activated genes (pags) encoding proteins essential to bacterial survival and virulence. One such gene, pagC, encodes an envelope protein with amino acid similarity to an epithelial cell invasion protein of Yersina enterocolitica, Ail, and a bacteriophage λ outer membrane protein, Lom. The PhoP and PhoQ proteins can also repress the synthesis of proteins, encoded by phoP repressed genes (prgs), when pags are maximally expressed. If prgs encode receptors for toxic compounds, prg repression may protect the cell within macrophages when pag expression is most necessary. At least one prg locus, prgH, is required for full S. typhimurium mouse virulence. Within the macrophage, different environments may stimulate a switch from pag to prg expression that is necessary to Salmonella survival, prg expression may also be necessary for surviving non-macrophage environments. Study of the PhoP regulon should lead to the discovery of new virulence factors, increase knowledge of how gene regulation is essential to bacterial virulence, and perhaps lead to the development of better vaccines for typhoid fever.     Notizen: Five days after implantation with 11-ketotestosterone (KT) female Lythrypnus dalli exhibited elevated urinary KT levels and male-like genital papilla morphology. Papilla length increased, width decreased, and length : width ratio increased. Control fish exhibited increases in papilla length, and length : width ratio on day 3, but not on day 5. Changes in the total area of the papilla were not statistically significant. It is suggested that KT mediates genital papilla morphology rapidly and thus may be a useful indicator of circulating KT in L. dalli.

    Notizen: The maintenance of alveolar bone is a major clinical objective in dentistry. This is particularly difficult following such local inflammatory episodes as those of periodontitis or the loss of dentition (residual ridge resorption). We present evidence from beagle dogs that local infusion of prostaglandin E1 (PGE) for 3 weeks at doses of 500 to 2000μg per week produces a dramatic, localized formation of alveolar bone in the mandible which exhibits a normal lamellar architecture and mineralization pattern when evaluated by fluorescence microscopy and microradiography. Whether this newly formed bone becomes functionally integrated into the skeleton and can replace bone lost from surgical resections or trauma remains to be established. Nevertheless, these data indicate that predictable local osteogenesis may eventually be produced by infusions of PGE.
