    Description: This study tested the interactive effects of increased seawater temperature and CO2 partial pressure (pCO2) on the photochemistry, bleaching, and early growth of the reef coral Pocillopora damicornis. New recruits were maintained at ambient or high temperature (29 or 30.8 °C) and pCO2 (500 and 1100 µatm) in a full-factorial experiment for 3 weeks. Neither a sharp decline in photochemical efficiency (Fv/Fm) nor evident bleaching was observed at high temperature and/or high pCO2. Furthermore, elevated temperature greatly promoted lateral growth and calcification, while polyp budding exhibited temperature-dependent responses to pCO2. High pCO2 depressed calcification by 28% at ambient temperature, but did not impact calcification at 30.8 °C. Interestingly, elevated temperature in concert with high pCO2 significantly retarded the budding process. These results suggest that increased temperature can mitigate the adverse effects of acidification on the calcification of juvenile P. damicornis, but at a substantial cost to asexual budding.

    Description: Diurnal pCO2 fluctuations have the potential to modulate the biological impact of ocean acidification (OA) on reef calcifiers, yet little is known about the physiological and biochemical responses of scleractinian corals to fluctuating carbonate chemistry under OA. Here, we exposed newly settled Pocillopora damicornis for 7 days to ambient pCO2, steady and elevated pCO2 (stable OA) and diurnally fluctuating pCO2 under future OA scenario (fluctuating OA). We measured the photo-physiology, growth (lateral growth, budding and calcification), oxidative stress and activities of carbonic anhydrase (CA), Ca-ATPase and Mg-ATPase. Results showed that while OA enhanced the photochemical performance of in hospite symbionts, it also increased catalase activity and lipid peroxidation. Furthermore, both OA treatments altered the activities of host and symbiont CA, suggesting functional changes in the uptake of dissolved inorganic carbon (DIC) for photosynthesis and calcification. Most importantly, only the fluctuating OA treatment resulted in a slight drop in calcification with concurrent up-regulation of Ca-ATPase and Mg-ATPase, implying increased energy expenditure on calcification. Consequently, asexual budding rates decreased by 50% under fluctuating OA. These results suggest that diel pCO2 oscillations could modify the physiological responses and potentially alter the energy budget of coral recruits under future OA, and that fluctuating OA is more energetically expensive for the maintenance of coral recruits than stable OA.

    Notes: Peripheral tissue injury-induced central sensitization may result from the altered biochemical properties of spinal dorsal horn. However, peripheral nerve injury-induced modification of genes in the dorsal horn remains largely unknown. Here we identified strong changes of 14 channels, 25 receptors and 42 signal transduction related molecules in Sprague–Dawley rat dorsal spinal cord 14 days after peripheral axotomy by cDNA microarray. Twenty-nine genes were further confirmed by semiquantitative RT-PCR, Northern blotting, in situ hybridization and immunohistochemistry. These regulated genes included Ca2+ channel α1E and α2/δ1 subunits, α subunits for Na+ channel 1 and 6, Na+ channel β subunit, AMAP receptor GluR3 and 4, GABAA receptor α5 subunit, nicotinic acetylcholine receptor α5 and β2 subunits, PKC α, βI and δ isozymes, JNK1–3, ERK2–3, p38 MAPK and BatK and Lyn tyrosine-protein kinases, indicating that several signal transduction pathways were activated in dorsal spinal cord by peripheral nerve injury. These results demonstrate that peripheral nerve injury causes phenotypic changes in spinal dorsal horn. Increases in Ca2+ channel α2/δ1 subunit, GABAA receptor α5 subunit, Na+ channels and nicotinic acetylcholine receptors in both dorsal spinal cord and dorsal root ganglia indicate their potential roles in neuropathic pain control.

    Notes: Nanometric ammonium aluminum carbonate hydroxide (AACH) with a particle size less than 5 nm was produced from ammonium aluminum sulfate and ammonium hydrocarbonate under an optimized adding condition, and phase transformations of the AACH on heating and the effect of α-Al2O3 seeding on the transformation kinetics were investigated. The phase transformation sequence of AACH on heating was found to be AACH → amorphous Al2O3→γ-Al2O3→θ-Al2O3→α-Al2O3, where the formation temperatures of θ-Al2O3 and α-Al2O3 are 850° and 1050°C, respectively. The phase transformation sequence on heating of the AACH seeded with 5 wt%α-Al2O3 seed crystals 100 nm in diameter was found to be AACH → amorphous Al2O3→θ-Al2O3→α-Al2O3. γ-Al2O3 was not observed, and the θ-Al2O3→α-Al2O3 transformation temperature was reduced to 900°C by the seeding. By calcining the γ-AlOOH, the AACH, and the seeded AACH at their lowest temperatures to transform completely to α-Al2O3 (i.e., about 1200°, 1100°, and 900°C, respectively), α-Al2O3 powders have been obtained with mean particle sizes of about 150, 70, and 30 nm, respectively. The α-Al2O3 powder produced from the seeded AACH has the highest sintering reactivity. After sintering at 1400°C for 2 h, relative densities of the powder compacts of α-Al2O3 obtained by calcining γ-AlOOH, AACH, and the seeded AACH are 74.3%, 95.0%, and 98.5%, respectively.

    Notes: Research into the relative importance of P-glycoprotein (pgp) overexpression, in comparison to other mechanisms of multidrug resistance (mdr) phenotype development, has been hampered by difficulties of measurement in clinical samples. The small size, heterogeneity and often poor quality of clinical specimens renders the quantitation of mdr mRNA species by Northern analysis or RNAse protection difficult. The reverse transcriptase-polymerase chain reaction (RT-PCR) assay has both the sensitivity and specificity to make it suitable for the analysis of mdr mRNA levels in clinical samples. It also has a significant speed advantage over Northern and RNAse protection analysis. Unfortunately the variable nature of the reactions involved have made it difficult to obtain accurate and reproducible quantitative data. To overcome this difficulty we constructed internal RNA standards to each human and rat mdr mRNA species. When included in the RT-PCR reaction these internal standards allow both comparative and absolute quantitation of individual mdr family mRNA levels.

    Description: Humic substances (humics) are ubiquitous in terrestrial and aquatic environments where they can serve as electron acceptors for anaerobic oxidation of organic compounds. Methane is a powerful greenhouse gas, as well as the least reactive organic molecule. Anaerobic oxidation of methane (AOM) coupled to microbial reduction of various electron acceptors plays a crucial role in mitigating methane emissions. Here, we reported that humics could serve as terminal electron acceptors for AOM using enriched nitrate-reducing AOM microorganisms. AOM coupled to the reduction of humics was demonstrated based on the production of 13C-labelled carbon dioxide, and AOM activity was evaluated with different methane partial pressures and electron acceptor concentrations. After three-cycle reduction, both AOM activity and copy numbers of the archaea 16S rRNA and mcrA genes were the highest when anthraquinone-2,6-disulfonic acid and anthraquinone-2-sulfonic acid were electron acceptors. The high-throughput sequencing results suggested that ANME-2d were the dominant methane oxidation archaea after humics reduction, although the partner bacteria NC10 trended downward, other reported humics reduction bacteria (Geobactor and Anammox) appeared. The potential electron transfer models from ANME-2d to humics were proposed. These results enable a better understanding of available electron acceptors for AOM in natural environments and broaden our insight into the significant role of ANME-2d.

    Description: Exposure to ambient airborne particulate matter (PM) with an aerodynamic diameter less than 10 μm (PM 10 ) links with public health hazards and increases risk for lung cancer and other diseases. Recent studies have suggested that oxidative stress is a key mechanism underlying the toxic effects of exposure to PM 10 . Several components of water-soluble fraction of PM 10 (sPM 10 ) have been known to be capable of inducing oxidative stress in in vitro studies. In this study, we investigated if water-insoluble fraction of PM 10 (iPM 10 ) could be also capable of inducing oxidative stress and oxidative damage. Human lung epithelial A549 cells were exposed to 10 μg/mL of sPM 10 , iPM 10 or total PM 10 (tPM 10 ) preparation for 24 h. Here, we observed that all three PM 10 preparations reduced cell viability and induced apoptotic cell death in A549 cells. We further found that, similar to the exposure to sPM 10 and tPM 10 , the intracellular level of hydrogen peroxide (H 2 O 2 ) in the iPM 10 -exposed cells was increased significantly; meanwhile the activity of catalase was decreased significantly as compared with the unexposed control cells, resulting in significant DNA damage. Our data obtained from inductively coupled plasma-mass spectrometry (ICP-MS) assays showed that iron is the most abundant metal in all three PM 10 preparations. Thus, we have demonstrated that, similar to sPM 10 , iPM 10 is also capable of inducing oxidative stress by probably inducing generation of H 2 O 2 and impairing enzymatic antioxidant defense, resulting in oxidative DNA damage and even apoptotic cell death through the iron-catalyzed Fenton reaction. © 2012 Wiley Periodicals, Inc.
