    Description: Chinese sturgeon (Acipenser sinensis), mainly distributed in the Yangtze River, has been listed as a grade I protected animal in China because of a dramatic decline in population owing to loss of natural habitat for reproduction and interference by human activities. Understanding the proteome profile of Chinese sturgeon liver would provide an invaluable resource for protecting and increasing the stocks of this species. In this study, we have analyzed proteome profiles of juvenile Chinese sturgeon liver using a one-dimensional gel electrophoresis coupled to LC-MS/MS approach. A total of 1059 proteins and 2084 peptides were identified. The liver proteome was found to be associated with diverse biological processes, cellular components and molecular functions. The proteome profile identified a variety of significant pathways including carbohydrate metabolism, fatty acid metabolism and amino acid metabolism pathways. It also established a network for protein biosynthesis, folding and catabolic processes. The proteome profile established in this study can be used for understanding the development of Chinese sturgeon and studying the molecular mechanisms of action under environmental or chemical stress, providing very useful omics information that can be applied to preserve this species.

    Description: The South China Sea (SCS) is the largest semienclosed marginal sea in the western Pacific (WP) and connects to the west Pacific through the Luzon Strait (LU). In this study, we use the observation of transient tracer chlorofluorocarbon‐12 (CFC‐12) to calculate the ventilation time scales of the SCS, LU, and WP. The CFC‐12 and oxygen data are used together to identify the sandwiched structure vertically of the flows across the LU. The CFC‐12 and oxygen distributions reveal a pronounced decrease westward across the LU and a slight decrease southward in the transport of the SCS. The mean age gradient of the salinity minimum (Smin) water between the WP and the northern SCS could be a consequence of intensive mixing and entrainment of the inflow water from the WP. An expected difference in age between the LU and SSCS is verified to reflect the transit time for the given water layers in the SCS. Thus, a mean transit time of 77 ± 20 years is estimated for the intermediate water in the SCS interior.

    Description: Background: Immunomagnetic nanobead (IMNB) labeled with specific antibody, has been demonstrated to be useful for the capturing and detection of viruses. Results: In this study, we developed an imunomagnetic bead based on carboxyl-magnetic beads (MNB) labeled with a single-domain antibody (sdAb) for capturing foot-and-mouth disease (FMD) Asia 1 virus. After magnetic separation, complexes of MNB-sdAb-virus were detected with either a sandwich ELISA or QDs-C5 probe under a fluorescence microscope, and the complexes were used as templates for extraction of total RNA for amplification of the VP1 or 3D gene fragments using RT-PCR and real-time RT-PCR. The Asia 1 VLPs were efficiently captured through IMNB with a high binding rate of 5.09 μg of antigen/μl of bead suspension. Moreover, this method has been successfully used to capture Asia 1 antigen in synthetic samples. Conclusion: Ultimately, a specific and highly sensitive capture FMDV Asia 1 tool has been established that has the potential to enhance the sensitivity and reliability when diagnosing FMDV Asia 1.

Previous studies mostly focused on how the amount of developed land affected forest loss and fragmentation, but neglected the impacts of its spatial pattern. This paper examines the effects of both the amount and spatial pattern of urban expansion on forest loss and fragmentation. We conducted a comparison study in the six largest urban megaregions in China—Beijing-Tianjin-Hebei (BTH), Yangtze River Delta (YRD), Pearl River Delta (PRD), Wuhan (WH), Chengdu-Chongqing (CY), and Changsha-Zhuzhou-Xiangtan (CZT) urban megaregions. We first quantified both the magnitude and speed of urban expansion, and forest loss and fragmentation from 2000 to 2010. We then examined the relationships between urban expansion and forest loss and fragmentation by Pearson correlation and partial correlation analysis using the prefecture city as the analytical unit. We found: (1) urban expansion was a major driver of forest loss in the CZT, PRD, and CY megaregions, with 34.05%, 22.58%, and 19.65% of newly-developed land converted from forests. (2) Both the proportional cover of developed land and its spatial pattern (e.g., patch density) had significant impacts on forest fragmentation at the city level. (3) Proportional cover of developed land was the major factor for forest fragmentation at the city level for the PRD and YRD megaregions, but the impact of the spatial pattern of developed land was more important for the BTH and WH megaregions.

    Description: Background: A major production constraint on the important ornamental species chrysanthemum is black spot which is caused by the necrotrophic fungus Alternaria tenuissima. The molecular basis of host resistance to A. tenuissima has not been studied as yet in any detail. Here, high throughput sequencing was taken to characterize the transcriptomic response of the chrysanthemum leaf to A. tenuissima inoculation. Results: The transcriptomic data was acquired using RNA-Seq technology, based on the Illumina HiSeqTM 2000 platform. Four different libraries derived from two sets of leaves harvested from either inoculated or mock-inoculated plants were characterized. Over seven million clean reads were generated from each library, each corresponding to a coverage of 〉350,000 nt. About 70% of the reads could be mapped to a set of chrysanthemum unigenes. Read frequency was used as a measure of transcript abundance and therefore as an identifier of differential transcription in the four libraries. The differentially transcribed genes identified were involved in photosynthesis, pathogen recognition, reactive oxygen species generation, cell wall modification and phytohormone signalling; in addition, a number of varied transcription factors were identified. A selection of 23 of the genes was transcription-profiled using quantitative RT-PCR to validate the RNA-Seq output. Conclusions: A substantial body of chrysanthemum transcriptomic sequence was generated, which led to a number of insights into the molecular basis of the host response to A. tenuissima infection. Although most of the differentially transcribed genes were up-regulated by the presence of the pathogen, those involved in photosynthesis were down-regulated.
