    Notes: Abstract The wild Bolivian potato, Solanum berthaultii Hawkes, has been used as a source of resistance to the Colorado potato beetle (CPB), Leptinotarsa decemlineata Say, one of the most significant pests of potato. In this study, two reciprocal backcross S. tuberosum x S. berthaultii potato progenies, BCB and BCT, were mapped with RFLP markers and screened for resistance to CPB consumption, oviposition and defoliation. The genotypic and phenotypic data were combined and analysed to locate quantitative trait loci (QTLs) for resistance to CPB. Three QTLs on three chromosomes in BCB, and two QTLs on two chromosomes in BCT influenced resistance. The QTLs were generally additive but one instance of epistasis was noted. Each QTL accounted for 4–12% of the phenotypic variation observed in resistance. In the more resistant BCB population, a three QTL model explained ca. 20% of the variation in CPB oviposition. When alleles at the three QTLs were homozygous S. berthaultii, oviposition was reduced ca. 60% compared to the heterozygotes. The QTLs for resistance to CPB were compared to those previously identified for the type A and B glandular trichomes, which have been implicated in resistance in the same progenies. Generally, the QTLs for resistance to CPB coincided with loci associated with the glandular trichomes confirming the importance of the glandular trichomes in mediating resistance. However, a relatively strong and consistent QTL for insect resistance in both BCB and BCT on chromosome 1 was observed that was not associated with any trichome traits, suggesting the trichomes may not account for all of the resistance observed in these progenies.

    Notes: Abstract The linkage relationships and chromosomal locations of 14 enzyme-coding genes were investigated in Capsicum annuum L. (garden pepper) by monitoring segregations in backcross and F2 progeny of an interspecific cross between C. annuum cv. NM6-4 and C. chinense CA4 and by studying allele dosage effects in five hybrid primary trisomics. These conclusions can be reached: 6Pgdh-i is on the metacentric chromosome corresponding to the noir trisomie; Idh-1 and Est-3 are on chromosome 12; an aerocentric chromosome which corresponds to the pourple trisomie; Idh-1 is near the centromere and Est-3 is distal on the long arm of that chromosome. The other loci can be arranged in the following linkage groups and are apparently not located on the trisome corresponding to any of the trisomics tested: Est-4-18cM-(Pgi-1-3cM-Pgm-1); Prx-7-2cM-Tk-1; Pgm-2-2cM-Skdh-1; Est-1-OcM-Est-7. Linkage and dosage data combined with karyotype and meiotic analyses of the two species and F1 hybrids suggest that Idh-1 and Skdh-1/Pgm-2 are near the breakpoints on the two chromosomes involved in a reciprocal translocation for which the two species differ. One locus, Pgm-3, was detected only in C. annuum cultivars and is apparently the result of a duplication of Pgm-2 which codes for cytosolic phosphoglucomutase activity. Pgm-2 and Pgm-3 are not tightly linked (approximately 20% recombination) which supports the proposal that Pgm-3 originated from a mechanism other than unequal crossing over. A comparison of the linkage relationships of enzyme-coding genes in pepper with those of putative orthologous loci in tomato reveals that two linkage blocks, Est-1-Est-7 and Pgi-1-Est-4, may have remained intact since the divergence of Capsicum and Lycopersicon.

    Notes: Abstract The linkage relationship and chromosomal association of two genes in tomato coding for the respiratory enzymes alcohol dehydrogenase and phosphoglucomutase are reported. Also included is a description of where in the life cycle of the plant these genes are expressed. The finding that in pollen and germinating seeds most if not all of the detected activity of these two enzymes apparently arises from these genes suggests that the two isozymes might function in the same respiratory pathway.

    Notes: Abstract Three independent genes are responsible for 6-phosphogluconate dehydrogenase (6PGDH) activity in tomato. 6Pgdh-2, located on chromosome 12, codes for subunits of an active dimer which is restricted to the plastids. 6Pgdh-1, chromosome 4, and 6Pgdh-3, chromosome 5, code for subunits which form three dimers—two homodimers and an intergenic heterodimer. The latter three isozymes are found in the cytosol. 6Pgdh-1, 6Pgdh-2, and 6Pgdh-3 code for subunits with estimated molecular weights of 49,500, 50,500, and 51,200, respectively. The intergenic heterodimer encoded by 6Pgdh-1 and 6Pgdh-3 is thus composed of subunits that differ in length by approximately 15 amino acid residues. Divergence in the length and primary subunit structure may account for the lower thermal stability of the intergenic heterodimer compared with the corresponding homodimers. A limited survey of other solanaceous plant species suggests that the duplication of cytosolic 6PGDH-coding genes found in tomato may be widespread in the family.

    Notes: Abstract Subjecting tomato seedlings to anaerobic conditions results in expression of a previously undescribed Adh gene, Adh-2. Induction profiles were similar for all tissues, including roots, hypocotyls, cotyledons, and true leaves. In sharp contrast to ADH-1, ADH-2 showed no induction under anaerobic stress. The only time ADH-2 activity was expressed (under noninduced conditions) was during the early stages of embryogenesis. By late embryogenesis, ADH-2 activity approached a zero level, concomitant with a sharp rise in ADH-1 activity, which is found in the cotyledons of quiescent embryo. Despite striking differences in the regulation of these two genes, their homology is demonstrated in the ability of their enzyme subunits to form presumed intergenic heterodimers, which are visible during the transient period of embryogenesis when the polypeptides encoded by both genes are expressed. A multiple point linkage test using isozymic marker genes places the Adh-2 locus on chromosome 6 near Aps-1, whereas Adh-1 resides on chromosome 4.

    Notes: Abstract We have previously reported the isolation and characterization of tomato nuclear genes encoding two types of chlorophyll a/b-binding (CAB) polypeptides localized in photosystem (PS) I and two types of CAB polypeptides localized in PSII. Sequence comparisons shows that all these genes are related to each other and thus belong to a single gene family. Here we report the isolation and characterization of an additional member of the tomato CAB gene family, the single tomato nuclear gene, designated Cab-8, which encodes a third type of CAB polypeptide localized in PSI. The protein encoded by Cab-8 is 65% and 60% divergent from the PSI Type I and Type II CAB polypeptides, respectively. The latter two are 65% divergent from each other. Only some short regions of the polypeptides are strongly conserved. The Cab-8 locus maps to chromosome 10, 9 map units from Cab-7, the gene encoding the Type II PSI CAB polypeptide. The Cab-8 gene contains two introns; the first intron matches in position the single intron in the Type II PSII CAB genes and the second intron matches in position the second intron in the Type II PSI CAB gene. Like other CAB genes, Cab-8 is light-regulated and is highly expressed in the leaf and to a lesser extent in other green organs.

    Notes: Abstract A tomato lambda genomic library was screened with the human minisatellites 33.6 and 33.15. Similar tomato sequences are estimated to occur on average every 4000 kb. In thirteen hybridizing clones characterized, the size of minisatellite arrays varied between 100 bp and 3 kb. The structure of the repetitive elements is complex as the human core sequence is interspersed with other elements. In three cases, sequences similar to the human minisatellites were part of a higher-order tandem repeat. The chromosomal position of these sequences was established by ascertaining linkage to previously mapped RFLP markers. In contrast to the human genome, no clustering of minisatellite loci was observed in tomato. The fingerprints generated by hybridizing tomato minisatellites to genomic DNA of a set of cultivars were, in two cases, more variable than those obtained with 33.6 or 33.15. Two of the characterized probes detected 4–8 alleles of a single locus, which displayed 10–15 times more polymorphism than random RFLP clones. Some minisatellites contain di- and tri-nucleotide microsatellite repeated motifs which may account for the high level of polymorphism detected with these clones.

    Notes: Abstract Pulsed-field gel electrophoresis (PFGE) has been applied to analyze the rice nuclear genome. Probing 56 RFLP probes selected from the 12 rice chromosomes to PFGE blots of nine rare-cutting restriction enzymes revealed that there are relatively high numbers of ‘rare-cutting’ restriction sites in the rice genome. The average sizes of restriction fragments detected by single-copy probes are smaller than 200 kb for all of the rare-cutting restriction enzymes examined. Sizes of fragments detected by repetitive probes are variable, depending on the probes analyzed. By using PFGE, a tandemly repeated sequence, Os48, was found to be tightly linked to telomeric tandem repeats but not physically linked to r5s genes with which sequence homology had been observed. Relationships between genetic and physical distances have been established for three different chromosomal segments. In these regions 1 cm corresponds to ca. 260 kb on average. Analysis of a cluster of RFLP markers on chromosome 3 revealed that genetically clustered RFLP markers are also physically closely linked, suggesting that clustering of genetic markers may result in part from uneven distribution of single-copy sequences.
