    Notes: Beta-haemolytic streptococci are important human and animal pathogens: their genetic traits that are associated with the ability to infect human hosts remain, however, unclear. The surface protein, Lmb, mediates the adherence of Streptococcus agalactiae to human laminin. For further analysis of the corresponding gene, the adjacent genomic regions were sequenced. Lmb is localized on a putative composite transposon of 16 kb and is flanked by two copies of a novel insertion sequence element (ISSag2). It harbours the genes scpB and lmb, which are 98% identical with the respective genes of Streptococcus pyogenes. Analysis of the distribution of these genes and ISSag2 among 131 streptococcal strains revealed that all of the human isolates, but only 20% (12 of 61) of the animal isolates, contained scpB and lmb or their homologues. To investigate if the putative transposon can be mobilized, an erythromycin resistance marker was incorporated into the lmb gene of S. agalactiae. Screening for mutant strains with a regained susceptibility for erythromycin identified strains with a deletion of scpB, lmb, and one copy of ISSag2. We hypothesize that a horizontal gene transfer caused the exchange of scpB and lmb and that the ability of S. pyogenes, S. agalactiae and group C and G streptococcal strains to colonize or infect human hosts is dependent on their presence.

    Notes: Zusammenfassung Es wird über Untersuchungen berichtet, in denen der Einfluß von peroral verabreichter Adenosintriphosphorsäure auf die körperliche Leistungsfähigkeit geprüft wurde. Mittels eines Knippingschen Spiroergographen wurden 6 männliche und 6 weibliche Probanden vor und nach Applikation von 50 mg ATP (in 3 Dosen) 4 Belastungsuntersuchungen unterzogen, wovon 2 unter konstanten, 2 unter ansteigenden Arbeitsstufen stattfanden. Bei der konstanten Tätigkeit zeigte sich nach der ATP-Verabfolgung bei 10 von 12 Personen eine Verringerung des Sauerstoffverbrauches im steady state der Arbeit, ohne daß sich eine statistische Signifikanz nachweisen ließ. Hingegen wiesen der Wirkungsgrad der weiblichen Personen und der Summenwert von O2-Aufnahme im steady state und oxygen debt eine statistisch signifikante Verbesserung nach ATP-Einnahme auf. Der O2-Verbrauch unter ansteigender Belastung läßt keine Beeinflussung durch ATP erkennen. Die in Frage kommenden Ursachen eines möglichen sauerstoffeinsparenden ATP-Effektes werden diskutiert.

    Description: Streptococcus agalactiae (group B Streptococcus [GBS]) is a leading cause of sepsis in neonates. The rate of invasive GBS disease in nonpregnant adults also continues to climb. Aminoglycosides alone have little or no effect on GBS, but synergistic killing with penicillin has been shown in vitro . High-level gentamicin resistance (HLGR) in GBS isolates, however, leads to the loss of a synergistic effect. We therefore performed a multicenter study to determine the frequency of HLGR GBS isolates and to elucidate the molecular mechanisms leading to gentamicin resistance. From eight centers in four countries, 1,128 invasive and colonizing GBS isolates were pooled and investigated for the presence of HLGR. We identified two strains that displayed HLGR (BSU1203 and BSU452), both of which carried the aacA-aphD gene, typically conferring HLGR. However, only one strain (BSU1203) also carried the previously described chromosomal gentamicin resistance transposon designated Tn 3706 . For the other strain (BSU452), plasmid purification and subsequent DNA sequencing resulted in the detection of plasmid pIP501 carrying a remnant of a Tn 3 family transposon. Its ability to confer HLGR was proven by transfer into an Enterococcus faecalis isolate. Conversely, loss of HLGR was documented after curing both GBS BSU452 and the transformed E. faecalis strain from the plasmid. This is the first report showing plasmid-mediated HLGR in GBS. Thus, in our clinical GBS isolates, HLGR is mediated both chromosomally and extrachromosomally.
