    Notes: Floral induction in the long-day plant spinach (Spinacia oleracea L. cv. Nobel) was accompanied by a thickening of the plasma membrane. Densitometry analyses showed that the light space of the dark-light-dark pattern of the membrane was not changed upon photoinduction. Rather, the increase was due to an enhancement of the dark layer adjacent to the cell wall. Parallel analyses of protein and phospholipid composition revealed no marked changes in protein composition or biosynthetic rate, protein phosphorylation, glycolipids and/or phospholipids as a result of the 24 h of continuous light sufficient to induce flowering. Photoinduction, however, was accompanied by an increase in the relative amount of plasma membrane sterols which may be related to the membrane thickening.

    Notes: NADH-ferricyanide oxido-reductase (EC 1,6,99,3) of purified plasma membrane vesicles isolated by aqueous two-phase partition from segments of etiolated soybean [Glycine max (L.) Merr. cv. Williams] hypocotyls was used as a measure of plasma membrane redox activity. Elongation growth of hypocotyl segments floated on the solutions was determined in parallel. Cis-platinum (II) diammine dichloride (cis-platin), adriamycin and p-nitrophenylacetate, agents known to inhibit cell proliferation and plasma membrane redox activities in mammalian cells inhibited both NADH-ferricyanide oxido-reductase of the isolated membrane vesicles and elongation growth of intact hypocotyl segments. Auxin(2,4-dichlorophenoxyacetic acid)-induced growth of the isolated segments was inhibited preferentially at drug concentrations where control growth was affected only slightly. The findings suggest a connection between plasma membrane redox reactions and the control of elongation growth in plants.

    Notes: A generalized two-step and interdependent control of basic and acidic peroxidases (EC 1.11.1.7) is observed in plant responses to different physical and chemical stimuli. An interpretative model consisting of a pathway of reactions is presented on the basis of our own data and the literature. Stress-induced membrane depolarization would generate different species of free radicals and peroxides, which in turn initiate lipid peroxidation. The degradation of cell membranes is suggested to bring about rapid changes in ionic fluxes (especially release of K+ which would result in an enhanced endogenous Ca/K ratio) and in leakage of solutes (among them electron donors such as ascorbic acid and phenolic substances). The increased intracellular relative calcium level results in: 1) activated secretion of basic peroxidases into the free space where, in association with the electron donors and maybe with the circulating indole-3-acetic acid (IAA), they eliminate the peroxides; and 2) facilitated binding of basic peroxidases to membrane structures allowing a role as 1-aminocyclopropane-1-carboxylic acid (ACC)-oxidases. The resulting IAA and ACC oxidase-mediated changes in ethylene production would further induce (this time through the protein synthesis machinery) an increase in activity of phenylalanine ammonia-lyase (EC 4.31.5) and acidic peroxidases. The resulting lignification and cell wall rigidification determines the growth and/or the developmental response to the initial stress.

    Notes: Ca2+ and Mn2+ activate the conversion of 1-aminocyclopropane-1-carboxylic acid (ACC) by root microsomes of Vicia lens as they do in other similar systems. The preparation of microsomes in the presence of Mn2+ greatly increases their ability to convert ACC into ethylene, without addition of Mn2+ in the reaction mixture. Ca2+ does not have this property. The effect could not be attributed to Mn2+ entrapping into membrane vesicles (sonication followed by repelleting had no effect) but, possibly, in part to Mn2+-mediated binding to microsomes of a soluble factor favouring the conversion of ACC to C2H4. Although no direct correlation could be established in vitro between ethylene-forming-enzyme (EFE) and peroxidase activities, some soluble peroxidases might be this soluble factor. Mn2+ favoured attachment to membranes of some peroxidase activity from the soluble fraction and from commercial HRP and lipoxygenase. This binding effect of Mn2+ cannot be readily distinguished from its role in the generation of a chain of free radicals and in redox mechanisms.

    Notes: Habituated calli have long been classified as neoplasms together with tumors from different origins. The general opinion is that habituation is a reversible process with an epigenetic basis. This is probably true in most cases examined. However, we show here that there might be several degrees of habituation, which can be considered as steps of a neoplastic progression leading to cancerisation in the absence of an introduced oncogenic pathogen. Cell rejuvenation, loss of the capacity to organize meristematic centers, and loss of totipotency are proposed to define plant cancer through this neoplastic progression of a callus.Habituated tissues share many morphological and biochemical similarities with so-called vitreous shoots from micropropagation. Vitrification and hyperhydric malformations of shoots raised in vitro may be considered as steps of another neoplastic progression, which leads to cancerisation also in the absence of introduced oncogenic pathogens. In this case death of the whole organism occurs either through direct apex necrosis or indirectly, from the loss of the capacity for the primary meristems to function normally, which gives rise to completely anarchic structures. As in the animal kingdom, carcinogenesis in plants is the final result of a multistep process involving the irreversible conversion of a stem cell to a terminal-differentiation-resistant cell.

    Notes: A membrane fraction enriched in plasma membrane and tonoplast vesicles was isolated from green leaves of Spinacia oleracea L. and subjected to subfractionation by free-flow electrophoresis. The most electronegative membrane vesicle fraction collected after the free-flow electrophoretic separation was identified as derived from tonoplast, while the least electronegative fraction was identified as derived from plasma membrane. The identification of the fractions was based on membrane morphology, and on the presence or absence of biochemical markers. The plasma membrane fraction was enriched in thick (9–11 nm) membranes which bound N-1-naphthylphthalamic acid (NPA), and reacted with phosphotungstic acid at low pH on thin sections for electron microscopy. The tonoplast fraction was enriched in vesicles with 7–9 nm thick membranes that neither bound NPA nor reacted with phosphotungstic acid at low pH. Both the plasma membrane and the tonoplast fraction were about 90% pure, with a cross-contamination of not more than 2%. Membrane vesicles originating from dictyosomes, endoplasmic reticulum, mitochondria, plastids, or peroxisomes contaminated the plasma membrane and the tonoplast fractions by a few % only. In leaves of photoinduced plants (24 h light period), the plasma membranes were thicker than in control leaves (8 h light, 16 h dark). The plasma membrane fraction obtained from photo-induced leaves by free-flow electrophoresis retained this increase in thickness, showing not only that photoinduction alters plasma membrane structure, but also that this change is stable to isolation.

    Notes: The effect of in vitro irradiations on ihe pelletability of peroxitlase activity was tested in extracts prepared from leaves of light-grown spinach (Spinacia oleracea). It appeared that far red light increased the pelletable activity and that red light reversed this effect. This phytochrome-mediated effect occurred in a particulate fraction which banded at 39% (w/w) sucrose on an isopycnic density gradient.

    Notes: Abstract. We investigated in a colon adenocarcinoma cell line, the exclusive role of extracellular matrix (ECM) components in the absence of soluble factors regarding the integrin clustering processes, and their implication in cell adhesion, spreading and organization of the actin cytoskeleton. Caco-2 cells were shown to express at the plasma membrane 11 integrins, some of which (e.g. α3β1, α5β1, α6β1/β4, α8β1 and αvβ1/β5/β6) were identified for the first time in this cell line. Cell adhesion and spreading processes were governed essentially by lamellipodium, the regulation of which was shown to be induced by two types of integrin clustering processes mediated by ECM proteins alone. During these phenomena, α2β1, αvβ6 and α6β1 integrins, the Caco-2 cell specific receptors of type IV collagen, fibronectin and laminin, respectively, were clustered in small focal complexes (point contacts), whereas αvβ5, the vitronectin receptor in this cell line, was aggregated in focal adhesions. The two levels of integrin clustering induced only F-actin cortical web formation organized in thin radial and/or circular filaments. We conclude thus that ECM components per se through their action on integrin clustering are involved in cell adhesion, cortical actin cytoskeleton organization and cell spreading.

    Notes: Abstract When cultured on collagen coated nitrocellulose filters, thyroid epithelial cells form morphologically and functionally polarized monolayers. The bioelectric parameters of these monolayers were measured after mounting in Ussing chambers; transepithelial potential (V ab), short circuit current (I sc) and transepithelial resistance were respectively 12±1 mV (apical side negative), 3.8±0.2 μA cm−2 and 3250±214 Ω cm2 (mean±SEM,n=75). Eighty two percent of the short circuit current was related to sodium absorption as shown by inhibition by apical amiloride (K m=0.2 μM) and by basal ouabain (K 1/2=0.3 μM). Amphotericin B (5–25 μg/ml) added to the apical bath increasedI sc suggesting an apical rate-limiting step. Step by step replacement of choline by Na+ in a Na+-free medium resulted in a progressive increase inV ab andI sc with half maximal effect at 20±1 mM Na+. Thyrotropin (TSH) increasedI sc andV ab in a biphasic way with a transient maximum after 5 min and a plateau after 20 min (about four times the basal level at 100 μU/ml TSH). This increase in sodium transport was also inhibited by apical amiloride. Thus, in culture, the thyroid cell monolayer behaves as a tight sodium absorbing epithelium controlled by TSH, with a rate limiting apical sodium channel as the entry mechanism and a basolateral Na+, K+-ATPase as the electromotive force.

    Notes: Abstract Poplar shoots raised in vitro were induced to root by incubation on an auxin (NAA) containing medium for 7 h. After 13 days on an auxin-free medium, 97% of the treated shoots had rooted. The introduction of known antiauxins (PCIB, PBA, POAA) into the rooting expression auxin-free medium, after the 7-h induction by NAA, completely (PCIB and PBA) or severely (POAA) inhibited rooting. The exclusion of calcium from the expression auxin free medium reduced the percentage of rooting by about 42%. The inhibition was still higher in the presence of EGTA, a calcium chelator. Lanthanum chloride, a calcium channel blocker, also completely inhibited rooting, when incorporated into the auxin free medium, with or without calcium. These results support previous hypotheses about the implication of both endogenous auxin and calcium in the late phases of the adventitious rooting process.
