    Notizen: Abstract In meiosis of human males DNA is packaged along pachytene chromosomes about 20 time more compactly than in meiosis of yeast. Nevertheless, a human-derived yeast artificial chromosome (YAC) shows the same degree of compaction of DNA as endogenous chromosomes in meiotic prophase nuclei of yeast. This suggests that in yeast meiosis, human and yeast DNA adopt a similar organization of chromatin along the pachytene chromosome cores. Therefore meiotic chromatin organization does not seem to be an inherent chromosomal property but is governed by the host-specific cellular environment. We suggest that there is a correlation between the less dense DNA packaging and the increased rate of recombination that has been reported for human-derived YACs as compared with human DNA in its natural environment.

    Notizen: Abstract An extensive synaptonemal complex (SC) is found at pachytene in whole mount spread preparations of a haploid yeast, Saccharomyces cerevisiae, strain. Whereas unsynapsed axial elements are present only in a few nuclei, in others non-homologous synapsis involves virtually the whole chromosome set. This suggests that homology is not an indispensable precondition for SC formation in yeast but that chromosomes engage in non-homologous synapsis if no homologous partner is available. Recent evidence that in the sporulation deficient yeast mutants rad50 and mer1 axial elements do form but remain unsynapsed in the majority of nuclei is discussed in the light of the above findings.

    Notizen: Abstract Non-isotopic high resolution in sity hybridization was applied to cytological preparations of sporulating yeast cells. Ribosomal DNA (rDNA) and chromosome V-specific recombinant lambda clones were used to tag individual chromosomes and chromosome subregions. This allowed the study of chromosome behaviour during early meiotic prophase. It was found that chromatin becomes condensed and homologous DNA sequences then appear to become aligned prior to synaptonemal complex formation.

    Notizen: Abstract Chiasmata in male meiosis of Allium flavum from early diplotene to metaphase I are found to occur almost exclusively very close to the terminal and intercalary C-bands. Heterozygosity of intercalary C-bands, where one homologue has one band missing, allows the direct proof that chiasmata do not terminalise and therefore mark the point of exchange.

    Notizen: Abstract Synaptonemal complex (SC) formation in microsporocytes of Allium ursinum is severely affected by exposure of plants to 35° C for 30 h or longer. In spread preparations made from fresh and freeze-conserved material it was found that a high proportion of meiocytes is arrested at leptotene and shows no synapsis at all. In another group of nuclei synaptonemal polycomplex-like structures do occur between converging axial elements at presumed rudimentary SC initiation sites. Axial elements are virtually always thickened at these sites which seem to involve primarily heterologous chromosomes. A third situation is seen in nuclei where two or more lateral elements are engaged in the formation of longer stretches of aberrant SCs. These feature surplus material filling the central space. It may be assumed that this abnormal condition precludes crossing over and hence may be one of several ways by which elevated temperatures cause the chiasma reduction described here for A. ursinum and reported for several other organisms in the literature.

    Notizen: Abstract Quantitatively evaluated C-banding karyograms are presented forAllium carinatum, A. carinatum ssp.pulchellum, andA. flavum of the sectionCodonoprasum Reichenb. Accurate measurements revealed that constitutive heterochromatin (C-bands) is probably additional chromosomal material. The distribution of the C-heterochromatin follows the principle of the equilocal heterochromatin-distribution byHeitz (1933). Furthermore, the pattern shows a relationship to the relative arm-length of the chromosomes in the karyotype. Fluorochrome banding revealed various heterochromatintypes. The C-band patterns ofAllium cupani (sect.Scorodon Koch) andA. vineale (sect.Allium), which are also rich in heterochromatin, are described.

Accumulating information about structures like lateral element thickenings and -doublings allows to draw conclusions about their nature. The occurrence of discrete intercalary and terminal homologous associations prior to synapsis is confirmed for a range ofAllium species and it is argued that they are a general phenomenon. Several hypotheses on homologous recognition and/or attraction are discussed in the light of the observations on homologous alignment inAllium.

    Notizen: Abstract B-Chromosomes ofAllium flavum, A. stamineum andA. carinatum were C-banded. InA. flavum different types of B's were found, one of them possessing a nucleolus organizer. In the B's ofA. flavum andA. stamineum the banding patterns resemble those found in the standard chromosomes. The B inA. carinatum is only terminally banded. InA. flavum chiasma localization in the B's appears to be dependent on C-band location, just as is the case in the A-chromosomes. An increase in B's was found to cause a small (but significant) increase in chiasma frequency in the PMC's. This may result from an alteration in the nucleotype by the B's.
