    Description: The tools of geodesy have the potential to transform the Ocean Observing System. Geodetic observations are unique in the way that these methods produce accurate, quantitative, and integrated observations of gravity, ocean circulation, sea surface height, ocean bottom pressure, and mass exchanges among the ocean, cryosphere, and land. These observations have made fundamental contributions to the monitoring and understanding of physical ocean processes. In particular, geodesy is the fundamental science to enable determination of an accurate geoid model, allowing estimate of absolute surface geostrophic currents, which are necessary to quantify ocean’s heat transport. The present geodetic satellites can measure sea level, its mass component and their changes, both of which are vital for understanding global climate change. Continuation of current satellite missions and the development of new geodetic technologies can be expected to further support accurate monitoring of the ocean. The Global Geodetic Observing System (GGOS) of the International Association of Geodesy (IAG) provides the means for integrating the geodetic techniques that monitor the Earth's time-variable surface geometry (including ocean, hydrologic, land, and ice surfaces), gravity field, and Earth rotation/orientation into a consistent system for measuring ocean surface topography, ocean currents, ocean mass and volume changes. This system depends on both globally coordinated ground-based networks of tracking stations as well as an uninterrupted series of satellite missions. GGOS works with the Group on Earth Observations (GEO), the Committee on Earth Observation Satellites (CEOS) and space agencies to ensure the availability of the necessary expertise and infrastructure. In this white paper, we summarize the community consensus of critical oceanographic observables currently enabled by geodetic systems, and the requirements to continue such measurements. Achieving this potential will depend on merging the remote sensing techniques with in situ measurements of key variables as an integral part of the Ocean Observing System.

    Notes: This work investigates the variations of the infrared transmission spectra of yttrium-hydride films YHx during the hydrogen loading process for the frequency range 500–4000 cm−1. The results indicate that the transmittance slightly decreases in the dihydride phase, followed by a significant increase in the trihydride phase. In addition, the carrier concentration decreases, whereas the carrier relaxation time increases with hydrogen content. The hydrogen vibration modes at interstitial sites are completely screened in the dihydride phase. The screening effect decreases as the system goes through the metal–insulator transition. Moreover, the screening effect can be continuously tuned by simply varying the hydrogen content in the yttrium-hydride system. Analysis indicates that the absorption intensity of the vibration mode depends on the carrier concentration. This effect can be used as a diagnostic tool for estimating the carrier concentration and hydrogen content in rare-earth hydrides. © 2001 American Institute of Physics.

    Notes: Hybrid resonant optical reflector lasers of less than 1 cm length are described. These external cavity devices operate with very narrow linewidth approaching the resonator-loss-limited minimum. Best linewidth results are 7.1 kHz at 1.3 μm and 24 kHz at 1.55 μm.

    Notes: Frequency modulation (FM) response of a 1.3 μm two-electrode distributed feedback laser is studied in detail. We found that by adjusting the injection current applied to the modulated section, the FM bandwidth can be increased to a value as high as 900 MHz. However, as the bandwidth increases, the FM sensitivity decreases proportionally. This behavior is described satisfactorily by below-threshold carrier rate equation and shown to be closely related to the injected carrier lifetime.

    Notes: Summary Carbon isotope ratios were used to survey the distribution of photosynthetic pathways among taxa, the relationship between photosynthetic pathway and habitat light levels, and the relationship between intercellular CO2 levels of C3 plants and habitat light levels within a subtropical monsoon forest in southern China. Of 128 species, most (94) possessed the C3 photosynthetic pathway; 33 species possessed the C4 pathway and all of these were restricted to high light locations. There was one epiphytic CAM species. The C3 species were classified as occurring in open, intermediate, and closed canopy sites. Among C3 species, carbon isotope ratios tended to become more negative with decreasing light availability in the habitat.

    Notes: Summary Pancreatic islet transplantation shows promise as a future method for the treatment of insulin-dependent diabetes mellitus. However, as with reports of whole organ transplants in man, there are likely to be too few human donor organs. One potential approach to try and circumvent this problem is to seek alternative sources of pancreatic tissue. We have studied and developed a technique for isolating functional islets from one-day-old piglets with this in mind. Twelve pancreata, mean weight 1.27±0.36 g, were retrieved aseptically. The islets were isolated using a collagenase digestion procedure. An average of 116,000±45,000 islets/g tissue were isolated from each pancreas. After 24–48 h of culturing, a membrane-boundary was formed around the islets. Dithizone (DTZ) staining revealed greater than 50% of the presumed islets contained at least 5–6 red stained cells (i.e. insulin-containing cells). The insulin content of islet-like structures containing 50%, 10%, and 0% of DTZ stained cells was measured and revealed 32.1, 10.8 and 2.9 µIU, respectively, of immuno-reactive-insulin. In static-perfusion assays, a biphasic insulin secretion was observed in response to increased glucose stimuli. Preliminary studies showed that these islets were able to reverse the hyperglycemic condition of streptozotocin-induced diabetic rats. The present study indicates that one-day-old piglet islets can be utilized as the source of functioning islet cells. Isolation of these cells is reproducible and there appears to be no advantage technically and economically in using fetal pig pancreatic tissue rather than tissue from neonatal piglets for the purpose of obtaining pancreatic islets.

    Description: The DNA damage response (DDR) involves both the control of DNA damage repair and signaling to cell cycle checkpoints. Therefore, unraveling the underlying mechanisms of the DDR is important for understanding tumor suppression and cellular resistance to clastogenic cancer therapeutics. Because the DDR is likely to be influenced by chromatin regulation at the sites of DNA damage, we investigated the role of heterochromatin protein 1 (HP1) during the DDR process. We monitored double-strand breaks (DSBs) using the H2AX foci marker and found that depleting cells of HP1 caused genotoxic stress, a delay in the repair of DSBs and elevated levels of apoptosis after irradiation. Furthermore, we found that these defects in repair were associated with impaired BRCA1 function. Depleting HP1 reduced recruitment of BRCA1 to DSBs and caused defects in two BRCA1-mediated DDR events: (i) the homologous recombination repair pathway and (ii) the arrest of cell cycle at the G 2 /M checkpoint. In contrast, depleting HP1 from cells did not affect the non-homologous end-joining (NHEJ) pathway: instead it elevated the recruitment of the 53BP1 NHEJ factor to DSBs. Notably, all three subtypes of HP1 seemed to be almost equally important for these DDR functions. We suggest that the dynamic interaction of HP1 with chromatin and other DDR factors could determine DNA repair choice and cell fate after DNA damage. We also suggest that compromising HP1 expression could promote tumorigenesis by impairing the function of the BRCA1 tumor suppressor.
