    Description / Table of Contents: Summary Tick-borne encephalitis (TBE) is the most severe arbovirus disease transmitted by ticks. The mortality of the central European form is 0.7–2%. Active immunisation is recommended for endemic regions. We report on 4 patients with TBE aquired in Middle-Hessen between 1994 and 1997 (2 in 1997). After repeated CSF and serum testing the TBE-specific antibodies were found in all 4 cases. In one case there was also evidence for a prior infection with borrelia burgdorferi. The results of the initial CSF-analysis were atypical in 2 cases (high cell count of 136 cells/mm3, total protein up to 1.5 g/l). The endemic region for TBE has expanded in northern direction into Middle-Hessen, a region in which Lyme borreliosis is also endemic. Thus, true double infections are possible. This and the initially frequently atypical CSF-findings make the differential diagnosis difficult. Therefore, repetitive CSF and blood examinations are recommended.
    Notes: Zusammenfassung Die FSME ist die häufigste, schwere durch Zecken übertragene Arbovirose Mitteleuropas. Die Letalität der in Teilen Deutschlands vorkommenden westlichen Variante beträgt 0,7–2%. Eine aktive Immunisierung ist möglich und für exponierte Gruppen in Endemiegebieten empfohlen. In Mittelhessen waren bisher keine Endemiegebiete bekannt. Wir berichten über 4 Patienten mit in Mittelhessen erworbenen FSME-Infektionen und typischem Krankheitsverlauf im Zeitraum von 1994–1997 (davon zwei 1997). Bei allen Patienten ließ sich die Infektion serologisch sichern, bei einem der 4 Patienten bestand zusätzlich der Hinweis auf eine abgelaufene oder möglicherweise gleichzeitige Borrelia-burgdorferi-Infektion. Der initiale Liquorbefund war in 2 Fällen atypisch, der Nachweis von spezifischen Antikörpern war im Verlauf in allen Fällen möglich. Mittelhessen ist seit 1997 als FSME-Endemiegebiet anzusehen, welches sich weiter nach Nordwesten auszubreiten scheint. Das gleichzeitige Vorkommen von Borrelia burgdorferi erschwert die Diagnostik und muß an Doppelinfektionen denken lassen.

    Notes: Summary Yield and properties of mitochondria, isolated from hind leg muscles of normal hamsters, are different, when 4 preparation procedures are applied simultaneously: 1. mechanical homogenization of muscle with a glass-Teflon homogenizer followed by rehomogenization of the isolated nuclear fraction for 1–6 times. The procedure yields impure mitochondria showing the highest amounts of mitochondrial enzymes (50–65% of total), protein and DNA; the isolated mitochondria exhibit disrupted outer membranes and loosened inner membranes causing a better supply with substrates of enzymes located at the inner membrane and in the matrix. 2. Single mechanical homogenization of muscle for increasing periods (2–40 min) yields lower amounts of all constituents (25–40% of total). 3. The yields of Nagarse treatment with one step of 2 min mechanical homogenization range between 20–35% of total. Mitochondria isolated with the latter procedure exhibit compact membranes and are very pure, but still contaminated with Nagarse, causing destructions of mitochondrial membranes already after 24 h storage in buffer with sucrose and mannitol at 2°C. 4. One single Nagarse treatment of muscle for 40 min produces a minor yield of these constituents. Therefore, at least two different isolation procedures are proposed for optimum investigation of skeletal muscle mitochondria.

    Notes: Abstract With cartilage slices from calf ribs, cGMP as well as cAMP accelerate dose-dependently and specifically label rates of Ch-4-,-6-S protein; they slightly elevate rates of anaerobic glycolysis dose-independently and unspecifically, similar to their 5-monophosphate compounds. cAMP, but not cGMP, slightly stimulates labeling of total protein dosedependently. Guanosine and adenosine (as well as adenine) accelerate more significantly all three anabolic processes in the order Ch-4-,-6-S protein formation ≧ total protein labeling 〉 anaerobic glycolysis. Acceleration of some of the processes rises further after adding theophylline or SQ 20.009, dependeing on the nucleoside used. diBu-cAMP (but not 8-Br-cAMP) stimulates the three processes more than cAMP; diBu-cAMP and 8-Br-cGMP alone increase the labeling rates of protein more than cGMP. cCMP and cIMP slightly accelerate at least one of the three processes doseindependently and unspecifically, similar to their 5-monophosphate compounds. cUMP was almost inactive. The results point to specific and unspecific effects of cGMP similar or different to those of cAMP.

    Notes: Abstract A 5 min equilibration of 0.250 mL serum sample with room air containing 5.6% CO2, by placing the gas tube 〈 1 cm above the sample-surface, enables a standardized determination of calcium ion concentration (cCa2+), referred to pH 7.40 and 37 °C, in aerobically handled specimens. Using this method, standardized measurement of calcium ions in serum is possible with a bias less than 2%, even in samples with pH-changes acquired during extended storage.

    Notes: Abstract Different mixtures from a serum pool and a cerebrospinal fluid (CSF) pool were used as models to study CSF contamination in secretions by determining two CSF specific proteins: β-trace protein (β-TP) and the asialo-transferrin (a-Tf) band which was detected by isoelectric focusing (IEF) with Tf specific immunofixation. β-TP and total Tf were measured immunonephelometrically. Secretion/serum ratios of β-TP content 〉 2.0 indicated CSF contaminations with ≥ 5% (v/v) CSF; this was confirmed by detecting the a-Tf band by IEF. Reliable ¶a-Tf bands were only revealed with secretion/serum rations of Tf contents 〈 0.1, indicating an interference of major sialo-Tf fractions with the a-TF band detection in the sample. For CSF detection in rhinorrhea and otorrhea, complementary use of β-TP assay and a-Tf assay is recommended. Preanalytically, dilution or concentration of the sample as well as denaturation of Tf and β-TP should be prevented by optimizing sample collection.

    Description / Table of Contents: Zusammenfassung In Hautschnitten von 16 Gesunden und 10 Patienten mit progressiver Sklerodermie wurde die Biosynthese von Glycosaminoglycanen (GAG) und Proteinen mit Hilfe autoradiographischer, histochemischer und radiobiochemischer Methoden in vitro unter aeroben und anaeroben Bedingungen untersucht. Die Autoradiographie mit radioaktiven Vorstufen der Sulfopolysacharid- (35SO4,3H-Glucosamin) und Proteinbiosynthese (3H-Prolin) indizierte deren Lokalisation in Epithelund Coriumzellen. Die Einbauraten aller 3 Vorstufen waren in der Haut von Sklerodermiepatienten 2- bis 20fach gegenüber Kontrollen gesteigert. Die Auszählung der Silberkörner pro 1000 Zellen läßt folgende Einbauverhältnisse im Corium zu Stratum spinosum zu Stratum basale für35SO4 erkennen: Kontrollen: 2,7:2,1:1,0, Sklerodermie: 12,3:2,7:1,0; für3H-Glucosamin: Kontrollen: 8,3:3,2:1,0, Sklerodermie: 22,0:5,6:1,0; für3H-Prolin: Kontrollen: 21,1:7,2:1,0, Sklerodermie: 15,6:3,4:1,0. Die in der Haut von Sklerodermiepatienten histochemisch nachweisbar vermehrte Ablagerung saurer und sulfatierter GAG stand in guter Korrelation mit den autoradiographisch nachweisbaren Syntheseorten (Stratum spinosum, Coriumbindegewebszellen, Hautanhangsgebilde, Gefäßwände). Damit kommen als Syntheseort von GAG Zellen ektodermalen und mesenchymalen Ursprungs in Frage. Die radiobiochemische und Infrarot-Spektralanalyse des Syntheseproduktes von Hautzellen zeigte bei Gesunden eine Sulfopolysaccharidfraktion mit überwiegend am C4- weniger am C6-Atom veresterten Sulfatgruppen, bei Sklerodermiepatienten eine Zunahme der am C6-Atom veresterten Sulfatgruppen. Der Uronsäuregehalt dieser Fraktion nahm in der Haut von Sklerodermiepatienten 4,4fach zu. Die spez. Radioaktivität (IpM/µMol Uronsäure) von Sulfatestergruppen, Hexosaminen und3H-Prolin war bei Sklerodermiepatienten gegenüber Kontrollen um 3-, 1,1- bzw. 1,9fach erhöht. Hieraus wird auf eine bevorzugte Neubildung von Sulfopolysacchariden in der Haut von Sklerodermiepatienten geschlossen. Radioaktive Einbauraten, anaerobe Glycolyserate und Hexosaminausscheidung im Urin zeigen eine vom Krankheitsverlauf abhängige Steigerung.
    Notes: Summary The biosynthesis of glycosaminoglycans and proteins was studied in skin slices of 16 healthy subjects and 10 patients with progressive skleroderma, using autoradiographic, histochemical and radiobiochemical methods in vitro under aerobic and anaerobic conditions. The sulphopolysaccharide and protein biosynthesis was localized autoradiographically in the cells of the epithelium and of the corium using35SO4 and3H-glucosamine as precursors of the sulphopolysaccharide biosynthesis and3H-proline as a precursor of the proteinbiosynthesis. Incorporation rates of these 3 precursors were 2 to 20 times higher in the skin of patients than in that of controls. The following proportions of the incorporation rates (silver grains per 1000 cells) were found for35SO4 incorporation in controls: corium to stratum spinosum to stratum basale as 2.7:2.1:1.0; in patients 12.3:2.7:1.0; for3H-glucosamine in controls 8.3:3.2:1.0 and in patients 22.0:5.6:1.0; for3H-proline in controls 21.0:7.2:1.0 and in patients 15.6:3.4:1.0. An increased deposition of acid and sulphated glycosaminoglycans was demonstrated with histochemical methods in the skin of patients with progressive skleroderma. It correlated well with the spots of glycosaminoglycan biosynthesis as shown autoradiographically in human skin (stratum spinosum, cells in corium and in wall of little vessels, perifollicular space). These findings evidenced a biosynthesis of glycosaminoglycans taking place in cells of ectodermal as well as of mesodermal origin. Radiobiochemical and infrared analysis of the in vitro synthesized product of human skin cells of healthy persons indicated a sulphopolysaccharide fraction exhibiting mainly C4- (less C6-) esterified sulphate groups whereas a relative increase of C6-esterified sulphate groups was found in patients with skleroderma. The content of uronic acid grew up to the 4.4fold in the skin of patients. The specific radioactivity (IpM/µmole uronic acid) of sulphate ester groups, radioactive hexosamine and proline residues increased to 3-, 1.1- and 1.9fold in patients compared to controls. The incorporation rate of radioactive precursors, the rate of anaerobic glycolysis and the excretion of hexosamine in urine depended on the malignancy of skleroderma.
