    Notes: Abstract The reduction and fragmentation of forest habitats is expected to have profound effects on plant species diversity as a consequence of the decreased area and increased isolation of the remnant patches. To stop the ongoing process of forest fragmentation, much attention has been given recently to the restoration of forest habitat. The present study investigates restoration possibilities of recently established patches with respect to their geographical isolation. Because seed dispersal events over 100 m are considered to be of long distance, a threshold value of 100 m between recent and old woodland was chosen to define isolation. Total species richness, individual patch species richness, frequency distributions in species occurrences, and patch occupancy patterns of individual species were significantly different among isolated and nonisolated stands. In the short term no high species richness is to be expected in isolated stands. Establishing new forests adjacent to existing woodland ensures higher survival probabilities of existing populations. In the long term, however, the importance of long-distance seed dispersal should not be underestimated because most species showed occasional long-distance seed dispersal. A clear distinction should be made between populations colonizing adjacent patches and patches isolated from old woodland. The colonization of isolated stands may have important effects on the dynamics and diversity of forest networks, and more attention should be directed toward the genetic traits and viability of founding populations in isolated stands.

    Description: We investigated the effects of fire on population growth rate and invasive spread of the perennial tussock grass Molinia caerulea. During the last decades, this species has invaded heathland communities in Western Europe, replacing typical heathland species such as Calluna vulgaris and Erica tetralix. M. caerulea is considered a major threat to heathland conservation. In 1996, a large and unintended fire destroyed almost one-third of the Kalmthoutse Heide, a large heathland area in northern Belgium. To study the impact of this fire on the population dynamics and invasive spread of M. caerulea, permanent monitoring plots were established both in burned and unburned heathland. The fate of each M. caerulea individual in these plots was monitored over four years (1997–2000). Patterns of seed dispersal were inferred from a seed germination experiment using soil cores sampled one month after seed rain at different distances from seed-producing plants. Based on these measures, we calculated projected rates of spread for M. caerulea in burned and unburned heathland. Elasticity and sensitivity analyses were used to determine vital rates that contributed most to population growth rate, and invasion speed. Invasion speed was, on average, three times larger in burned compared to unburned plots. Dispersal distances on the other hand, were not significantly different between burned and unburned plots indicating that differences in invasive spread were mainly due to differences in demography. Elasticities for fecundity and growth of seedlings and juveniles were higher for burned than for unburned plots, whereas elasticities for survival were higher in unburned plots. Finally, a life table response experiment (LTRE) analysis revealed that the effect of fire was mainly contributed by increases in sexual reproduction (seed production and germination) and growth of seedlings and juveniles. Our results clearly showed increased invasive spread of M. caerulea after fire, and call for active management guidelines to prevent further encroachment of the species and to reduce the probability of large, accidental fires in the future. Mowing of resprouted plants before flowering is the obvious management tactic to halt massive invasive spread of the species after fire.

    Description: Quantifying the abundances of fungi is key to understanding natural variation in mycorrhi-zal communities in relation to plant ecophysiology and environmental heterogeneity. High-throughput metabarcoding approaches have transformed our ability to characterize and com-pare complex mycorrhizal communities. However, it remains unclear how well metabarcodingread counts correlate with actual read abundances in the sample, potentially limiting their useas a proxy for species abundances. Here, we use droplet digital PCR (ddPCR) to evaluate the reliability of ITS2 metabarcodingdata for quantitative assessments of mycorrhizal communities in the orchid speciesNeottiaovatasampled at multiple sites. We performed specific ddPCR assays for eight families oforchid mycorrhizal fungi and compared the results with read counts obtained from metabar-coding. Our results demonstrate a significant correlation between DNA copy numbers measured byddPCR assays and metabarcoding read counts of major mycorrhizal partners ofN. ovata,highlighting the usefulness of metabarcoding for quantifying the abundance of orchid mycor-rhizal fungi. Yet, the levels of correlation between the two methods and the numbers of falsezero values varied across fungal families, which warrants cautious evaluation of the reliabilityof low-abundance families. This study underscores the potential of metabarcoding data for more quantitative analysesof mycorrhizal communities and presents practical workflows for metabarcoding and ddPCRto achieve a more comprehensive understanding of orchid mycorrhizal communities
