    Notes: Abstract. Four type-1 (single-chain) ribosome-inactivating proteins (RIPs), with isoelectric points between 9.5 and 9.7, were isolated from leaves of Phytolacca dioica L. The purification procedure furnished the four proteins with an overall yield of about 16% and separated them from a protein of 29 407 ± 2 Da, as determined by electrospray mass spectrometry, whose N-terminal amino acid sequence differed from that of pokeweed (Phytolacca americana L.) leaf chitinase (PLC-B) by only one amino acid (R17I). The four RIPs (PD-L1 to PD-L4) inhibited protein synthesis by a rabbit reticulocyte lysate with 50% inhibition at the picomolar level, and produced the β-fragment, diagnostic of the specific enzymatic action of RIPs, on yeast ribosomes. Comparison of their N-terminal sequences, up to residue 45, showed that PD-L1 is identical to PD-L2 [designated the isoleucine (Ile) form from the N-terminal residue] and PD-L3 is identical to PD-L4 [designated the valine (Val) form from the N-terminal residue] and that there are 35 identical residues between the two forms. Furthermore, the Val form presents the same number of identical residues as PD-S2, an RIP isolated from the seeds of the same plant. With the exception of PD-L4, the purified RIPs gave a positive reaction when stained for sugars on SDS-PAGE gels and, when analyzed by electrospray mass spectrometry, had Mr values of 32 715 ± 1 (PD-L1), 31 542 ± 1 (PD-L2), 30 356 ± 1 (PD-L3) and 29 185 ± 1 Da (PD-L4). The 1171 kDa difference in Mr, within the same RIP form, could be due to glycosylation. Like leaf saporins and many other RIPs, the four RIPs released several adenines from poly(A), herring sperm DNA and rRNA 16S + 23S, thus acting as polynucleotide:adenosine glycosidases. This property was less pronounced in PD-L1 and PD-L3 than in PD-L2 and PD-L4, respectively. The proteins PD-L1 and PD-L4 showed 3.7% reactivity with the antiserum anti-dianthin 32 and no reactivity with antisera to PAP-R saporin-S6, momordin I and even PD-S2, an RIP isolated from the seeds of the same plant. Protein PD-L4 showed 12.5% cross-reactivity with anti-PD-L1, while the opposite cross-reactivity was 100%.

    Description: This paper presents a methodology taking advantage of the GPOD-SBAS service to study the surface deformation information over high mountain regions. Indeed, the application of the advanced DInSAR over the arduous regions represents a demanding task. We implemented an iterative selection procedure of the most suitable SAR images, aimed to preserve the largest number of SAR scenes, and the fine-tuning of several advanced configuration parameters. This method is aimed at minimizing the temporal decorrelation effects, principally due to snow cover, and maximizing the number of coherent targets and their spatial distribution. The methodology is applied to the Valle d’Aosta (VDA) region, Northern Italy, an alpine area characterized by high altitudes, complex morphology, and susceptibility to different mass wasting phenomena. The approach using GPOD-SBAS allows for the obtainment of mean deformation velocity maps and displacement time series relative to the time period from 1992 to 2000, relative to ESR-1/2, and from 2002 to 2010 for ASAR-Envisat. Our results demonstrate how the DInSAR application can obtain reliable information of ground displacement over time in these regions, and may represent a suitable instrument for natural hazards assessment.

    Description: Background: Timely and appropriate hospital treatment of acute cerebrovascular diseases (stroke and Transient Ischemic Attacks - TIA) improves patient outcomes. Emergency Medical Service (EMS) dispatchers who can identify cerebrovascular disease symptoms during telephone requests for emergency service also contribute to these improved outcomes. The Italian Ministry of Health issued guidelines on the management of AC patients in pre-hospital emergency service, including Cincinnati Prehospital Stroke Scale (CPSS) use.We measured the sensitivity and Positive Predictive Value (PPV) of EMS dispatchers' ability to recognize stroke/TIA symptoms and evaluated whether the CPSS improves accuracy. Methods: A cross-sectional multicentre study was conducted to collect data from 38 Italian emergency operative centres on all cases identified with stroke/TIA symptoms at the time of dispatch and all cases with stroke/TIA symptoms identified on the scene by the ambulance personnel from November 2010 to May 2011. Results: The study included 21760 cases: 18231 with stroke/TIA symptoms at dispatch and 9791 with symptoms confirmed on the scene. The PPV of the dispatch stroke/TIA symptoms identification was 34.3% (95% CI 33.7-35.0; 6262/18231) and the sensitivity was 64.0% (95% CI 63.0-64.9; 6262/9791). Centres using CPSS more often (〉10% of cases) had both higher PPV (56%; CI 95% 57--60 vs 18%; CI 95% 17--19) and higher sensitivity (71%; CI 95% 87--89 vs 52%; CI 95% 51--54).In the multivariate regression a centre's CPSS use was associated with PPV (beta 0.48 p = 0.014) and negatively associated with sensitivity (beta -0.36; p = 0.063); centre sensitivity was associated with CPSS (beta 0.32; p = 0.002), adjusting for PPV. Conclusions: Centres that use CPSS more frequently during phone dispatch showed greater agreement with on-the-scene prehospital assessments, both in correctly identifying more cases with stroke/TIA symptoms and in giving fewer false positives for non-stroke/TIA cases. Our study shows an extreme variability in the performance among OCs, highlighting that form many centres there is room for improvement in both sensitivity and positive predictive value of the dispatch. Our results should be used for benchmarking proposals in the effort to identify best practices across the country.

    Description: In the years between 2011 and 2014, at the edge between the Apennines collapsing chain and the subducting Calabrian arc, intense seismic swarms occurred in the Pollino mountain belt. In this key region, 〈2.5mm/yr of NE-trending extension is accommodated on an intricate network of normal faults, having almost the same direction as the mountain belt. The long-lasting seismic release consisted of different swarm episodes, where the strongest event coinciding with a ML 5.0 shock occurred in October 2012. This latter comes after a ML four nucleated in May 2012 and followed by aseismic slip episodes. In this study, we present accurate relocations for ∼6,000 earthquakes and shear-wave splitting analysis for ∼22,600 event-station pairs. The seismicity distribution delineates two main clusters around the major shocks: in the north-western area, where the ML 5.0 occurred, the hypocenters are localized in a ball-shaped volume of seismicity without defining any planar distribution, whilst in the eastern area, where the ML 4.3 nucleates, the hypocenters define several faults of a complex system of thrusts and back-thrusts. This different behavior is also imaged by the anisotropic parameters results: a strong variability of fast directions is observed in the western sector, while stable orientations are visible in the eastern cluster. This tectonic system possibly formed as a positive flower structure but as of today, it accommodates stress on normal faults. The deep structure imaged by refined locations is overall consistent with the complex fault system recently mapped at the surface and with patterns of crustal anisotropy depicting fractures alignment at depth. The possible reactivation of inherited structures supports the important role of the Pollino fault as a composite wrench fault system along which, in the lower Pleistocene, the southward retreat of the ionian slab was accommodated; in this contest, the inversion of the faults kinematics indicates a probable southward shift of the slab edge. This interpretation may help to comprehend the physical mechanisms behind the seismic swarms of the region and defining the seismic hazard of the Pollino range: nowadays a region of high seismic hazard although no strong earthquakes are present in the historical record.

    Description: Since the late the late ’60s-early ’70s era seismologists started developed theories that included variations of the elastic property of the Earth crust and the state of stress and its evolution crust prior to the oc- currence of a large earthquake. Among the others the theory of the dilatancy (Scholz et al., 1973): when a rock is subject to stress, the rock grains are shifted generating micro-cracks, thus the rock itself in- creases its volume. Inside the fractured rock, fluid saturation and pore pressure play an important role in earthquake nucleation, by modulating the effective stress. Thus measuring the variations of wave speed and of anisotropic parameter in time can be highly informative on how the stress leading to a major fault failure builds up. In 80s and 90s such kind of research on earthquake precursor slowed down and the priority was given to seismic hazard and ground motions studies, which are very important since these are the basis for the building codes in many countries. Today we have dense and sophisticated seismic networks to measure wave-fields characteristics: we archive continuous waveform data recorded at three components broad-band seismometers, we almost routinely obtain high resolution ear- thquake locations. Therefore we are ready to start to systematically look at seismic-wave propagation properties to possibly reveal short-term variations in the elastic properties of the Earth crust. In active fault areas and volcanoes, tectonic stress variation influences fracture field orientation and fluid migration processes, whose evolution with time can be monitored through the measurement of the anisotropic pa- rameters ( Piccinini et al., 2006). Through the study of S waves anisotropy it is therefore potentially possible to measure the presence, migration and state of the fluid in the rock traveled by seismic waves, thus providing a valuable route to understanding the seismogenic phenomena and their precursors (Crampin & Gao, 2010). Variations of anisotropic parameter and of the ratio between the compressional (P-wave) and the shear (S-wave) seismic velocities, the Vp/Vs (Nur, 1972) have been recently observed and measured during the preparatory phase of a major earthquake (Lucente et al. 2010). Here we show the anisotropic parameters at station MMN during the Pollino seismic sequence 2010-2013.
