    Notes: Abstract Purpose: We studied vincristine disposition after 169 weekly i.v. bolus injections in 32 children with acute lymphoblastic leukemia, non-Hodgkin lymphoma, or Wilms' tumor. The aim of the study was to determine intrapatient and interpatient variability in vincristine disposition and demographic, clinical, and biochemical characteristics influencing this variability. Methods: Vincristine plasma concentrations were measured by a high-performance liquid chromatography assay with electrochemical detection. A limited sampling strategy was used based on a bayesian parameter estimation algorithm that is part of the ADAPT II software package. A two-compartment, first-order model was fitted to the data, and pharmacokinetic parameters were calculated from the model using the ADAPT II software. For statistical analysis, analysis of variance (ANOVA), t test, simple and multiple regression analysis, and non-parametric or robust equivalents were used. Results: Results showed a large intrapatient and interpatient variability in distribution half-life, elimination half-life, total body clearance, apparent volume of distribution at steady state, and area under the concentration–time curve. Intrapatient variability was significantly smaller than interpatient variability for all these parameters except distribution half-life. The diagnosis or treatment protocol turned out to be the most predictive characteristic; leukemia and non-Hodgkin lymphoma patients had a significantly higher total body clearance than Wilms' tumor patients. Conclusions: We conclude that both intrapatient and interpatient variability in vincristine pharmacokinetics is large in pediatric cancer patients and that variability, although significantly influenced by diagnosis, largely remains unpredictable.

    Description: An eleven-week feeding trial was conducted to evaluate the growth effect of partial replacement of fishmeal by the microalga Spirulina platensis in the diet of juvenile Pacific white shrimp Litopenaeus vannamei (Boone). The results clearly indicate that Spirulina platensis constitutes an effective food ingredient for shrimp. Growth rates of a Spirulina-fed group differed highly significantly (p 〈 0,001) compared to two groups fed to less suitable diets and were slightly, even though not significantly superior to that based on an optimal commercial reference fish diet. As a side effect, Spirulinafed shrimps showed measurable differences in pigmentation.

    Description: Quantum limited amplifiers are sought after for a wide range of applications within quantum technologies and sensing. One promising candidate is the travelling wave parametric amplifier which exploits the non-linear kinetic inductance of a superconducting transmission line. This type of microwave amplifier promises to deliver a high gain, a quantum limited noise performance over several GHz bandwidth, and a high dynamic range. However, practical realizations of this type of device have so far been limited by fabrication defects, since the length of the superconducting transmission line required for achieving substantial parametric gain is on the order of ∼1 m. Here, we report on a design for a microwave traveling wave amplifier based on a slow propagation line comprising a central strip with high kinetic inductance and quasi-fractal line-to-ground capacitors. Due to an enhanced per unit length inductance (73 nH cm −1 ) and capacitance (15 pF cm −1 ), the line has a microwave propagation velocity as low as 9.8 × 10 8  cm s −1 . This translates into parametric gain up to 0.5 dB cm −1 and a total gain of 6 dB for just a ∼10 cm long transmission line. Moreover, the flexibility of the presented design allows balancing the line inductance and capacitance in order to keep the characteristic impedance close to 50 Ω and to suppress standing waves, both factors being essential in order to implement a practical parametric amplifier in the microwave domain.

    Description: Objective Immunoglobulin G4-related disease (IgG4-RD) is a multiorgan immune-mediated disease that predominantly affects the biliary tract (IgG4-associated cholangitis, IAC) and pancreas (autoimmune pancreatitis, AIP). We recently identified highly expanded IgG4+ B-cell receptor clones in blood and affected tissues of patients with IAC/AIP suggestive of specific (auto)antigenic stimuli involved in initiating and/or maintaining the inflammatory response. This study aimed to identify (auto)antigen(s) that are responsible for the clonal expansion of IgG4+ B cells in IgG4-RD. Design We screened sera of patients with IAC/AIP (n=50), in comparison to control sera of patients with primary sclerosing cholangitis (PSC) and pancreatobiliary malignancies (n=47), for reactivity against human H69 cholangiocyte lysates on immunoblot. Subsequently, target antigens were immunoprecipitated and analysed by mass spectrometry. Results Prominent reactivity against a 56 kDa protein was detected in human H69 cholangiocyte lysates exposed to sera of nine patients with IAC/AIP. Affinity purification and mass spectrometry analysis identified annexin A11, a calcium-dependent phospholipid-binding protein. Annexin A11-specific IgG4 and IgG1 antibodies were only detected in serum of patients with IgG4-RD of the biliary tract/pancreas/salivary glands and not in disease mimickers with PSC and pancreatobiliary malignancies. Epitope analysis showed that two annexin A11 epitopes targeted by IgG1 and IgG4 autoantibodies were shared between patients with IAC/AIP and IgG4 antibodies blocked binding of IgG1 antibodies to the shared annexin A11 epitopes. Conclusion Our data suggest that IgG1-mediated pro-inflammatory autoreactivity against annexin A11 in patients with IgG4-RD may be attenuated by formation of annexin A11-specific IgG4 antibodies supporting an anti-inflammatory role of IgG4 in IgG4-RD.

    Description: During yeast cell polarization localization of the small GTPase, cell division control protein 42 homologue (Cdc42) is clustered to ensure the formation of a single bud. Here we show that the disease-associated flippase ATPase class I type 8b member 1 (ATP8B1) enables Cdc42 clustering during enterocyte polarization. Loss of this regulation results in increased apical membrane size with scattered apical recycling endosomes and permits the formation of more than one apical domain, resembling the singularity defect observed in yeast. Mechanistically, we show that to become apically clustered, Cdc42 requires the interaction between its polybasic region and negatively charged membrane lipids provided by ATP8B1. Disturbing this interaction, either by ATP8B1 depletion or by introduction of a Cdc42 mutant defective in lipid binding, increases Cdc42 mobility and results in apical membrane enlargement. Re-establishing Cdc42 clustering, by tethering it to the apical membrane or lowering its diffusion, restores normal apical membrane size in ATP8B1-depleted cells. We therefore conclude that singularity regulation by Cdc42 is conserved between yeast and human and that this regulation is required to maintain healthy tissue architecture.

    Description: Deep hydrothermal Mo, W and base metal mineralization found in the Detroit City portal of the Sweet Home mine in the Alma district (Colorado Mineral Belt) was deposited in response to magmatic activity and the formation of Climax-type Mo deposits during the Oligocene. This study presents extensive geochemical analyses of fluid inclusions inminerals from early greisen-like vein mineralization to better understand the fluid system responsible for ore formation. Quartz and fluorite, which are associated with molybdenite, huebnerite and/or pyrite mineralization, precipitated from low- to medium-salinity (1.5-11.5 wt.% equiv. NaCl), CO2-bearing fluids at temperatures between 360 and 415°C and probably under a fluctuating pressure regime at depths of at least 3.5 km. The formation of greisen-like and base metal mineralization at the Detroit City portal of the Sweet Home mine is related to fluids of different origin. Early magmatic fluids were the principal source for mantle-derived volatiles (CO2, H2S/SO2, noble gases) and mixed with significant amounts of heated meteoric water. Fluid mixing of magmatic fluids with meteoric water is constrained by δ 2H-δ 18O relationships of fluid inclusion water in different minerals (Fig. 1). Whether molybdenum was derived from magmatic fluids remains unclear. Fluid inclusions in huebnerite suggest that W originated from source rocks that are enriched in organic matter rather than from magmatic fluids. The deep hydrothermal mineralization at the Detroit City portal of the Sweet Home mine shows features similar to deep hydrothermal vein mineralization found in Climax-type Mo deposits and their periphery, suggesting that fluid migration and the deposition of ore and gangue minerals in the Sweet Home mine was triggered by a deep-seated magmatic intrusion. The findings of this study are in good agreement with the results of previous fluid inclusion studies of the mineralization of the Sweet Home mine (Lüders et al., 2009) and from Climaxtype Mo porphyry deposits in the Colorado Mineral Belt (Hall et al., 1974).

    Description: Stibnite was mined until the end of the twentieth century in the Schlaining ore district, Austria, near the easternmost border of the Eastern Alps where windows of Penninic ophiolites and metasediments are exposed below Austroalpine tectonic units. In Early Miocene, structurally controlled small vein and metasomatic stibnite-quartz deposits were formed in Penninic Mesozoic calcareous marbles and calcite schists. Fluid inclusion studies identified two fluids involved in the mineralization: (i) a low-salinity, low-CO2 metamorphic fluid that precipitated quartz at approximately 240 °C and (ii) a stibnite-forming ore fluid that had a meteoric origin. There is no evidence of boiling or that the fluids mixed during mineralization. The ore components Sb and H2S were leached by fluid/rock interaction from buried rock units. Stibnite mineralization occurred by cooling the ore fluid to below 300 °C, at less than 2000 m depth. Quartz precipitated at slightly lower temperatures, approximately contemporaneous with stibnite. Fluid migration and ore deposition are probably related to high heat flow during the exhumation of the Rechnitz Window in response to Neogene extension and/or shallow Early Miocene andesitic magmatism. The study emphasizes that data obtained from the analyses of gangue minerals alone cannot routinely be used to infer the origin and depositional conditions of the associated ore minerals.
