    Notes: Final fruit size is the consequence of complex metabolic events that occur between fruit set and maturation. Disruption of these biochemical and molecular processes at any stage during fruit growth will impact on final fruit size. Because fruit size is a function of cell number rather than cell size, factors affecting cell division cycle activity assume importance. In this paper, we focus attention on the metabolic control of fruit growth using avocado as a model system. Three areas of current interest are highlighted, viz. the contribution by isoprenoid metabolism in the control of cell proliferation, the role played by carbohydrate content and composition in signalling changes in metabolite status and gene expression and maintenance of plant hormone homeostasis. Central to the process of fruit growth and control of final fruit size by cell division is 3-hydroxy-3-methylglutaryl coenzyme A reductase (HMGR) and activity of the sucrose non-fermenting 1-related protein kinase (SnRK1) complex. It is argued that sugar content and composition of sink cells impact on SnRK1 (and hexokinase) to modulate expression of sugar-metabolizing enzymes, HMGR and molybdenum cofactor (MoCo)-containing enzymes. These changes, in turn, impact on hormone metabolism by affecting allocation of the purine-derived MoCo to aldehyde oxidase and thus the endogenous concentration of indole-3-acetic acid, abscisic acid and cytokinin (CK) to alter plant hormone homeostasis. These aspects are integrated into a model to explain the metabolic control of avocado fruit growth and final fruit size.

    Notes: Samples of fed and unfed hatchery reared Atlantic salmon fry and parr were analysed for body water and whole body Na, K and Ca content. From these body parameters it is possible to estimate the relative proportions of the skeletal mass, and intra- and extracellular spaces. These data can be applied to estimating the nutritional status of parr sampled at random from hatchery, and possibly from wild populations. When fish with a dry weight of about 25 and 70 mg were deprived of artificial food for 27 and 21 days, respectively, the Na/K ratio rose to 0.59 and 0.80 compared with 0.37 in the controls. With fish of about 475 mg in weight, percentage water content was a better indicator of undernourishment. The use of the K+: Ca2+ ratio to compare the amount of cellular material with the skeletal mass was a good indicator of nutritional depletion, especially in smaller fish.

    Notes: Water content of Atlantic salmon parr fell from about 84% at emergence (late May) to just under 79% in September but rose again towards March. Na+ content consequently rose from 3·3 mg g−1 dry wt at the beginning of June to 6·2 mg g−1 in early July. It then fell to 4·4 mg g−1 in September, rising again towards March. K+ content rose to a maximum in July to stabilize at 16·6 mg g−1 dry wt in September. The resultant Na+/K+ ratio peaked at 0·43: 1 in mid-June, falling to a minimum in mid-August but rising again in March reflecting changes in the relative proportions of intra and extracellular water. The changes in whole-body chemistry suggest a period of nutritional stress immediately after emergence and during the winter. In streams at higher altitude and of lower nutrient status, nutritional stress during the winter appears to be more severe.

    Notes: Abscisic acid is considered an apocarotenoid formed by cleavage of a C-40 precursor and subsequent oxidation of xanthoxin and abscisic aldehyde. Confirmation of this reaction sequence is still awaited, and might best be achieved using a cell-free system capable of both carotenoid and abscisic acid biosynthesis. An abscisic acid biosynthesizing cell-free system, prepared from flavedo of mature orange fruits, was used to demonstrate conversion of farnesyl pyrophosphate, geranylgeranyl pyrophosphate and all-trans-β-carotene into a range of β,β-xanthophylls, xanthoxin, xanthoxin acid, 1′,4′-trans-abscisic acid diol and abscisic acid. Identification of product carotenoids was achieved by high-performance liquid chromatography and on-line spectral analysis of individual components together with co-chromatography. Putative C-15 intermediates and product abscisic acid were identified by combined capillary gas chroma-tography-mass spectrometry. Kinetic studies revealed that β-carotene, formed from either famesyl pyrophosphate or geranylgeranyl pyrophosphate, reached a maximum within 30 min of initiation of the reaction. Thereafter, β-carotene levels declined exponentially. Catabolism of substrate β-carotene into xanthophylls, putative abscisic acid precursors and product abscisic acid was restricted to the all-trans-isomer. However, when a combination of all-trans- and 9-cis-β-carotene in the ratio 1:1 was used as substrate, formation of abscisic acid and related metabolites was enhanced. Biosyn-thetically prepared [14C]-all-trans-violaxanthin, [14C]-all-trans-neoxanthin and [14C]-9′-cis-neoxanthin were used as substrates to confirm the metabolic interrelationship between carotenoids and abscisic acid. The results are consistent with 9′-cis-neoxan-thin being the immediate carotenoid precursor to ABA, which is oxidatively cleaved to produce xanthoxin. Formation of abscisic aldehyde was not observed. Rather, xanthoxin appeared to be converted to abscisic acid via xanthoxin acid and 1′,4′-trans-abscisic acid diol. An alternative pathway for abscisic acid biosynthesis is therefore proposed.
