    Notes: The effect of various experimental parameters upon the frequency of callus formation from cultured anthers of Oryza sativa has been investigated. Although certain medium components were found to be critical to callus formation, the concentration of these components had little effect upon the frequency of callus formation. The degree to which the callus formation frequency was influenced by cold pretreatment of the flowers was variable. Even though plants were grown under uniform conditions and flowers containing pollen in the microspore stage of development were selected for dissection, the frequency of callus formation varied nonrandomly between flowers. In experiments with populations of flowers in a physiological and developmental state favorable to callus formation 35% of plated anthers produced callus and at least 60% of these calluses gave rise to green plants.

    Notes: Summary Cultures of Nicotiana tabacum cells homozgous for a mutation (S4) at the SuRB locus that confers resistance to the sulfonylurea herbicides chlorsulfuron and sulfometuron methyl (Chaleff and Ray 1984; Chaleff and Bascomb 1987) were used to isolate a doubly mutant cell line (S4 Hra/S4+) resistant to even higher herbicide concentrations. Growth of cells homozygous for both the S4 and Hra mutations (S4 Hra/S4 Hra) was uninhibited by a herbicide concentration 500-fold higher than a concentration by which growth of S4+/S4+ callus was inhibited by 75%. Plants homozygous for both mutations were at least five-fold more resistant to foliar applications of chlorsulfuron than were singly mutant S4+/S4+ plants. This enhanced resistance was inherited as a single, semidominant, nuclear trait that is genetically linked to the S4 mutation. Acetolactate synthase (ALS) activity in extracts of leaves of doubly mutant (S4 Hra/S4 Hra) plants was approximately 20-fold more resistant to inhibition by chlorsulfuron and sulfometuron methyl than was ALS activity in singly mutant (S4+/ S4+) leaf extracts, which was in turn more resistant to inhibition by these compounds than was the normal enzyme. Extracts prepared from plants of these three genotypes possessed the same ALS specific activities. Therefore, Hra represents a second independent mutation at or near the SuRB locus that reduces the sensitivity of tobacco ALS activity to inhibition by sulfonylurea herbicides.

    Notes: Abstract Rice callus was initiated from cultured anthers and was maintained on medium containing ammonium as sole nitrogen source and supplemented with either a buffer or an organic acid. Plant regeneration also was obtained on this medium, but the morphogenetic capacity of the callus culture declined as rapidly as on a medium containing a mixture of ammonium and nitrate. The results of the studies suggest that organic acids provide an additional function beyond that of buffering the medium during growth on ammonium.

    Notes: Summary Mutations conferring resistance to sulfonylurea herbicides have been identified at two unlinked genetic loci (SuRA and SuRB) in Nicotiana tabacum. A mutation (S4) residing at the SuRB locus causes production of an altered form of acetolactate synthase (ALS) that is insensitive to inhibition by sulfonylurea herbicides and that constitutes approximately 50% of total ALS activity. Another mutation (C3) at the SuRA locus similarly results in herbicide resistance of approximately half of the total ALS activity. These results suggest that N. tabacum possesses at least two isozymic forms of ALS. The relative and absolute amounts of the isozymes are affected by a mutation that segregates independently of C3.

    Notes: Summary Tobacco cell lines selected for resistance to picloram (4-amino-3,5,6-trichloropicolinic acid) and plants regenerated from these cell lines manifest several traits not shown by the parental strains. Genetics analyses of the regenerated plants have permitted the sources of this variability to be identified. Tricotyledenous seedlings appeared at a much higher frequency among the progeny of a heterozygous mutant plant (PmR1/+) regenerated from culture than they did among progeny of normal regenerated plants. In crosses with the regenerated heterozygous mutant plant and with homozygous progeny of this plant (PmR1/PmR1) the frequency of tricotyly was influenced more by the generation than by the genotype of the parent plant. Therefore, it is concluded that tricotyly is a physiological response to passage through cell culture. More than half of the picloram-resistant cell lines isolated were also resistant to hydroxyurea. Segregation of these two resistances was analyzed in progeny of crosses with regenerated plants. In all cases hydroxyurea-resistance was genetically stable and inherited as a single dominant nuclear mutation (designated HuR). In crosses with plants PmR1/+ and PmR7/+ the HuR and PmR mutations assorted independently. In contrast, the HuR mutation recovered from plant PmR6/+ was linked to the PmR6 mutation.
