    Notes: Abstract Two different series collected at the University Hospital of Freiburg are presented as a basis for the critical appraisal of available imaging tests and their correlation with anatomic pain sites. The first series comprises 284 patients of non-traumatic acute addomen admitted to the emergency department during 1990. The radiological approach revealed an increased use of immediate ultrasound (58%) followed by plain film radiography (53%), contrast studies (15%), and computed tomography (9.8%). Five groups of entities were identific according to the diagnostic imaging procedure prior to surgery. In a second series, the clinical presentation, the distribution and the efficacy of imaging studies for the confirmation of acute mesenteric ischemia were evaluated in 55 patients over a period of 14 years. A reduced mortality rate emphasizes the value of early and aggressive diagnosis and angiographie management.

    Notes: Abstract To assess the value of three-dimensional (3D) image reconstructions of two-dimensional (2D) data from contiguous non-overlapping slices in the diagnosis of spinal trauma, 21 patients with a total of 36 injured vertebrae and 4 normal controls were examined. Forty plain films in two planes, 40 axial CT scans with multiplanar reformatted (MPR) 2D reconstructions and 40 sets of 3D images (surface rendering technique) were independently evaluated by four readers. The final diagnosis was defined after a panel review of clinical, surgical and imaging findings on all modalities. The 3D image reconstructions were not as accurate as axial CT with MPR 2D reconstructions in the diagnosis of vertebral body (n = 20) and posterior element fractures (n = 35). Dislocation (n = 3) was equally well detected by all imaging modalities. Narrowing of the spinal canal (n = 17) was best assessed by either MPR 2D CT or 3D images. A rotational component was diagnosed more accurately by 3D images, followed by 2D CT and plain films. Thus, 3D images combined with MPR 2D CT reconstructions are an important adjunct for an accurate diagnosis of spinal trauma, especially when a rotational component is suspected.

    Description / Table of Contents: Zusammenfassung Es wird über die Analyse von Citratcyclusintermediaten mit Hilfe der modernen Flüssigkeits-Chromatographie (LC) berichtet. Der photometrische Nachweis der kurzkettigen Carbonsäuren im Säuleneluat erfolgt mit Hilfe eines gruppenspezifischen Indicators (o-Nitro-Phenol). Bei gradientenfreier Elution beträgt der Rauschpegel der Basislinie des Chromatogramms max. 1 · 10−3 E und der Flächenwert für 10 nVal Essigsäure 25 E · sec. Bei wiederholten Probenaufgaben betragen die Abweichungen vom Flächenmittelwert max. ± 5% (VB 95%). Die Aufbereitung biologischer Proben erfolgt durch Extraktion mit Diäthyläther in einer einfachen säulen-chromatographischen Anlage, die gleichzeitig der Vortrennung sog. „kritischer Säurenpaare” dient. Die Anwendbarkeit der LC auf die Analyse von biologischem Material wird mit einem Chromatogramm von 10 μl Humanserum gezeigt.
    Notes: Abstract The analysis of TCA intermediates and related compounds with modern liquid chromatography (LC) is described. Short-chain carboxylic acids are detected by adding a compound specific indicator (o-nitro-phenol) to the eluent. Without gradient elution, the noise level of the chromatogram baseline does not exceed 1×10−3 O.D. and the peak area for 10 nanoequivalent acetic acid is 25 absorbance seconds. With repeated applications of sample to column, deviations of 5% of the medium peak area value are not exceeded (range of confidence 95%). Preparation of the biological samples is achieved by ether extraction with a simple column-chromatography apparatus, which at the same time, preseparates critical acids otherwise not completely separable by elution without gradient. The application of LC to the analysis of biological material is demonstrated by a chromatogram of 10 μl of human serum.

    Description: Many insects raise their offspring on organic substrates or in the soil where microorganisms are abundant. Microbes may pose a serious threat to offspring development and survival by either decomposing food resources or directly infecting the offspring. Selection to cope with these effects may favor social defenses, for example, through forms of parental care that can limit or eliminate these threats to offspring fitness. In this study, we experimentally tested if maternal egg attendance in the European earwig Forficula auricularia has a function as a social defense against mold infection of eggs by manipulating exposure of eggs to mold spores and the presence of the mother in a fully factorial design. Furthermore, we investigated the potential roles of egg grooming behavior and maternal transfer of chemicals as underlying mechanisms. As predicted, the beneficial effect of egg attendance on hatching success was significantly enhanced when eggs were exposed to the mold. Females significantly increased their egg grooming duration in response to mold exposure of her eggs, and the quantity of chemicals (identified as hydrocarbons) was maintained among attended eggs but decreased substantially among unattended eggs. Maternal transfer of chemicals was confirmed in extractions of glass beads that were mingled into attended or unattended clutches. This study shows that maternal egg attendance in the European earwig has a social defense function protecting offspring against mold infection. The maternal egg grooming behavior seems to be key for this effect, probably through both the mechanical removal of spores and the continued application of chemical substances on the egg surface.

    Description: Tuberculosis remains a global health problem so that a more effective vaccine than bacillus Calmette–Guérin is urgently needed. Cytomegaloviruses persist lifelong in vivo and induce powerful immune and increasing ("inflationary") responses, making them attractive vaccine vectors. We have used an m1–m16-deleted recombinant murine CMV (MCMV) expressing Mycobacterium tuberculosis Ag 85A to show that infection of mice with this recombinant significantly reduces the mycobacterial load after challenge with M. tuberculosis , whereas control empty virus has a lesser effect. Both viruses induce immune responses to H-2 d –restricted epitopes of MCMV pp89 and M18 Ags characteristic of infection with other MCMVs. A low frequency of 85A-specific memory cells could be revealed by in vivo or in vitro boosting or after challenge with M. tuberculosis. Kinetic analysis of M. tuberculosis growth in the lungs of CMV-infected mice shows early inhibition of M. tuberculosis growth abolished by treatment with NK-depleting anti–asialo ganglio- N -tetraosylceramide Ab. Microarray analysis of the lungs of naive and CMV-infected mice shows increased IL-21 mRNA in infected mice, whereas in vitro NK assays indicate increased levels of NK activity. These data indicate that activation of NK cells by MCMV provides early nonspecific protection against M. tuberculosis, potentiated by a weak 85A-specific T cell response, and they reinforce the view that the innate immune system plays an important role in both natural and vaccine-induced protection against M. tuberculosis .
