    Notes: Background Allergen-induced T-helper type 2 (Th2) responses can be inhibited with Th1 directing vaccines. However, studies comparing the efficacy of the different adjuvants have not been performed in detail.Objective For this reason we compare the effects of live Bacillus–Calmette–Guerin(BCG), heat-killed (hk)-BCG, CpG-ODN (oligodeoxynucleotide) or PPD on the development of allergen-induced Th2 responses in mice.Methods Ovalbumin (OVA)-specific allergic responses were induced in C57BL/6 mice by two intraperitoneally (i.p.) applications of OVA/alum followed by the intranasal challenge with OVA. The different Th1-inducing adjuvants were applied to the mice together with OVA/alum i.p. during the OVA-sensitization period and, subsequently, different parameters of allergic immune responses were evaluated.Results All the adjuvants were effective in inhibiting the development of allergen-induced airway eosinophilia, mucous production and, with the exception of PPD, also airway hyper-reactivity, when they were applied together with OVA/alum. However, allergen-specific IgG1 and IgE serum levels were only reduced in live BCG- and PPD-treated mice. Suppression of airway eosinophilia was not observed in IFN-γ- or IL-12-deficient mice (hk-BCG, CpG-ODN and PPD). Interestingly, live BCG was still able to suppress allergen-induced Th2 responses in the absence of either IFN-γ or IL-12. When mice vaccinated with the different adjuvants together with OVA/alum were subjected to a second period of OVA/alum immunization, only live and hk-BCG were able to efficiently suppress the development of airway inflammation. This effect could be adoptively transferred by splenic CD4+ T cells.Conclusions Taken together our data suggest that live BCG〉hk-BCG〉CpG-ODN 〉PPD are effective in suppressing allergen-induced Th2 responses. The degree of suppression and the component of the Th2 response affected (airway inflammation vs. the production of allergen-specific IgE and IgG1) were dependent upon the adjuvant used and how it was applied. Our results contribute to the design of novel vaccines protecting humans from developing allergic disorders.

    Notes: Abstract High resolution benthic oxygen isotope records combined with radiocarbon datings, from cores retrieved in the North, Equatorial, and South Atlantic are used to establish a reliable cronostratigraphy for the last 60 ky. This common temporal framework enables us to study the timing of the sub-Milankovitch climate variability in the entire surface Atlantic during this period, as reflected in planktonic oxygen isotope records. Variations in sea surface temperatures in the Equatorial and South Atlantic reveal two warm periods during the mid-stage 3 which are correlated to the warming observed in the North Atlantic after Heinrich events (HL) 5 and 4. However, the records show that the warming started about 1500 y earlier in the South Atlantic. A zonally averaged ocean circulation model simulates a similar north-south thermal antiphasing between the latitudes of our coring sites, when pertubated by a freshwater flux anomaly. We infer that the observed phase relationship between the northern and the southern Atlantic is related to periods of reduced NADW production in the North Atlantic, such as during HL5 and HL4.

    Notes: Abstract  The Silurian of Gotland, Sweden, consists of 440 m of carbonate deposits. Repeatedly, uniform sequences of micritic limestones and marls are interrupted by complex-structured reefs and by adjacent platform sediments. Generally, the alteration of facies is interpreted as the result of sea-level fluctuations caused by a gradual regression with superimposed minor transgressive pulses. The purpose of this study is a facies interpretation based on both field observations and stable isotope measurements of brachiopod shells. Approximately 700 samples from stratigraphically arranged localities in different facies areas have been investigated. The carbon and oxygen isotopes show principally parallel curves and a close relationship to the stratigraphic sequence. Lower values occur in periods dominated by deposition of marly sequences. Higher values are observed in periods dominated by reefs and extended carbonate platforms. The oxygen isotope ratios are interpreted to reflect paleosalinity changes due to varying freshwater input, rather than to paleotemperature. Carbon isotope ratios are believed to have been connected to global changes in the burial of organic carbon in black shales during periods of euxinic deep water conditions. Consequently, the facies succession on Gotland results from global paleoclimatic conditions. Changes in terrigenous input due to different rates of weathering and freshwater runoff, rather than sea-level fluctuations, control the carbonate formation of the Silurian on Gotland.

    Description: An undisturbed 16 m late Pliocene-Pleistocene sediment core spanning 2.6 Myr of deposition was recovered from the Manihiki Plateau by the German research vessel Sonne in 1990. This core 34KL complements the heavily disturbed late Pliocene-Pleistocene core sections of DSDP Site 317, and is well suited for stratigraphic correlation. The sediments consist of calcareous microfossils (93–97% CaCO3), minor portions of siliceous microfossils and detrital minerals. All important calcareous microfossil zones could be identified and correlated with the magnetostratigraphic and isotope stratigraphic events, stages and periods. Due to the high degree of silica dissolution, the late Quaternary radiolarian Buccinosphaera invaginata Zone is the only siliceous fossil zone which could be identified. An interval with Globigerinoides gomitulus/pink was found within the middle Pleistocene which is clearly distinguished from the Globigerinoides ruber/pink interval of the late Pleistocene. The magnetostratigraphic Gauss-Matuyama and Matuyama-Brunhes boundaries as well as the Olduvai and Jaramillo events were clearly identified. The δ180 curve displays the 100 kyr (Milankovitch) and the 41 kyr (Laplace) cyclicity periods. The beginning of the Laplace Period and the last occurrence of the calcareous nannofossil Discoaster brouweri, which marks the end of the Pliocene, fall within the base of the Olduvai Event. There is also a marked drop in sedimentation rates around this time, which seems to be a regional phenomenon. Prominent 3.5 kHz subbottom reflectors at 8 ms and 15 ms reflection time are related to lithologic changes near the Mid-Pleistocene Revolution (MPR) and the beginning of the Laplace Period or the Pleistocene, respectively. It is possible to trace these reflectors laterally to the eroded eastern edge of the Manihiki Plateau where they can be tied to older strata.

    Description: We report on epibenthic foraminiferal δ18O and δ13C and percentage coarse fraction records from Caribbean Ocean Drilling Program (ODP) Site 999 (12°44′N, 78°44′W, water depth 2828 m) spanning the interval from 8.5 to 5.3 Ma. Low epibenthic δ13C values and low amounts of sand-sized particles (mostly foraminifer shells) indicate a poorly ventilated deep Caribbean throughout the late Miocene. At this time the deep Caribbean was dominated by a nutrient-rich and corrosive water mass. A generally constant δ13C gradient between the Caribbean and deep Atlantic records during the late Miocene suggests that the fluctuations in δ13C reflect global changes in δ13C of the dissolved inorganic carbon due to varying erosion of organic carbon from terrigenous soils and shelf sediments. The observed 100-kyr cyclicity of epibenthic δ13C is in good accordance with the variability of terrigenous input to the equatorial Atlantic as recorded by magnetic susceptibility records of the Ceara Rise. However, some short-term gradient changes between 7.0 and 4.5 Ma indicate a poorer ventilation of the deep Atlantic related to a reduced production of deep water in the Atlantic. The Messinian Salinity Crisis between 6.0 and 5.3 Ma did not affect the intermediate to deep water gradient between the Caribbean and the Atlantic. Comparison to the Bahama platform record of ODP Site 1006, however, indicates a poorer ventilation of the shallower northern Caribbean basins coincident with the isolation of the Mediterranean Sea.

    Description: On a transect between 20° and 70°S in the eastern Atlantic Ocean and Weddell Sea, water samples from 19 hydrographic stations and bottom water from 55 surface sediment samples taken with a multiple corer were investigated for the stable carbon isotopic composition of the total dissolved inorganic carbon (δ13CΣCO2). These measurements were compared to δ13C values determined on live specimens of the benthic foraminifer Fontbotia wuellerstorfi and closely related genera from the same stations. In addition, at 16 stations the stable carbon isotope composition of sedimentary organic carbon was measured. General deepwater and bottom-water mass circulation patterns as inferred from the δ13CΣCO2 are in close agreement with those known from other nonconservative tracers. Very low δ13C values of upper Circumpolar Deep Water (〈0.3‰ Pee Dee belemnite (PDB)) in the Polar Front region and the eastern limb of the Weddell gyre coincide with nutrient maxima. However, a significant decoupling of the dissolved phosphate signal from the δ13CΣCO2 signal is indicated in the abyssal Weddell Sea. We attribute this to temperature-dependent fractionation processes during gas exchange of surface waters with the atmosphere at sites of bottom-water formation. Multiple corer water from the sediment/water interface is slightly δ13C depleted relative to deepwater and bottom-water δ13ΣCO2. The surface sediment organic carbon δ13C is 3 to 4‰ lower south of the Polar Front than north of it, and the δ13Corg in freshly accumulated phytodetritus is 3 to 4‰ lower than surface sediment organic carbon δ13C. Comparison of live F. wuellerstorfi δ13C and related genera with bottom-water δ13CΣCO2 exhibits at most stations between the Subtropical Front (≈41°S) and the southern boundary of the Antarctic Circumpolar Current (≈55°S) a significant lowering of foraminiferal δ13C values. Compilation of a mean last glacial/interglacial δ13C amplitude (Δδ13C) from six published southern ocean cores results in a shift of −0.99± 0.13‰ PDB; this shift is greater than that in all other regions. However, all of these cores are from positions close to Recent oceanic fronts. Thus, for these peripheral areas of the southern ocean, we suggest about half of the glacial/interglacial shift can be explained by varying frontal zone positions and widths accompanied by a change in mode and height of export production.

    Description: Objective— Previously, we demonstrated the relevance for endothelial carcinoembryonic antigen–related cell adhesion molecule 1 (CEACAM1) expression in collateral formation. However, a proarteriogenic role for CEACAM1 + myeloid cells is unknown. Here, we investigated the contribution of CEACAM1 + myeloid cells on collateral formation. Methods and Results— Collateral growth and vascular remodeling were analyzed in CEACAM1-competent and CEACAM1 null mice after femoral artery ligation in hindlimb ischemia. Reperfusion of the adductor muscles was evaluated by Laser Doppler measurements and microcomputed tomography imaging. In CEACAM1 null mice, poor reperfusion and reduced collateral formation were observed, accompanied by reduction in arterial diameters. Using flow cytometry, we identified an increase of the muscle-resident CD11b + /granulocyte receptor-1+ (Gr-1+) population in CEACAM1 null mice only, pointing toward a CEACAM1-dependent functional deviation. Direct and reciprocal bone marrow transplantations between CEACAM1-competent and CEACAM1 null mice, and antibody-mediated depletion of the CD11b + /Gr-1 + population, confirmed the requirement of CEACAM1 expression on the CD11b + /Gr-1 + population for reestablishment of perfusion after arterial occlusion. Conclusion— CEACAM1 expression on CD11b + /Gr-1 + myeloid cells is a prerequisite for adequate collateral formation.

    Description: Incorporation of boron into foraminiferal shells is thought to be primarily governed by the carbonate chemistry of the ambient seawater, suggesting that it can be reconstructed from B/Ca ratios. To this end, B/Ca ratios of the benthic foraminifer Planulina wuellerstorfi from South Atlantic core top samples have been analyzed using laser ablation-inductively coupled plasma-mass spectroscopy (LA-ICP-MS) to provide additional information on intratest trace element heterogeneity. Results show that boron is heterogeneously distributed within and between shells, with content variations of approximately {+/-}43% displayed within a single shell. B/Ca is higher in the youngest chambers, opposite to the observed between-chamber variability of Mg/Ca. This may be explained by ontogenetic changes of physiological processes that increase the pH of the calcifying fluid and thus the borate concentration while decreasing Mg/Ca to promote calcification. Despite this heterogeneity, mean B/Ca ratios are positively correlated with the deepwater calcite saturation state ({Delta}[CO32-]), in line with previous studies. We apply this empirical relationship to reconstruct {Delta}[CO32-] for the late Pleistocene to Holocene using samples from a depth transect in the equatorial Atlantic. Reconstructed {Delta}[CO32-] values confirm previous studies suggesting that CaCO3-oversaturated North Atlantic Deep Water was reduced during glacial periods, whereas CaCO3-undersaturated Antarctic Bottom Water expanded vertically and propagated northwards. In summary, our data demonstrate that bulk B/Ca in P. wuellerstorfi reliably reflects variations in {Delta}[CO32-], despite the strong physiological control of boron incorporation.
