    Notizen: SYNOPSIS. An empirical method is used to determine mixtures of the chelating agent, EDTA (ethylenediamine tetraacetate), and trace metals which allow maximum growth rates of Chlamydomonas mundana under optimum environmental conditions. In this method which resembles an immunological titration one trace metal (Ca, Fe, Mn or Zn), in increasing concentrations, is used to “saturate” a standard amount of EDTA. Other required trace metals are added in a range of minimum concentrations. Extent of growth with each mixture is evaluated after 24 hours of incubation. Mixtures allowing greatest growth are then used to evaluate growth rate. Media with manganese as the EDTA-saturating trace metal allow most rapid growth rates equivalent to 11.0 doublings per day. Magnesium as the EDTA-saturating metal does not allow growth, suggesting that Mg does not displace other required trace metals from EDTA-complex. Since the mixtures allowing maximum growth rate are “underchelated” it appears that the free metals rather than the metal-chelate complexes are utilized by the alga in rapid growth.

    Notizen: Abstract Nitrate reductase (NR) activity was assayed by measuring the NADH-dependent formation of nitrite in phytoplankton extracts. NR specific activity increased with the nitrate concentration of the water in upwelling areas of the Peru Current. The temperature optimum for NR for natural phytoplankton was 15° to 20°C. NR activity showed diel periodicity, with maximum activity about noon and minimum activity near midnight local time. Rate of nitrate reduction in the extracts averaged only about 15% of the rate of nitrate assimilation in the whole cells.

    Notizen: Abstract Recent improvements in the analytical determination of nitrate and nitrite allow measurements of nanomolar concentrations in nitrogen-depleted surface waters, revealing variability not previously observable. The new methods allow direct observation of net nitrate consumption and production measured as changes in nitrate concentration over time in incubated samples of seawater. We find that nitrate concentrations in nitrogendepleted surface waters show regional differences, and sometimes diel differences. The variability in nitrate concentration reflects differences in atmospheric inputs, in physical forcing, as well as in the biological processes of nitrate uptake and nitrification. Examples are shown from the Southern California Bight (1986–1987), the equatorial Pacific (February–March, 1988), and the eastern subtropical Atlantic (March–April, 1989).

    Notizen: Abstract Assimilation rates of 15N-labelled ammonium, urea, and nitrate by plankton in the upper euphotic zone were measured in 2 summer, 2 winter, and 1 spring cruise in the central North Pacific Ocean. Average rates of ammonium plus urea assimilation could not be determined precisely, but were estimated to be 7 to 25 μg-at. N m-3 day-1. Indirect evidence suggested that non-photosynthetic microorganisms contributed to these rates. Nitrate assimilation was negligible in the upper waters considered in this report (above the chlorophyll maximum and the nutricline). Potential, nitrate-saturated rates were in the range 1 to 8 μg-at. N m-3 day-1. Seasonal comparison showed lowest rates of both carbon and nitrogen assimilation rates per chlorophyll a in winter.

    Notizen: Abstract Chlorophyll images suitable for the regional analysis of the Southern California Bight were obtained by satellite (Nimbus-7 Coastal Zone Color Scanner) on 27 February, 6 March and 11 March 1979. An algorithm to convert satellite chlorophyll data to primary production has been developed. In addition, primary production was measured along with other biological and physical factors from ships cruising in the Southern California Bight during late February and early March 1979. There was fair agreement between shipboard measurements and satellite estimations; for example, primary production (mg carbon m-2 d-1) averaged 554 mg from shipboard measurement and 403 mg from satellite estimation when averaged for the entire Bight for the course of the study. The high standard error associated with the variance in productivity index used in our algorithm prevented more accurate conversion of chlorophyll concentrations into primary productivity. However, the synoptic data provided by concurrent ship and satellite measurements can be used to quantify, and perhaps reduce, sampling errors associated with ship sampling alone and to provide estimates of the accuracy of primary productivity on a regional basis.

    Notizen: Abstract A study of phosphate assimilation by coastal marine plankton revealed that both phytoplankton and microheterotrophs incorporated radioactive phosphorus (33P). Size fractionation of the particulate matter (using 1 μm pore diameter Nucleopore® membrane filters), antibiotic treatment (using garamycin), and independent estimaties of photoautotrophic (14CO2 uptake) and heterotrophic (3H-glucose uptake) activities were employed to separate phyto- and bacterioplankton phosphate uptake. Results indicated that phytoplankton 33P-uptake was best estimated by the fraction of particulate matter retained on the 1 μm membrane filters. Usually, less than 10% of the phytoplankton (based on chlorophyll a measurements) passed the 1 μm pore-diameter filters, whereas about 90% of the heterotrophic activity passed. At least 50% of the 33P-uptake was associated with the 〈1 μm fraction. It may be possible to resolve the phytoplankton and bacterial contributions to 33P-uptake by comparing the percent of total 33P-uptake with the percent of total 3H-glucose uptake associated with the 〉1 μm fraction.

    Notizen: Abstract Ceratium tripos dominated a multi-species dinoflagellate patch in the sub-surface chlorophyll maximum in August 1978 on the Southern California shelf. The specific growth rate (μ) ofC. tripos averaged 0.25 d-1. Patch length was about 45 km along the shelf. Several members of the subsurface dinoflagellate assemblage were also present in surface samples, but only during the daytime. These apparent vertical migrators includedProrocentrum micans, C. furca, Gonyaulax polyedra and other less common forms. The growth ofC. tripos in the California patch is compared with that in aC. tripos patch off New York in 1976.

    Beschreibung: Nitrate concentration was measured in seawater samples from the euphotic zone at the beginning and end of 12-h, daytime, in situ incubations. The changes in concentration are considered to be measurements of new production. During periods of 2-3 weeks in March-April 1989, important time scales for NO3- input to the euphotic zone (i.e. residence times) and new production were approximately 26 d at 18-degrees-N, 31-degrees-W and approximately 10 d near 33-degrees-N, 21-degrees-W. The average rate of NO3- use in the two areas was 2.63 and 0.62 mmol N m-2 (12 h)-1, or, in carbon equivalents 209 and 49 mg C m-2 d-1, respectively. These values bracket the large-scale estimate by Jenkins of new production in the nearby beta triangle of 150 mg C m-2 d-1.

    Beschreibung: During a R.V. Meteor JGOFS-NABE cruise to a tropical site in the northeast Atlantic in spring 1989, three different vertical regimes with respect to nitrate distribution and availability within the euphotic zone were observed. Besides dramatic variations in the depth of the nitracline, a previously undescribed nose-like nitrate maximum within the euphotic zone was the most prominent feature during this study. Both the vertical structure of phytoplankton biomass and the degree of absolute and relative new production were related to the depth of the nitracline, which in turn was dependent on the occurrence/non-occurrence of the subsurface subtropical salinity maximum (S(max)). The mesoscale variability of the nitracline depth, as indicated from a pre-survey grid, and published data on the frequent occurrence of the S(max) in tropical waters suggest higher variability of new production and F-ratio than usually expected for oligotrophic oceans. The importance of salt fingering and double diffusion for nitrate transport into the euphotic zone is discussed.
