Abstract:
What's new? Tumor‐associated lymphocytes contribute to tumor angiogenesis and tumor progression. Here, the authors show that co‐culture of lymphocytes with head and neck squamous cell carcinoma (HNSCC) cell line Cal27 significantly up‐regulated the secretion of pro‐inflammatory cytokine LT‐α in lymphocytes. LT‐α promoted endothelial cell glycolysis, proliferation, migration and tube formation in a PFKFB3‐dependent manner. High expression of LT‐α in the HNSCC tissue array correlated with enhanced angiogenesis and endothelial PFKFB3 expression. Genetic and pharmacological inhibition of PFKFB3 attenuated tumor angiogenesis in HNSCC xenograft mice models, suggesting PFKFB3 as a promising therapeutic target for suppressing angiogenesis in HNSCC.
