Summary By using a blotting and incubation procedure with radioactive DNA probes, several protein species capable of high-affinity DNA binding have been detected among the proteins associated with mustard (Sinapis alba) chloroplast transcription complexes. Species with different affinity for single- and double-stranded DNA have been distinguished by using probes labelled by two different procedures in combination with unlabelled competitor DNA. Proteins of Mr 145,000, 125,000, 115,000, and 33,000 bind strongly to single-stranded DNA, while those of Mr 81,000, 57,000, 19,000, and 17,000 appear to bind both single- and double-stranded DNA.
