    Notes: Nine substituted benzoyl hydrazonyl, three semicarbazonyl, and four thiosemicarbazonyl tridentate ligands were synthesized. They were used to coordinate with Bu2SnCl2 or (PhCH2)2SnCl2 to form 18 novel tin complexes that contained pentacoordinate organotin(IV) in a heterobicyclic ring. All these complexes were characterized by MS, NMR, and IR spectroscopy elemental analyses. © 1995 John Wiley & Sons, Inc.

    Notes: Changes in chromatin structure during spermatogenesis were investigated using a monoclonal antibody that immunoreacts with the N-terminus of the testis-specific histone TH2B. This monoclonal antibody, which had been raised against rat tyrosine hydroxylase (TH), cross-reacted with TH2B because of sequence homology at the N-termini of TH and TH2B. The epitope was localized to the N-terminus of TH2B as trypsin-digested chromatin which lacked the N-terminal tail did not react with anti-TH and preincubating anti-TH with a synthetic peptide made from the homologous sequence between TH2B and TH inhibited its binding to TH and TH2B. In histological sections of rat testis, the primary spermatocytes and round spermatids immunoreacted weakly, whereas elongating spermatids at steps 10-12 immunoreacted intensely with anti-TH. Increased staining of elongating spermatids was also observed in mouse and hamster by immunohistochemistry. However, immunoblotting proteins extracted from separated rat testis cells showed no increase in the TH2B content of these late steps of spermatids. The apparent increase in the immunohistochemical staining corresponds to increased accessibility of the epitope in the elongating spermatids. This indicated that the N-terminus of TH2B is less tightly bound to DNA or to other proteins at this time in preparation for the removal of TH2B and other histones. © 1995 wiley-Liss, Inc.

    Description: Nek2A (NIMA-related kinases 2A) has been known as an important centrosome regulatory factor. The aim of this study was to investigate the expression of Nek2A and the role it played in different stages of breast cancer. We detected the expression of Nek2A in both mRNA and protein levels in MCF10 cell lines including MCF-10A, MCF-10DCIS.com, MCF-10CA1a and in human breast samples which contained normal breast tissue (NBT), breast ductal carcinoma in situ (DCIS), and invasive ductal carcinoma (IDC). Our study revealed that the mRNA and protein expression of Nek2A were significantly up-regulated in MCF-10DCIS.com and MCF-10CA1a cell lines as well as in human primary breast cancer tissue (DCIS and IDC). Our study also presented a correlation between Nek2A mRNA expression and some clinic pathological factors. We found that Nek2A mRNA expression was associated with molecular subtypes, ER, PR and Ki-67 immunoreactivity ( P  〈 0.05) in DCIS and associated with histological grade, lymph node metastasis, molecular subtypes, c-erbB-2, and Ki-67 expression ( P  〈 0.05) in IDC. In addition, we observed that ectopic expression of Nek2A in “normal” immortalized MCF-10A breast epithelial cell resulted in increased Nek2A which lead to abnormal centrosomes. Furthermore, knockdown of Nek2A in MCF-10DCIS.com could remarkably inhibit cell proliferation and induce cell cycle arrest in MCF-10DCIS.com cell line. These data suggested that Nek2A might bear a close relationship with development and progression of breast carcinoma, and highlighted its role as a novel potential biomarker for diagnosis and a possible therapeutic target for human breast cancer especially for DCIS.     Description: Zhang C Z Y, Cao Y, Yun J P, Chen G G & Lai P B S (2012) Histopathology 00, Increased expression of ZBP-89 and its prognostic significance in hepatocellular carcinoma Aims:  ZBP-89 plays a role in cell growth and death. Its expression in hepatocellular carcinoma (HCC) is not well documented. This study aimed to analyse ZBP-89 expression in HCC. Methods and results:  We examined ZBP-89 expression in five HCC cell lines and 182 HCC tissue samples by reverse transcription–polymerase chain reaction (RT–PCR), Western blot analysis and immunofluorescence staining. Our results showed that the expression of ZBP-89 was higher in HCC than adjacent non-tumour liver, at both mRNA and protein levels. ZBP-89 was localized in the nucleus in most HCC tissue samples, but was found in the cytoplasm in 11.5% of cases. Patient survival in those tumours showing high ZBP-89 expression was better than in those with low expression. High ZBP-89 expression tended to be more common in World Health Organization (WHO) grade I than grades II–IV HCC. There was a significant association between HBV positivity and high ZBP-89 expression. Colony formation was reduced dramatically in those HCC cell lines in which ZBP-89 overexpression was demonstrated; this appeared to correlate with increased apoptosis, inferred by finding elevated levels of cleaved poly(ADP-ribose)polymerases (PARP), the probable mechanisms for which may involve increased p53 or p21 expression. Conclusions:  ZBP-89 has anti-tumour properties and is a potential biomarker for prognosis of HCC.

    Description: The effects of β adrenergic receptors (β-ARs) and p38 mitogen-activated protein kinases (MAPK) pathways on cardiosphere-derived cells (CDCs) are largely unknown. This study aimed to investigate the roles of β-ARs and p38MAPK pathways on the proliferation, apoptosis and differentiation capacity of CDCs. The CDCs were treated with β1-AR blocker (Met group), β2-AR antagonist (ICI group) and p38MAPK inhibitor (SB group), nonselective β-AR blocker (PRO group), and β-AR agonist (ISO group). The viability, apoptotic rate and differentiation status of CDCs were determined by MST-1 assay, flow cytometery, and Western blot, respectively. The CDCs viability significantly reduced in ICI group (all P  〈 0.05), and SB group had a significant high viability after 48h treatment (P 〈 0.05). Compared with control group, all treated groups had a low apoptotic rate. After treatment for 72h, ISO treatment elevated the expression of Nkx2.5, and could partially or fully attenuate the inhibitory effects of β-AR antagonists and/or p38MAPK inhibitor. A similar overall trend of protein expression levels among all groups could be observed between protein pairs of cTnT and β1-AR as well as c-Kit and β2-AR, respectively. These results suggested that β-ARs and p38MAPK signaling pathways play crucial roles in the proliferation and differentiation of CDCs. Our findings should be helpful for better understanding the molecular mechanism underlying the physiological processes of CDCs. This article is protected by copyright. All rights reserved

    Description: The climatic effects of irrigation over the Huang-Huai-Hai Plain (3HP) in China are investigated by using the weather research and forecasting model coupled with an operational-like irrigation scheme. Multiple numerical experiments with irrigation off/on during spring, summer, and both spring and summer are conducted. Results show that the warm bias in surface temperature and dry bias in soil moisture are reduced over the 3HP region during the growing seasons by considering the irrigation in the model. The air temperature during nongrowing seasons is also affected by irrigation because of the persistent effects of soil moisture on land–air energy exchanges and ground heat storage. Irrigation can induce significant cooling in the planetary boundary layer (PBL) during the growing seasons and lead to a relatively wet PBL with increased low-level clouds during spring but a relatively dry condition in summer. Further analyses indicate that irrigation leads to increased summer precipitation over the Yangtze River Basin and decreased summer precipitation in southern and northern China. These responses are associated with the changes in the large-scale circulation induced by irrigation. Irrigation tends to cool the atmosphere and forces a possible southward shift of the upper-level jet that can further affect the precipitation distribution. Our model results suggest that in addition to local-scale processes, large-scale impacts should also be considered when studying the precipitation response to irrigation over East Asia.

    Description: ABSTRACT SMARCB1 inactivation occurs in a variety of tumors, being caused by various genetic mechanisms. Since SMARCB1 and EWSR1 genes are located close to each other on at the chromosome 22, larger SMARCB1 deletions may encompass the EWSR1 locus. Herein, we report four cases with SMARCB1 -deletions showing concurrent EWSR1 gene abnormalities by FISH, which lead initially to misinterpretations as EWSR1 -rearranged tumors. Our study group included various morphologies: a poorly differentiated chordoma, an extrarenal rhabdoid tumor, a myoepithelial carcinoma, and a proximal-type epithelioid sarcoma. All cases showed loss of SMARCB1 (INI1) by immunohistochemistry and displayed characteristic histologic features for the diagnoses. The SMARCB1 FISH revealed homozygous or heterozygous deletions in three and one case, respectively. The co-hybridized EWSR1 probes demonstrated either unbalanced split signals or heterozygous deletion in two cases each. The former suggested bona fide rearrangement, while the latter resembled an unbalanced translocation. However, all the FISH patterns were quite complex and distinct from the simple and uniform split signals seen in typical EWSR1 rearrangements. We conclude that in the context of 22q11-12 regional alterations present in SMARCB1 -deleted tumors, simultaneous EWSR1 involvement may be misinterpreted as equivalent to EWSR1 rearrangement. A detailed clinicopathologic correlation and supplementing the EWSR1 FISH assay with complementary methodology is mandatory for correct diagnosis. This article is protected by copyright.     Description: Differentiated embryo chondrocyte 1 (DEC1), a member of basic-helix–loop–helix transcription factor Bhlhe40, also called stimulated by retinoic acid 13, STRA13, plays an important role in the regulation of adipogenesis, tumorigenesis, peripheral circadian output, response to hypoxia and development of metabolic syndrome. Previous studies suggested that DEC1 was involved in skeletal muscle development; however, its precise role in myoblast differentiation has not been determined. In the present study, we showed that DEC1 expressed ubiquitously in different bovine tissues and was down-regulated in differentiated bovine satellite cells. Expression of muscle specific transcription factors (Myf5, MyoD, MyoG and MHC) was significantly down-regulated when DEC1 was over-expressed by both CoCl 2 -simulated hypoxia and Adenovirus-mediated transduction in bovine satellite cells. Consistent with that, promoter analyses via luciferase reporter assay also revealed that overexpression of bovine DEC1 could inhibit MyoG promoter activity. In conclusion, overexpression of DEC1 blocked myogenesis by inhibiting MyoG promoter activity in bovine.     Description: A simple grid cell based distributed hydrologic model was developed to provide spatial information on hydrologic components for determining hydrologically-based critical source areas. The model represents the critical process (soil moisture variation) to runoff generation accounting for both local and global water balance. In this way, it simulates both infiltration excess runoff and saturation excess runoff. The model was tested by multi-site and multi-variable evaluation on the 50 km 2 Little River Experimental Watershed I in Georgia, US, and two smaller nested sub-watersheds. Water balance, hydrograph, and soil moisture were simulated and compared to observed data. For streamflow calibration, the daily Nash-Sutcliffe coefficient was 0.78 at the watershed outlet and 0.56 and 0.75 at the two nested sub-watersheds. For the validation period, the Nash-Sutcliffe coefficients were 0.79 at the watershed outlet and 0.85 and 0.83 at the two sub-watersheds. The percent bias was less than 15% for all sites. For soil moisture, the model also predicted the rising and declining trends at four of the five measurement sites. The spatial distribution of surface runoff simulated by the model was mainly controlled by local characteristics (precipitation, soil properties, and land cover) on dry days, and by global watershed characteristics (relative position within the watershed and hydrologic connectivity) on wet days when saturation excess runoff was simulated. The spatial details of runoff generation and travel time along flow paths provided by the model are helpful for watershed managers to further identify critical source areas of non-point source pollution and develop best management practices.
