    Notes: Abstract The testing behavior and test probing which are a part of host-plant acceptance behavior were studied in P. manihoti. Attention was focused on the testing behavior of three plants of the Manihot genus (Incoza, M'Pembé and Faux-caoutchouc) and a weed of the cassava fields (Talinum). This enabled us not only to characterize the associated behavior but also to show that the mealybug is able to distinguish between different host plants when walking on the leaf. The video description of the test probing of first- and fourth-instar larvae on M'Pembé and on the Faux-caoutchouc shows that the succession of the phases is similar. After a first phase characterized by the repeated intervention of the mealybug sensorial organs, a second phase, more mechanical, with up-and-down head movements, is observed. At this time, the stylets pass through the epidermic and inner tissues. Finally, a third phase, during which the mealybug becomes more agitated, is observed: it stands up using its rear legs and pushes the upper part of its body against the plant. The stylets continue their progression, which is principally intercellular, until they reach the phloem. Longer and longer immobility periods are observed over a period of time. We used the coupled videocamera and electrical penetration graph technique to see the relations existing between outer and inner plant events, i.e., between the behavioral items and the electrical signals characteristic of the stylets pathway in the plant. We observed that the stylet progression in the plant was more difficult in the resistant hybrid Faux-caoutchouc than in the sensitive M'Pembé variety.

    Notes: Abstract  Fluorescein isothiocyanate (FITC) is largely used in immunofluorescence methods. We propose to analyse the quality of some recent fluorochromes using image analysis. Fluorochromes tested include FITC and dichlorotriazinylaminofluorescein (DTAF), dipyrrometheneboron difluoride (BODIPY), Rhodol Green and cyanine 2. RAMOS cells were immunolabelled against the proliferating cell nuclear antigen (PCNA) revealed by the biotin-streptavidin technique. Slides were mounted in anhydrous glycerol or in buffered glycerol (pH 7.0 or pH 8.5). No antifading medium was added. Cell fluorescence emission intensity and bleaching characteristics were measured. Rhodol Green exhibited the highest fluorescence intensity and the best photobleaching resistance. Although BODIPY also resisted well during the photobleaching assay, its fluorescence intensity was weak. FITC, DTAF and cyanine 2 showed intermediate fluorescence intensity and a fast decay of fluorescence. Among the green emitting fluorochromes tested, Rhodol Green appeared to be the best.

    Notes: Summary The variations and characteristics of o-diphenoloxidase activity (O-diphenol-O2-oxidoreductase EC 1.10.3.1) were examined in aging, isolated spinach chloroplasts to determine whether this activity, measured in the presence of 4-methylcatechol as substrate, could be responsible for the inhibition of O2 evolution during aging of these organelles in dark and light. The rate of the Hill reaction (oxygen evolution and the corresponding photoreduction of ferricyanide) during aging in the dark was inhibited at pH 8.0 and stimulated at pH 6.5. This difference did not depend on the nature of the buffer used (Tris-HCl or phosphate). Furthermore, the pH optimum for the ferricyanide-Hill reaction was shifted to lower values (from pH 8.0 to 6.5) on aging of chloroplasts. This phenomenon is probably due to uncoupling during aging. In the light, the Hill reaction was markedly inhibited. However, the ratio μmoles O2 evolved/μmoles ferricyanide reduced diminished slowly in darkness and rapidly when the chloroplasts were aged in the light. Aging of chloroplasts in darkness was accompanied by a slow decrease in the latent period which precedes the initiation of the oxidation, followed by an increase in O-diphenoloxidase activity. Light-aged chloroplasts showed an initial stimulation and then a smaller increase in enzyme activity compared with that of the dark-aged chloroplasts. This latter phenomenon was probably due to secondary reactions caused by photo-inactivation. Under light conditions, the latent period decreased rapidly and disappeared after one hour. This latent period varied considerably with the season and was reduced or obliterated by treatments with light, fatty acids, Triton-X, hypotonic medium and increasing concentrations of substrate: that is by treatments which generally enhance chloroplast swelling. Thus it appears that the latent period is not a characteristic of O-diphenoloxidase but depends on the integrity of chloroplast structure. The enzyme activity was characterized by a stoichiometry of about 1 μmoles O2 consumed per 1.2 μmoles substrate oxidized, indicating that oxidation was probably proceeding further than conversion of O-diphenol to O-diquinone. The latter compound could be used as a Hill oxidant and it permitted measurement of O2 evolution in the same reaction mixture in the presence of light. Under these experimental conditions, O2 evolution (a DCMU sensitive reaction) was first stimulated in dark-aged chloroplasts and rapidly inhibited in light-aged chloroplasts. At appropriate concentrations, KCN, a potent inhibitor of oxidases, enhanced O2 evolution, suggesting that O-diphenoloxidase activity interferes with O2 evolution. This possibility is discussed in view of our previous findings on chloroplast aging in vitro.

    Notes: Abstract The transmembrane distribution of monogalactosyldiacylglycerol and digalactosyldiacylglycerol was determined in chloroplast thylakoids from a range of temperate climate plants. These plants included dicotyledons, monocotyledons, C16:3 and C18:3 plants and herbicide-resistant species. In all the thylakoids examined monogalactosyldiacylglycerol was enriched in the outer leaflet (53–65%) while digalactosyldiacylglycerol was highly enriched in the inner leaflet (78–90%). The non-bilayer forming monogalactosyldiacylglycerol represented 55–81% of the total acyl lipids of the outer monolayer. The relative acyl lipid composition of both leaflets of the thylakoid membrane indicates that the lamellar structure is strongly favored in the inner monolayer, whereas the outer one presents a metastable character which allows the probable coexistence of both lamellar and non-lamellar phases. The consequence of this asymmetry for the stability and function of the thylakoid membrane is discussed.

    Notes: Abstract The double pendulum is a non-integrable Hamiltonian system which exhibits the scenario of transition to global chaos via the decay of a golden mean KAM torus. We apply Greene's method and the MacKay residue criterion and compute the threshold to global chaos. We find that MacKay's method is superior to Greene's since it requires much less numerical work but nevertheless gives accurate results.

    Notes: Abstract The mathematical theory of evidence has been introduced by Glenn Shafer in 1976 as a new approach to the representation of uncertainty. This theory can be represented under several distinct but more or less equivalent forms. Probabilistic interpretations of evidence theory have their roots in Arthur Dempster's multivalued mappings of probability spaces. This leads to random set and more generally to random filter models of evidence. In this probabilistic view evidence is seen as more or less probable arguments for certain hypotheses and they can be used to support those hypotheses to certain degrees. These degrees of support are in fact the reliabilities with which the hypotheses can be derived from the evidence. Alternatively, the mathematical theory of evidence can be founded axiomatically on the notion of belief functions or on the allocation of belief masses to subsets of a frame of discernment. These approaches aim to present evidence theory as an extension of probability theory. Evidence theory has been used to represent uncertainty in expert systems, especially in the domain of diagnostics. It can be applied to decision analysis and it gives a new perspective for statistical analysis. Among its further applications are image processing, project planning and scheduling and risk analysis. The computational problems of evidence theory are well understood and even though the problem is complex, efficient methods are available.
