    Notes: Abstract The kinetics of pentachlorophenol (PCP) degradation by a Flavobacterium sp. ATCC 39723 has been investigated. Sodium glutamate was supplied as an additional carbon source to increase the rate of cell growth and PCP degradation. A kinetic model including PCP toxicity for cell growth and PCP inhibition of its own degradation was developed. This model was also applied to 2,4-dichlorophenol (DCP) degradation by the same organism. Although PCP and DCP are degraded by different pathways, the model describes these two degradation processes very well.

    Notes: Abstract. Voltage operated calcium channels (VOCCs) are implicated in osteoblastic mechano- and hormonal transduction. Very little, however, is known about the expression of VOCCs in osteoblasts of load-bearing bones. Here we describe two types of whole-cell calcium current in rat femoral explant-derived osteoblasts. The first is high-voltage activated and sensitive to nifedipine, Bay K8644 and FPL 64176. The second is low-voltage activated and is sensitive to micromolar concentrations of Ni2+. The properties of these two currents are consistent with those of L-type and T-type calcium currents respectively. T-type currents were detected in most cells on the day of passage, the level of expression being significantly lower on subsequent days. L-type currents were also most common on the day of passage but were detected consistently throughout the 4-day period of study. The reverse transcription polymerase chain reaction with non-specific primers directed against all L-type VOCC α1 subunits and then with specific primers directed against sequences from rat brain α1C (L-type), α1D (L-type) and α1G (T-type) VOCC subunits detected transcripts of appropriate size in all four cases. Products from the three sets of specific primer pairs (α1C, α1D, α1G) were sequenced and were identical to their respective rat brain templates.

    Notes: Abstract A cofactor-independent phosphoglycerate mutase (PGAM-i) was isolated to homogeneity from monocotyledonous Lilium longiflorum Thunb. Two-dimensional (2D) polyacrylamide gel electrophoresis resolved three PGAM-i forms. This enzyme was originally identified by cross-reactivity to antibodies affinity-purified from 2D gels using human vitronectin (VN). Antibody produced against a denatured protein spot from a 2D gel did not recognize VN protein, but partial protein and DNA sequencing showed similarity of the former protein to maize PGAM-i. Immunoblots from roots, styles, leaves, and anthers showed the presence of PGAM-i in all tissues examined; it was isolated predominantly from the soluble cell fraction, with some present in the insoluble cell fraction. Immunoelectron microscopy demonstrated its localization in the cytoplasm and plastids in root cells near the apical meristem. In addition, immunogold labeling detected signals from the nucleus. The immunohistochemical localization of the enzyme in the nucleus, as well as in the cytosol and plastids, indicates that lily PGAM-i might have multiple functions in the cell.

    Notes: Abstract An ac calorimetric method for measuring the thermal diffusivity of thin-film materials has been widely applied. In the application of this method, the systematic errors caused by the heat loss effect, the edge reflection effect, etc., have been analyzed and corresponding correction methods have been developed. But when measuring films with low thermal diffusivity or with thickness comparable to the thermal diffusion length, a two-dimensional effect which will also result in a systematic error of the measurement is present. In this paper, the mechanism of two-dimensional heat conduction within a thin sample which is supplied a periodic heat flux by a chopped light beam is analyzed. A numerical analysis method is developed to study the effect of the two-dimensional heat conduction on the measured thermal diffusivity values. The relations between the measured thermal diffusivity and independent parameters such as frequency, thickness of sample, width of light spot, etc., are demonstrated to indicate the two-dimensional effect. The experimental precondition for minimizing the systematic error caused by the two-dimensional effect is determined. In addition, the analysis method presented in this paper should be useful for more difficult problems such as error estimation of the thermal diffusivity measurement of coatings or composite films.

    Notes: Abstract In the thermal diffusivity measurement of a CVD diamond film using an ac calorimetric method, the reflection of an ac temperature wave at the edge of the film sample should be considered due to the limited length of the sample and its high thermal diffusivity, i.e., the edge effect. In this case, the measured thermal diffusivity is given as a function of frequency. The relation between the measured thermal diffusivity and the frequency is represented as an analytical expression. The real thermal diffusivity is obtained by correcting the edge effect by two means. One is an iterative method using the directly measured edge length of the sample to fit the analytical expression. The other is a parameter estimation method by which a simplex method is used to estimate the edge length and the real thermal diffusivity. Thermal diffusivities of two diamond films were measured, and data were analyzed using the above methods. The result shows that the parameter estimation method is relatively accurate and convenient in processing test data.

    Notes: Abstract We have obtained results for converting focused laser radiation to X-ray line emissions in laser-heated NaF and Cu plasmas. These plasmas were used as the pump source for studying the phenomena of inner-shell photoionization of Zn-like ionic states. Soft X-ray emissions from transitions of 3d 94s 2 → 3d 104p in Cu-like Pd and Ag ions were observed.

    Notes: Abstract. We have previously investigated methods that image high-pressure processes such as combustion inside automobile cylinders and aircraft engines, or chemical phenomena in supercritical fluids. Here we show that vibrational Raman scattering can simply obtain, quantitatively, densities of some combustion-relevant molecules. We use narrow-band KrF excimer-laser light. Measurements for H2, N2, O2, CO2, and CH4 are in the pressure range from 1 to 60 bar, whereas those for C2H6 and C3H8 are up to their respective vapor pressures. All these species are at ambient temperature. Additional measurements are described for CO2 up to 96.8 bar and 318 K, where CO2 is a supercritical fluid. The O2 measurements are complicated by a photochemical formation of O3; those in supercritical CO2 by drastic bending of the laser beam within this medium. We show that, for each gas, the Raman signal is directly proportional to gas density, thereby making quantitative analysis particularly convenient. For each species, we present an estimate of its Raman cross-section relative to that of N2. However we recommend that future diagnostics users calibrate their own systems for relative species sensitivity.

The nucleotide sequence and organization of the human θ1 gene (exons, introns, promoter, and polyadenylation signals) are similar to those reported for the orangutan θ1-globin gene. If these genes are functional, the sequences of their θ1-globin chains would differ by only one amino acid residue (at position 137).
