    Abstract: Intestinal bacterial communities play an important role in the growth and health of aquatic animal hosts and have drawn increasing attention. However, the role of the intestinal microbiota in the growth of freshwater prawns remains unclear. Here, the intestinal microbiota of freshwater prawns (Macrobrachium rosenbergii) at different life stages (one, two, and three months old) were investigated using 16S rRNA sequencing. The results showed that community richness and diversity increased with growth, which might be one of the reasons that the prawns maintained a fast growth rate before sexual maturation. Three core phyla were identified in the one-month-old group, namely, Firmicutes (79.24%), Proteobacteria (17.09%) and Actinobacteriota (2.01%). Five core phyla were identified in the two-month-old group, including Firmicutes (47.84%), Proteobacteria (44.22%), Actinobacteriota (1.83%), Acidobacteriota (1.66%) and Bacteroidota (1.24%), and the core phyla in the three-month-old group were similar, except for Acidobacterota, which was not identified. A total of 12 core genera were identified in all samples, and significant differences were observed in the relative abundance of gut microbiota between the three groups (p 〈 0.05). Exiguobacterium, Lactococcus and Shewanella were the three most significantly differentially abundant genera between stages. In addition, Candidatus Hepatoplasma was detected only in the two- and three-month-old prawn groups. This study provides information on the differences in the intestinal microbiota in different developmental stages, which contribute to adaptation to salinity in the early developmental stage and digestive ability to meet the growth needs of Macrobrachium rosenbergii.

    Abstract: Three series of curcumin derivatives including phosphorylated, etherified, and esterified products of curcumin were synthesized, and their anti-tumor activities were assessed against human breast cancer MCF-7, hepatocellular carcinoma Hep-G2, and human cervical carcinoma HeLa cells. Compared with curcumin, compounds 3, 8, and 9 exhibited stronger antitumor cell line growth activities against HeLa cells. Compound 12 also showed higher antitumor cell line growth activities on MCF-7 cells than curcumin. Among them, 4-((1E,6E)-7-(4-Hydroxy-3-methoxyphenyl)-3,5-dioxohepta-1,6-dienyl)-2-methoxyphenyl dihydrogen phosphate(3) showed the strongest activity with an half maximal inhibitory concentration (IC50) of 6.78 µM against HeLa cells compared with curcumin with an IC50 of 17.67 µM. Stabilities of representatives of the three series were tested in rabbit plasma in vitro, and compounds 3 and 4 slowly released curcumin in plasma. The effect of compound 3 on HeLa cell apoptosis was determined by examining morphological changes by DAPI (4′,6-diamidino-2-phenylindole) staining as well as Annexin V-FITC/ Propidium Iodide (PI) double staining and flow cytometry. The results showed that 3 induced cellular apoptosis in a dose-dependent manner. Together our findings show that 3 merits further investigation as a new potential antitumor drug candidate.

    Abstract: Twelve new and four known alkaloids including five different structural scaffolds were isolated from the sponge Stylissa massa collected in the South China Sea. Compound 1 is the first identified precursor metabolite of the classic 5/7/5 tricyclic skeleton with unesterified guanidine and carboxyl groups, compounds 2–5 and 13–15 belong to the spongiacidin-type pyrrole imidazole alkaloids (PIAs). Z- and E-configurations of the spongiacidin-type PIAs often appeared concomitantly and were distinguished by the chemical shift analysis of 13C NMR spectra. The structures of all twelve new compounds were determined by NMR, MS, and ECD analysis combined with single-crystal data of compounds 1, 5, and 10. In the aldose reductase (ALR2) inhibitory assay, six 5/7/5 tricyclic compounds (2–5, 13–15) displayed significant activities. Compounds 13 and 14, as the representative members of spongiacidin-PIAs, demonstrated their ALR2-targeted activities in SPR experiments with KD values of 12.5 and 6.9 µM, respectively.
