    Abstract: Background: In the treatment of advanced urothelial (aUC) and renal cell carcinoma (aRCC), biomarkers such as PD-1 and PD-L1 are not robust prognostic markers for immunotherapy (IO) response. Previously, a significant association between IO and a change in splenic volume (SV) was described for several tumour entities. To the best of our knowledge, this study presents the first correlation of SV to IO in aUC and aRCC. Methods: All patients with aUC (05/2017–10/2021) and aRCC (01/2012–05/2022) treated with IO at our academic centre were included. SV was measured at baseline, 3 and 9 months after initiation of IO using an in-house developed convolutional neural network-based spleen segmentation method. Uni- and multivariate Cox regression models for overall survival (OS) and progression-free survival (PFS) were used. Results: In total, 35 patients with aUC and 30 patients with aRCC were included in the analysis. Lower SV at the three-month follow-up was significantly associated with improved OS in the aRCC group. Conclusions: We describe a new, innovative artificial intelligence-based approach of a radiological surrogate marker for IO response in aUC and aRCC which presents a promising new predictive imaging marker. The data presented implicate improved OS with lower follow-up SV in patients with aRCC.

    Abstract: Wendelstein 7-X (W7-X) is the leading experiment on the path of demonstrating that stellarators are a feasible concept for a future power plant. One of its major goals is to prove quasi-steady-state operation in a reactor-relevant parameter regime. The surveillance and protection of the water-cooled plasma-facing components (PFCs) against overheating is fundamental to guarantee a safe steady-state high-heat-flux operation. The system has to detect thermal events in real-time and timely interrupt operation if it detects a critical event. The fast reaction times required to prevent damage to the device make it imperative to automate fully the image analysis algorithms. During the past operational phases, W7-X was equipped with inertially cooled test divertor units and the system still required manual supervision. With the experience gained, we have designed a new real-time PFC protection system based on image processing techniques. It uses a precise registration of the entire field of view against the CAD model to determine the temperature limits and thermal properties of the different PFCs. Instead of reacting when the temperature limits are breached in certain regions of interest, the system predicts when an overload will occur based on a heat flux estimation, triggering the interlock system in advance to compensate for the system delay. To conclude, we present our research roadmap towards a feedback control system of thermal loads to prevent unnecessary plasma interruptions in long high-performance plasmas.

    Abstract: Background: Undesirable side effects from wearing face masks during the ongoing COVID-19 pandemic continue to be discussed and pose a challenge to occupational health and safety when recommending safe application. Only few studies examined the effects of continuously wearing a face mask for more than one hour. Therefore, the influence of wearing a medical mask (MedMask) and a filtering facepiece class II respirator (FFP2) on the physiological and subjective outcomes in the course of 130 min of manual work was exploratively investigated. Physical work load and cardiorespiratory fitness levels were additionally considered as moderating factors. Methods: Twenty-four healthy subjects (12 females) from three different cardiorespiratory fitness levels each performed 130 min of simulated manual work with light and medium physical workload using either no mask, a MedMask or FFP2. Heart rate, transcutaneous oxygen and carbon dioxide partial pressure (PtcO2, PtcCO2) as well as perceived physical exertion and respiratory effort were assessed continuously at discrete time intervals. Wearing comfort of the masks were additionally rated after the working period. Results: There was no difference in time-dependent changes of physiological outcomes when using either a MedMask or a FFP2 compared to not wearing a mask. A stronger increase over time in perceived respiratory effort occurred when the face masks were worn, being more prominent for FFP2. Physical workload level and cardiorespiratory fitness level were no moderating factors and higher wearing comfort was rated for the MedMask. Conclusion: Our results suggest that using face masks during light and medium physical manual work does not induce detrimental side effects. Prolonged wearing episodes appeared to increase respiratory effort, but without affecting human physiology in a clinically relevant way.

    Abstract: The ongoing COVID-19 pandemic requires wearing face masks in many areas of our daily life; hence, the potential side effects of mask use are discussed. Therefore, the present study explores whether wearing a medical face mask (MedMask) affects physical working capacity (PWC). Secondary, the influence of a filtering facepiece mask with exhalation valve class 2 (FFP2exhal) and a cotton fabric mask (community mask) on PWC was also investigated. Furthermore, corresponding physiological and subjective responses when wearing face masks as well as a potential moderating role of subjects’ individual cardiorespiratory fitness and sex on face mask effects were analyzed. Thirty-nine subjects (20 males, 19 females) with different cardiorespiratory fitness levels participated in a standardized submaximal bicycle ergometer protocol using either a MedMask, FFP2exhal, community mask, or no mask (control) on four days, in randomized order. PWC130 and PWC150 as the mechanical load at the heart rates of 130 and 150 beats per minute were measured as well as transcutaneous carbon dioxide partial pressure, saturation of peripheral capillary oxygen, breathing frequency, blood pressure, perceived respiratory effort, and physical exhaustion. Using the MedMask did not lead to changes in PWC or physiological response compared to control. Neither appeared changes exceeding normal ranges when the FFP2exhal or community mask was worn. Perceived respiratory effort was up to one point higher (zero-to-ten Likert scale) when using face masks (p 〈 0.05) compared to control. Sex and cardiorespiratory fitness were not factors influencing the effects of the masks. The results of the present study provide reason to believe that wearing face masks for infection prevention during the COVID-19 pandemic does not pose relevant additional physical demands on the user although some more respiratory effort is required.

    Abstract: Spatiotemporally accurate estimates of crop traits are essential for both scientific modeling and practical decision making in sustainable agricultural management. Besides efficient and concise methods to derive these traits, site- and crop-specific reference data are needed to develop and validate retrieval methods. To address this shortcoming, this study first includes the establishment of ’MISPEL’, a comprehensive spectral library (SpecLib) containing hyperspectral measurements and reference data for six key traits of ten widely grown crops. Secondly, crop-specific statistical leaf area index (LAI) models for winter wheat are developed based on a hyperspectral (MISPELFR) and a simulated Sentinel-2 (MISPELS2) SpecLib applying four nonparametric methods. Finally, an independent Sentinel-2 model evaluation at the DEMMIN test site in Germany is conducted, including a comparison with the commonly used SNAP-LAI product. To date, MISPEL comprises a set of 1411 spectra of ten crops and more than 6800 associated reference measurements. Cross-validations of winter wheat LAI models revealed that Elastic-net generalized linear model (GLMNET) and Gaussian process (GP) regressions outperformed partial least squares (PLS) and random forest (RF) regressions, showing RSQ values up to 0.86 and a minimal NRMSE of 0.21 using MISPELFR. GLMNET and GP models based on MISPELS2 further outperformed SNAP-based LAI estimates derived for the external validation site. Thus, it is concluded that the presented SpecLib ’MISPEL’ and applied methodology have a very high potential for deriving diverse crop traits of multiple crops in view of most recent and future multi-, super-, and hyperspectral satellite missions.

    Abstract: Genomic surveillance of the SARS-CoV-2 pandemic is crucial and mainly achieved by amplicon sequencing protocols. Overlapping tiled-amplicons are generated to establish contiguous SARS-CoV-2 genome sequences, which enable the precise resolution of infection chains and outbreaks. We investigated a SARS-CoV-2 outbreak in a local hospital and used nanopore sequencing with a modified ARTIC protocol employing 1200 bp long amplicons. We detected a long deletion of 168 nucleotides in the ORF8 gene in 76 samples from the hospital outbreak. This deletion is difficult to identify with the classical amplicon sequencing procedures since it removes two amplicon primer-binding sites. We analyzed public SARS-CoV-2 sequences and sequencing read data from ENA and identified the same deletion in over 100 genomes belonging to different lineages of SARS-CoV-2, pointing to a mutation hotspot or to positive selection. In almost all cases, the deletion was not represented in the virus genome sequence after consensus building. Additionally, further database searches point to other deletions in the ORF8 coding region that have never been reported by the standard data analysis pipelines. These findings and the fact that ORF8 is especially prone to deletions, make a clear case for the urgent necessity of public availability of the raw data for this and other large deletions that might change the physiology of the virus towards endemism.

    Abstract: DNA-binding protein A (DbpA) belongs to the Y-box family of cold shock domain proteins that exert transcriptional and translational activities in the cell via their ability to bind and regulate mRNA. To investigate the role of DbpA in kidney disease, we utilized the murine unilateral ureter obstruction (UUO) model, which recapitulates many features of obstructive nephropathy seen in humans. We observed that DbpA protein expression is induced within the renal interstitium following disease induction. Compared with wild-type animals, obstructed kidneys from Ybx3-deficient mice are protected from tissue injury, with a significant reduction in the number of infiltrating immune cells as well as in extracellular matrix deposition. RNAseq data from UUO kidneys show that Ybx3 is expressed by activated fibroblasts, which reside within the renal interstitium. Our data support a role for DbpA in orchestrating renal fibrosis and suggest that strategies targeting DbpA may be a therapeutic option to slow disease progression.

    Abstract: Background: There is increasing evidence that the reliance on synthetic chemical pesticides and mineral fertilizers in agriculture has significant negative environmental and/or health impacts and poses a risk for future food security. Systematic reviews/meta-analyses showed that organic production systems, which omit the use of agrochemicals, produce crops with lower yields, but superior nutritional composition. However, the agronomic parameters responsible for differences in crop yields and nutritional quality are poorly understood. Methods: Here we report results for four field vegetable crops from the Nafferton Factorial Systems Comparison (NFSC) trial. This long-term factorial field experiment was designed to (i) identify effects of growing season/climatic variation, and contrasting rotational designs, crop protection protocols and fertilization regimes used in organic and conventional systems on crop health, yield and nutritional parameters and (ii) estimate the relative importance of climatic and agronomic drivers for crop health, yield and nutritionally relevant quality parameters. Quality parameters monitored in harvested products, included phenolic, glucosinolate, vitamin C, vitamin B9, carotenoid, cadmium (Cd), nickel (Ni), lead (Pb) and glycoalkaloid concentrations. Results: Climatic conditions during the growing season were found to have a larger impact on crop yield and quality than the agronomic factors (pre-crop, crop protection, fertilization) studied. However, the (i) interactions between growing season with contrasting climatic conditions and agronomic factors identified by ANOVA for crop health, yield and quality parameters and (ii) the associations between the three climatic drivers (precipitation, temperature, radiation) and crop yield and quality parameters differed substantially between the four crop plant species. Among the agronomic factors, fertilization had a substantially larger impact compared with both pre-crop and crop protection. Specifically, crop yields were found to be significantly increased by the use of (i) conventional fertilization and crop protection methods in potato, (ii) conventional fertilization, but organic crop protection methods in cabbage, and (iii) conventional fertilization regimes in lettuce, while none of the agronomic factors had a significant effect on onion yields. When important crop pest and diseases were assessed, (i) conventional crop protection resulted in significantly lower late blight severity in potato, while (ii) organic crop protection resulted in lower bird damage and cabbage root fly (CRF) incidence in cabbages, and Sclerotinia incidence in lettuce and (iii) organic fertilization resulted in lower CRF and Sclerotinia incidence in cabbage and lettuce respectively. When concentrations of nutritionally relevant phytochemicals were compared, organic fertilization resulted in significantly higher phenolic concentrations in potato, cabbage and lettuce, higher glucosinolate and carotenoid concentrations in cabbage, higher vitamin C concentrations in potato and cabbage and higher vitamin B9 concentrations in potato and lettuce—but lower concentrations of toxic glycoalkaloids in potato. Significant effects of crop protection protocols on phytochemical concentrations were only detected in cabbage with conventional crop protection resulting in higher glucosinolate and vitamin B9 concentrations. When toxic metal concentrations were compared, organic fertilization resulted in significantly lower Cd concentrations in all four crops and lower Ni concentrations in potato, cabbage and onion. Significant effects of crop protection were only detected in cabbage, where organic crop protection resulted in lower Ni concentrations. Pb concentrations were not affected by any of the agronomic factors. The potential implications of results for improving (i) strategies to reduce the use of non-renewable resources and environmental impacts of vegetable production and (ii) the productivity of organic and other low-input vegetable production systems without compromising food quality are discussed. Conclusions: The study confirms that organic vegetable production protocols result in higher concentrations of phenolics and other nutritionally desirable phytochemicals, but lower concentrations of the toxic metals Cd and Ni in harvested products. It also demonstrates, for the first time, that this is primarily due to differences in fertilization regimes. The finding that in three of the four crops (cabbage, lettuce and onion) the application of synthetic chemical crop protection products had no measurable positive impact on crop health and yield should be considered in the context of the growing concern about health impacts of pesticide use in field vegetable crops.
