    Abstract: In recent decades, great efforts have been made to develop innovative, effective, and accurate nanofabrication techniques stimulated by the growing demand for nanostructures. Nowadays, mechanical tip-based emerged as the most promising nanolithography technique, allowing the pattern of nanostructures with a sub-nanometer resolution, high reproducibility, and accuracy. Unfortunately, these nanostructures result in contoured pile-ups that could limit their use and future integration into high-tech devices. The removal of pile-ups is still an open challenge. In this perspective, two different AFM-based approaches, i.e., Force Modulation Mode imaging and force-distance curve analysis, were used to characterize the structure of pile-ups at the edges of nanogrooves patterned on PMMA substrate by means of Pulse-Atomic Force Lithography. Our experimental results showed that the material in pile-ups was less stiff than the pristine polymer. Based on this evidence, we have developed an effective strategy to easily remove pile-ups, preserving the shape and the morphology of nanostructures.

    Abstract: Corallium rubrum and Eunicella cavolini are two octocorals, reported as co-occurring species in the deep rocky habitats of the Mediterranean Sea with a high hydrodynamic and moderate eutrophication. Their spatial distribution and demography in the deep sea are mainly affected by temperature and direct and indirect anthropogenic activities; however, knowledge of the factors that potentially influence their co-existence is scarce. This paper provides novel data on the distribution and demography of these two species, at depths between 50 and 290 m in the Western Mediterranean Sea, providing insights on their co-occurrence. Both species exhibited the highest population density at deeper sites ( 〉 150 m), showing an inverse size–density relation. Density values ranged from 0.03 colonies m−2 to 32 and 80 col. m−2 for yellow gorgonian and red coral, respectively. The two species co-occurred in 13% of the total frames examined, mostly dwelling between 120 and 160 m depth. Distance-based linear modeling (DistLM) emphasized that when co-occurring the variability of the two species’ densities were significantly driven by the density—rather than the morphology (i.e., height)—of the other species. We stress the need for further studies to elucidate the possible mutual effects of suspension feeders and to test the role of different environmental factors potentially influencing inter-specific relationships.

    Abstract: The recruitment process is a fundamental step in population life cycles that determines survival, population demographic structure, and dynamics. The success of recruitment events repeated over successive years greatly affects the survival of long-lived gorgonian populations. Here, we report the recruitment process of the precious, heavily harvested Mediterranean gorgonian Corallium rubrum (red coral) on both settlement tiles and natural substrates over different Mediterranean areas. Red coral is a gonochoric internal brooder that reproduces in early summer. Lecithotrophic planulae settle 15–30 days after release in semi-dark environments at depths between 15 and 800 m. In autumn, 0.58–0.68 mm-wide recruits can be observed on the vaults of small crevices and caves and on rocky cliffs and boulders. Owing to their small size, there is limited knowledge of C. rubrum recruitment in the field. In this study, we examined the recruitment density and distribution in Canadells (Banyuls sur Mer, France) and Calafuria (Livorno, Italy) and compared these findings with those collected over different Mediterranean areas. Red coral exhibited high recruitment values ranging from 0.43 to 13.19 recruits dm−2. The distribution pattern of recruits, examined at a small spatial scale via nearest-neighbor distance analysis, revealed a significantly higher patch frequency on the natural substrate than on settlement tiles, presumably because of the scarcely available spots of free space on the former substrate, which are crowded by competitor species.

    Abstract: Seascape mapping is critical to understanding ecosystem services and managing areas with potential for fishing, power generation, mining, and tourism. Despite advances in marine geophysics, the necessary equipment to make underwater cartography can be expensive and requires a certain degree of specialization. In areas with scarce data, ethnomapping can be used for the elaboration or complementation of marine cartography. In addition, it provides information about the nature, concepts, phenomena, and nomenclatures attributed by the local population. The aim of this study was to integrate the knowledge of artisanal fishermen from NE Brazil into the mapping of seascapes, validating the obtained cartography with scientific sampling. Focus groups were used to promote an open discussion of local users’ knowledge about seascapes and their importance for local fisheries. After analyzing, it was possible to correlate the products of participatory mapping with the scientific data available in the literature, resulting in the seascape ethnomap. Nine seascapes relevant to subsistence fisheries were identified. The mapping of seascapes and fauna captured by indigenous fishermen was similar to that produced from preexisting geology, geomorphology, and fishing data. This validated the methodological protocol and the importance of the participation of local populations in coastal conservation and management activities.

    Abstract: An accurate and easy-to-use Q3 system for on-chip quantitative real-time Polymerase Chain Reaction (qPCR) is hereby demonstrated, and described in detail. The qPCR reactions take place inside a single-use Lab-on-a-Chip with multiple wells, each with 5 to 15 µL capacity. The same chip hosts a printed metal heater coupled with a calibrated sensor, for rapid and accurate temperature control inside the reaction mixture. The rest of the system is non-disposable and encased in a 7 × 14 × 8.5 (height) cm plastic shell weighing 300 g. Included in the non-disposable part is a fluorescence read-out system featuring up to four channels and a self-contained control and data storage system, interfacing with an external user-friendly software suite. Hereby, we illustrate the engineering details of the Q3 system and benchmark it with seamlessly ported testing protocols, showing that Q3 equals the performance of standard commercial systems. Overall, to the best of our knowledge, this is one of the most mature general-purpose systems for on-chip qPCR currently available.

    Abstract: To define the role of spleen stiffness (SS) and liver stiffness (LS) in myelofibrosis and other Philadelphia (Ph)-negative myeloproliferative neoplasms (MPNs), we studied, by ultrasonography (US) and elastography (ES), 70 consecutive patients with myelofibrosis (MF) (no.43), essential thrombocythemia (ET) (no.10), and polycythemia vera (PV) (no.17). Overall, the median SS was not different between patients with MF and PV (p = 0.9); however, both MF and PV groups had significantly higher SS than the ET group (p = 0.011 and p = 0.035, respectively) and healthy controls (p 〈 0.0001 and p = 0.002, respectively). In patients with MF, SS values above 40 kPa were significantly associated with worse progression-free survival (PFS) (p = 0.012; HR = 3.2). SS also correlated with the extension of bone marrow fibrosis (BMF) (p 〈 0.0001). SS was higher in advanced fibrotic stages MF-2, MF-3 (W.H.O. criteria) than in pre-fibrotic/early fibrotic stages (MF-0, MF-1) (p 〈 0.0001) and PFS was significantly different in the two cohorts, with values of 63% and 85%, respectively (p = 0.038; HR = 2.61). LS significantly differed between the patient cohort with MF and healthy controls (p = 0.001), but not between the patient cohorts with ET and PV and healthy controls (p = 0.999 and p = 0.101, respectively). We can conclude that organ stiffness adds valuable information to the clinical work-up of MPNs and could be employed to define patients at a higher risk of progression.
