    Abstract: Postoperative peritoneal adhesion (PPA) is a major clinical complication after open surgery or laparoscopic procedure. Ligustrazine is the active ingredient extracted from the natural herb Ligusticum chuanxiong Hort, which has promising antiadhesion properties. This study is aimed at revealing the underlying mechanisms of ligustrazine in preventing PPA at molecular and cellular levels. Both rat primary peritoneal mesothelial cells (PMCs) and human PMCs were used for analysis in vitro. Several molecular biological techniques were applied to uncover the potential mechanisms of ligustrazine in preventing PPA. And molecular docking and site-directed mutagenesis assay were used to predict the binding sites of ligustrazine with PPARγ. The bioinformatics analysis was further applied to identify the key pathway in the pathogenesis of PPA. Besides, PPA rodent models were prepared and developed to evaluate the novel ligustrazine nanoparticles in vivo. Ligustrazine could significantly suppress hypoxia-induced PMC functions, such as restricting the production of profibrotic cytokines, inhibiting the expression of migration and adhesion-associated molecules, repressing the expression of cytoskeleton proteins, restricting hypoxia-induced PMCs to obtain myofibroblast-like phenotypes, and reversing ECM remodeling and EMT phenotype transitions by activating PPARγ. The antagonist GW9662 of PPARγ could restore the inhibitory effects of ligustrazine on hypoxia-induced PMC functions. The inhibitor KC7F2 of HIF-1α could repress hypoxia-induced PMC functions, and ligustrazine could downregulate the expression of HIF-1α, which could be reversed by GW9662. And the expression of HIF-1α inhibited by ligustrazine was dramatically reversed after transfection with si-SMRT. The results showed that the benefit of ligustrazine on PMC functions is contributed to the activation of PPARγ on the transrepression of HIF-1α in an SMRT-dependent manner. Molecular docking and site-directed mutagenesis tests uncovered that ligustrazine bound directly to PPARγ, and Val 339/Ile 341 residue was critical for the binding of PPARγ to ligustrazine. Besides, we discovered a novel nanoparticle agent with sustained release behavior, drug delivery efficiency, and good tissue penetration in PPA rodent models. Our study unravels a novel mechanism of ligustrazine in preventing PPA. The findings indicated that ligustrazine is a potential strategy for PPA formation and ligustrazine nanoparticles are promising agents for preclinical application.

    Abstract: Objectives. Factors prognostic of survival in liver transplant (LT) recipients with hepatitis B virus related acute-on-chronic liver failure (HBV-ACLF) remain unclear. This study evaluated risk factors for survival in LT recipients with HBV-ACLF and determined the scoring system optimal for assessing patient prognosis. Methods. This retrospective study included 323 HBV-ACLF related patients undergoing LT, including 112, 146, and 65 patients with HBV-ACLF grades 1, 2, and 3, respectively. Overall survival (OS) was estimated by the Kaplan–Meier method, and factors associated with survival were analysed by multivariate Cox proportional hazards models. Pretransplant prognostic scoring systems were compared by receiver operating characteristic (ROC) curve analysis. Results. The one-year survival rate was significantly lower in HBV-ACLF grade 3 (80.0%) than in grades 1 (93.8%) and 2 (91.8%) recipients ( p = 0.0063 ). Cox multivariate analysis showed that age 〉 53 years (hazard ratio (HR) 3.731; 95% confidence interval (CI) 1.640–8.407), WBC count 〉 8.6 × 109/L (HR 4.544; 95% CI 1.140–18.107), HBV-ACLF 3 (HR 2.729; 95% CI 1.050–7.096), and cold ischaemia time 〉 8.5 hours (HR 2.867; 95% CI, 1.38–5.921) were independently prognostic of 1-year survival. Comparisons of pretransplant scoring systems showed that chronic liver failure-consortium ACLF score (CLIF-C ACLFs) was superior to COSSH-ACLF, MELD-Na, and MELD scores in predicting 1-year OS in these patients. Conclusions. Age 〉 53 years, WBC counts 〉 8.6 × 109/L, HBV-ACLF grade 3, and cold ischaemia time 〉 8.5 hours are independently prognostic of OS in LT recipients with HBV-ACLF. CLIF-C ACLFs is superior to other scoring methods in predicting 1-year OS in these patients.

    Abstract: Previous studies reported discrepant white matter diffusivity in mild traumatic brain injury (mTBI) on the base of Glasgow Coma Scale, which are unreliable for some TBI severity indicators and the frequency of missing documentation in the medical record. In the present study, we adopted the Mayo classification system for TBI severity. In this system, the mTBI is also divided into two groups as “probable and symptomatic” TBI. We aimed to investigate altered microstructural integrity in symptomatic acute TBI ( 〈 1 week) by using tract-based spatial statics (TBSS) approach. A total of 12 patients and 13 healthy volunteers were involved and underwent MRI scans including conventional scan, and SWI and DTI. All the patients had no visible lesions by using conventional and SWI neuroimaging techniques, while showing widespread declines in the fractional anisotropy (FA) of gray matter and white matter throughout the TBSS skeleton, particularly in the limbic-subcortical structures. By contrast, symptomatic TBI patients showed no significant enhanced changes in FA compared to the healthy controls. A better understanding of the acute changes occurring following symptomatic TBI may increase our understanding of neuroplasticity and continuing degenerative change, which, in turn, may facilitate advances in management and intervention.

    Abstract: Maternal obesity is associated with placental oxidative stress. However, the mechanism underlying this association remains poorly understood. In the present study, a gilt obesity model was developed by exposure to different energy diets and used to investigate the role of NADPH oxidase 2 (Nox2) in the placenta. Specifically, 99 gilts (Guangdong Small-ear Spotted pig) at day 60 of gestation were randomly assigned to one of the following three treatments: low-energy group (L, DE = 11.50   MJ / kg ), medium-energy group (M, DE = 12.41   MJ / kg ), and high-energy group (H, DE = 13.42   MJ / kg ), with 11 replicate pens per treatment and 3 gilts per pen. At the start of the study, maternal body weight and backfat thickness were not significantly different in the three treatments. After the study, data indicated that the H group had higher body weight and backfat thickness gain for gilts during gestation and lower piglet birth weight compared with the other two groups. Additionally, the H group showed glucolipid metabolic disorders and increased triglyceride and nonesterified fatty acid contents in the placenta of gilts. Compared with the L group, the H group exhibited lower mitochondrial biogenesis and increased oxidative damage in the placenta. Importantly, increased mRNA expression and protein abundance of Nox2 were observed for the first time in H group placentae. Furthermore, compared with the L group, the H group showed a decrease in the density of placental vessels and the protein levels of vascular endothelial cadherin (VE-cadherin), vascular endothelial growth factor A (VEGF-A), and phosphorylation of vascular endothelial growth factor receptor 2 (p-VEGFR2) as well as the immunostaining intensity of platelet endothelial cell adhesion molecule-1 (CD31). Our findings suggest that maternal high-energy diet-induced obesity increases placental oxidative stress and decreases placental angiogenesis possibly through the upregulation of Nox2.

    Abstract: In NAND flash storage devices, the random access memory (RAM) is composed of a data buffer and mapping cache that play critical roles in storage performance. Furthermore, as the capacity growth rate of RAM chips lags far behind that of flash memory chips, determining how to take advantage of precious RAM is still a crucial issue. However, most existing buffer management studies on storage devices report performance degradation since these devices cannot refine reference regularities such as sequential, hot, or looping data patterns. In addition, most of these studies focus only on separately managing the data buffer or mapping cache. Compared with the existing buffer/cache management schemes (BMSs), we propose a unified RAM management (URM) scheme for not only the mapping cache but also the data buffer in NAND flash storage devices. URM compresses the mapping table to save memory space, and the remaining dynamic RAM space is used for the data buffer. For the data buffer part, we utilize the program counter-technique in the host layer that provides automatic pattern recognition for different applications, in contrast to existing BMSs. The program counter-technique in our design is able to distinguish four patterns. According to these patterns, the data buffer is divided into four size-adjustable zones. Therefore, our approach is linked to multimodal data and used in a data-intensive system. In particular, in URM, we use a multivariate classification to predict prefetching length in mapping buffer management. Our multivariate classification is transformed into multiple binary classifications (logistic regressions). Finally, we extensively evaluate URM using various realistic workloads, and the experimental results show that, compared with three data buffer management schemes, CFLRU, BPLRU, and VBBMS, URM can improve the hit ratio of data buffer and save response time by an average to 32% and 18%, respectively.

    Abstract: The accumulation of heavy metals (HMs) in the water-level fluctuation zone (WLFZ) of the Three Gorges Reservoir (TGR) area is potentially harmful to the water environment. In order to reveal whether nitrogen (N) deposition is a potential driving factor for the migration and transformation of HMs (Cd, Cr, Cu, Ni, and Pb), a simulated N deposition experiment was performed on the soil in the WFLZ of the TGR. The results showed that the accumulative release amounts of HMs increased with the increase of N deposition. It was found that the Elovich equation, double-constant equation, and parabolic diffusion equation could well describe the release process of Cu, Cd, Cr, and Ni, while the double-constant equation, parabolic diffusion equation, and first-order equation could be applicable for Pb. The exchangeable fractions of HMs increased to varying degrees after the N deposition treatment, wherein Ni was most significant, indicating that N deposition could increase the ecological risk of HM pollution in the TGR area. The results provide insight into the major factors affecting the release of different HMs under N deposition in this vulnerable region ecologically.

    Abstract: Vertically aligned silver nanorods were good substrates for surface-enhanced Raman scattering. The surface-enhanced Raman scattering sensitivity of nanorods can be regulated through the method that the silver nanorod is divided into four uniform silver sections using five uniform silicon sections. And the length of silicon sections is the key factor in regulating the surface-enhanced Raman scattering sensitivity. In the regulation, the best surface-enhanced Raman scattering performance is about 4 times as large as the worst performance. The study provides an effective way to regulate the surface-enhanced Raman scattering sensitivity of silver nanorods and its possible explanation about mechanism.

    Abstract: As structural material and moderator in high temperature gas-cooled reactor (HTGR), nuclear graphite endures large flux of irradiation in its service time. The microstructure of nuclear graphite is a topical issue studied to predict the irradiation property of graphite and improve manufacturing process. In our present work, the pores in graphite are focused, and the relationship between pore and irradiation behavior is discussed. Three kinds of nuclear graphite (IG-11, NBG-18, and HSM-SC) are concerned, and their porosity, pore size, and morphology before and after irradiation are studied, respectively. A comparison between the three graphites shows that dense small pores which are uniformly distributed in graphite bring better irradiation property because the pores can accommodate some of the internal stress caused by irradiation expansion. Coke particles of small size and a thorough mixture between coke and binder are suggested to obtain such pores in nuclear graphite and thus improve irradiation property.

    Abstract: Surface-enhanced Raman scattering (SERS) was discovered in 1974 and impacted Raman spectroscopy and surface science. Although SERS has not been developed to be an applicable detection tool so far, nanotechnology has promoted its development in recent decades. The traditional SERS substrates, such as silver electrode, metal island film, and silver colloid, cannot be applied because of their enhancement factor or stability, but newly developed substrates, such as electrochemical deposition surface, Ag porous film, and surface-confined colloids, have better sensitivity and stability. Surface enhanced Raman scattering is applied in other fields such as detection of chemical pollutant, biomolecules, DNA, bacteria, and so forth. In this paper, the development of nanofabrication and application of surface-enhanced Ramans scattering substrate are discussed.
