    Abstract: We propose a novel method that predicts binding of G-protein coupled receptors (GPCRs) and ligands. The proposed method uses hub and cycle structures of ligands and amino acid motif sequences of GPCRs, rather than the 3D structure of a receptor or similarity of receptors or ligands. The experimental results show that these new features can be effective in predicting GPCR-ligand binding (average area under the curve [AUC] of 0.944), because they are thought to include hidden properties of good ligand-receptor binding. Using the proposed method, we were able to identify novel ligand-GPCR bindings, some of which are supported by several studies.

    Abstract: The initial software fitting prescribed by the fitting formula largely depends on the patient’s hearing loss, which may not be the optimal preference for a particular user. Certain criteria must also be readjusted by an audiologist to meet the user-specific requirements. Therefore, this study focuses on the novel application of a neural network (NN) technique to build a suitable fitting algorithm with prescribed hearing loss and the corresponding preferred gain to minimize the gap between optimized fittings. The algorithm intended to learn the hearing preferences of an individual user such that the initial fitting may be optimized. These findings demonstrate the efficiency of the algorithm, with and without additional features. Using the clinical fitting data, the average mean square error (MSE) for the simple NN algorithm was 5.4183%. By adding additional features to the data, the algorithm performed better, and the average MSE was as low as 5.2530%. However, the algorithm outperformed Company A fitting software, as the MSE was the highest at 5.4748%. As the company’s automatic fitting has a noticeable discrepancy with clinical fitting records, the impeccable results from this study can lead to a better path towards fitting satisfaction, thus benefiting the hearing-impaired community to a larger extent.

    Abstract: Ursolic acid (UA) is the major active component of the loquat leaf extract (LLE) and several previous studies have indicated that UA may have the ability to prevent skeletal muscle atrophy. Therefore, we conducted a randomized, double-blind, and placebo-controlled study to investigate the effects of the LLE on muscle strength, muscle mass, muscle function, and metabolic markers in healthy adults; the safety of the compound was also evaluated. We examined the peak torque/body weight at 60°/s knee extension, handgrip strength, skeletal muscle mass, physical performance, and metabolic parameters at baseline, as well as after 4 and 12 weeks of intervention. Either 500 mg of LLE (50.94 mg of UA) or a placebo was administered to fifty-four healthy adults each day for 12 weeks; no differences in muscle strength, muscle mass, and physical performance were observed between the two groups. However, the right-handgrip strength of female subjects in the LLE group was found to be significantly better than that of subjects in the control group ( P = 0.047 ). Further studies are required to determine the optimal dose and duration of LLE supplementation to confirm the first-stage study results for clinical application.     Abstract: Most existing conventional security mechanisms are insufficient, mainly attributable to their requirements for heavy processing capacity, large protocol message size, and longer round trips, for resource-intensive devices operating in an Internet of Things (IoT) context. These devices necessitate efficient communication and security protocols that are cognizant of the severe resource restrictions regarding energy, computation, communication, and storage. To realize this, the IETF (Internet Engineering Task Force) is currently working towards standardizing an ephemeral key-based lightweight and authenticated key exchange protocol called EDHOC (Ephemeral Diffie–Hellman over COSE). The protocol’s primary purpose is to build an OSCORE (Object Security for Constrained RESTful Environments) security environment by supplying crucial security properties such as secure key exchange, mutual authentication, perfect forward secrecy, and identity protection. EDHOC will most likely dominate IoT security once it becomes a standard. It is, therefore, imperative to inspect the protocol for any security flaw. In this regard, two previous studies have shown different security vulnerabilities of the protocol using formal security verification methods. Yet, both missed the vital security flaws we found in this paper: resource exhaustion and privacy attacks. In finding these vulnerabilities, we leveraged BAN-Logic and AVISPA to formally verify both EDHOC protocol variants. Consequently, we described these security flaws together with the results of the related studies and put forward recommended solutions as part of our future work.

    Abstract: Triboelectric nanogenerator (TENG) is one of the emerging energy harvesting technologies with the potential to be an alternative energy source. Owing to the various advantages of TENG, such as low cost, simple design, and high applicability, several researchers reported wearable TENG devices that can power electronics by harvesting human motion. However, as the human body has limited movement, the existing wearable TENG devices can only generate low power to turn on the electronics. In this study, a flow ring-based TENG (FR-TENG) is fabricated, which can be applied to wearable devices to generate high voltage and current output by including an opposite charging intermediate layer. By the simulation and experimental results, FR-TENG is optimized to generate a high output, that is, peak open-circuit voltage and closed-circuit current of up to 1020 V and 260 mA, respectively, owing to the electrostatic discharge. By these results, sleeve-type wearable FR-TENG is fabricated which can effectively harvest energy from arm movement. The sleeve-type FR-TENG can generate a high output owing to the working mechanism of FR-TENG; the high output was used to turn on 200 LEDs.

    Abstract: Vestibular nucleus (VN) and cerebellar flocculus are known as the core candidates for the neuroplasticity of vestibular system. However, it has been still elusive how to induce the artificial neuroplasticity, especially caused by an electrical stimulation, and assess the neuronal information related with the plasticity. To understand the electrically induced neuroplasticity, the neuronal potentials in VN responding to the repeated electrical stimuli were examined. Galvanic vestibular stimulation (GVS) was applied to excite the neurons in VN, and their activities were measured by an extracellular neural recording technique. Thirty-eight neuronal responses (17 for the regular and 21 for irregular neurons) were recorded and examined the potentials before and after stimulation. Two-third of the population (63.2%, 24/38) modified the potentials under the GVS repetition before stimulation ( p = 0.037 ), and more than half of the population (21/38, 55.3%) changed the potentials after stimulation ( p = 0.209 ). On the other hand, the plasticity-related neuronal modulation was hardly observed in the temporal responses of the neurons. The modification of the active glutamate receptors was also investigated to see if the repeated stimulation changed the number of both types of glutamate receptors, and the results showed that AMPA and NMDA receptors decreased after the repeated stimuli by 28.32 and 16.09%, respectively, implying the modification in the neuronal amplitudes.

    Abstract: Chemoresistance is one of the major causes of therapeutic failure in breast cancer patients. In this study, the mechanism of chemoresistance in human triple-negative breast cancer (TNBC) cells (MDA-MB-231) induced by doxorubicin (DOX) gradient was investigated. These DOX-resistant cells showed higher drug efflux rate, increased anchorage-independent growth when cultured in suspension, and increased tumor-forming ability in nude mice, compared to the wild-type MDA-MB-231 cells. RNA sequencing analysis showed an increase in the expression of genes involved in membrane transport, antiapoptosis, and histone regulation. Kaplan-Meier plot analysis of TNBC patients who underwent preoperative chemotherapy showed that the relapse free survival (RFS) of patients with high HIST1H2BK (histone cluster 1 H2B family member k) expression was significantly lower than that of patients with low HIST1H2BK expression. Quantitative real-time PCR confirmed that the level of HIST1H2BK expression was increased in resistant cells. The cytotoxicity analysis showed that the DOX resistance of resistant cells was reduced by treatment with a histone deacetylase (HDAC) inhibitor. Our results suggest that, in DOX-resistant cells, HIST1H2BK expression can be rapidly induced by the high expression of genes involved in membrane transport, antiapoptosis, and histone regulation. In conclusion, chemoresistance in MDA-MB-231 cells can occur in a relatively short period by DOX gradient via this previously known mechanism of resistance, and DOX resistance is dependent on the specificity of resistant cells to HDAC.
