    Abstract: The black coral Anthipatella wollastoni forms marine animal forests in the mesophotic zone. The spatial extent of black coral forests is not well known in many regions. Due to its protein and chitin skeleton, the coral is difficult to image using acoustic remote sensing techniques compared to corals with carbonate skeletons. Several manufacturers have recently introduced an additional data type to their multibeam echosounders, called “multi-detection,” which provides additional target detections per beam in addition to the primary bottom detection. In this study, we used a Norbit chirp multibeam echosounder in multi-detect mode to acquire up to three targets in each beam in an area of black coral below 45 m depth off the coast of Lanzarote (Canary Islands, Spain). Multi-detect allows features above and below the primary bottom detection to be identified without the need to store and process water-column data. Black coral can be detected by comparing “multi-detection” data with ground truthing by technical divers and underwater cameras. However, the repeatability of the detections is limited and further sensitivity studies are required.

    Abstract: Corals are ecosystem engineers whose tree-like structures give three-dimensional complexity to the habitat. Their population dynamics are affected by recruitment and juvenile survival. Therefore, several defense strategies, such as the formation of hard skeletons and/or spicules, have evolved to protect these vulnerable stages. The family Coralliidae, to which “precious corals” belong, represent an exception in the order Scleralcyonacea, as they form hard CaCO 3 skeletons and small CaCO 3 structures, the sclerites. The skeletogenesis of Corallium species is relatively well documented in adult colonies but remains poorly known in the early stages of the development of new colonies. To shed light on the timing of Corallium rubrum ’s early skeleton formation and the role of sclerites, we focused on the first 4-years of life, applying different techniques, from scanning electron microscopy to synchrotron tomography and laser ablation inductively coupled plasma-mass spectrometry. Our results show that: 1) the first visible sclerites in the primary polyp appear at least 12 days after larval settlement, which is associated with a high CaCO 3 production rate (4.5 ± 2.3 μg of CaCO 3 per day). Furthermore, growth rings are visible in the sclerites, showing that fully matured sclerites grow fast, probably in 3 to 4 days. 2) Sclerites are the only biomineral product in the first year of life of C. rubrum ’s colonies. 3) The evidence of a consolidated axial skeleton, intended as the inner part of the skeleton characteristic of the adult red coral (the medullary zone, MZ), is recorded for the first time in 2-year-old colonies. 4) The annular zone (AZ) around the medullary zone starts forming not before four years after settlement. Thus, primary polyp builds a deformable armor made of only sclerites during the first year. This shelter provides mechanical protection from abrasion and predation to early settled colonies. After two years, settlers are firmly and mineralogically attached to the substratum, which makes them less vulnerable to predation than younger recruits that are not anchored by the skeleton.

    Abstract: Even with high-dose post-transplant cyclophosphamide (PT-Cy) which was initially introduced for graft-versus-host disease (GvHD) prevention in the setting of HLA-haploidentical transplantation, both acute and chronic GvHDs remain a major clinical challenge. Despite improvements in the understanding of the pathogenesis of both acute and chronic GvHDs, reliable biomarkers that predict their onset have yet to be identified. We recently studied the potential correlation between extracellular vesicles (EVs) and the onset of acute (a)GvHD in transplant recipients from related and unrelated donors. In the present study, we further investigated the role of the expression profile of membrane proteins and their microRNA (miRNA) cargo (miRNA100, miRNA155, and miRNA194) in predicting the onset of aGvHD in haploidentical transplant recipients with PT-Cy. Thirty-two consecutive patients were included. We evaluated the expression profile of EVs, by flow cytometry, and their miRNA cargo, by real-time PCR, at baseline, prior, and at different time points following transplant. Using logistic regression and Cox proportional hazard models, a significant association between expression profiles of antigens such as CD146, CD31, CD140a, CD120a, CD26, CD144, and CD30 on EVs, and their miRNA cargo with the onset of aGvHD was observed. Moreover, we also investigated a potential correlation between EV expression profile and cargo with plasma biomarkers (e.g., ST2, sTNFR1, and REG3a) that had been associated with aGVHD previously. This analysis showed that the combination of CD146, sTNFR1, and miR100 or miR194 strongly correlated with the onset of aGvHD (AUROC & gt;0.975). A large prospective multicenter study is currently in progress to validate our findings.
