    Notes: The evolution of cells of the monocyte-macropbage lineage (MML cells) in the spleen of Leishmania donovani (LD) infected BALB/c mice was studied. Spleen cells were fractionated on a discontinuous percoll gradient and adherent cells (AC) were purified from fractionated spleen cells by adherence steps that appeared at the interfaces of 25–35%, 35–40%, 40–45% and 45–50% percoll gradients. The AC were characterized as MML cells on the basis of positive staining for non-specific esterase. Adherent cells that appeared at the interfaces of 25–35% and 40–45% were defined as A and C, respectively, and both of them showed extreme variation in a progressive infection. It was observed that A supported parasite replication whereas C remained refractory when infected with LD in vitro. Furthermore, when A cells and C cells were used as antigen-presenting cells to stimulate mixed population of IFN-γ producing and IL-4 producing T-cells, it was observed that IL-4 and IFN-γ were the predominating cytokine in the T-cell supernatant, respectively. Both A and C were found to be increased hand-in-hand up to 5 months of infection and from then on A decreased and C increased in their numerical strength (A-C reciprocity). The evolution of A-C reciprocity coincided with the gradual reduction in the parasitaemia in the spleen suggesting that this may contribute to the acquisition of anti-leishmania immunity.

    Notes: Antibodies (Abs) raised against the l-glutamate-binding protein (GBP) purified from bovine brain were used to define the possible physiologic activity of GBP in synaptic membranes. Three processes were examined for their sensitivity to the Abs: the excitatory amino acid stimulation of thiocyanate (SCN−) flux, the transport of l-glutamic acid across the synaptic membrane, and the depolarization-induced release of l-glutamate. Only the amino acid-induced changes in ion flux were inhibited by the anti-GBP Abs. The change in membrane potential produced by exposure of synaptic membranes to excitatory amino acids was measured as the increase in the uptake of the lipophilic anion SCN−. The l-glutamate-induced SCN− influx was 40 times more sensitive to inhibition by the anti-GBP Abs than the stimulation of ion flux by kainate, and 60 times more sensitive than that produced by quisqualate. The anti-GBP Abs did not inhibit the activation of ion flux produced by N-methyl-d-aspartate. The inhibition of glutamate-stimulated ion fluxes by the Abs was complete, whereas the inhibition of l-glutamate binding to either the rat or bovine brain GBP was not. The results obtained indicated that although the majority of the anti-GBP Abs were not directed against the glutamate recognition site of the GBP and of presumed synaptic membrane receptors, they were effective in blocking the activation of receptor-associated ion channels. Thus, the GBP may be considered a component of some excitatory amino acid receptor complexes.

    Notes: Trays containing 100g of mushrooms overwrapped with PVC film were held at 12°C. Two 3 mm holes were made on top of the ovetwrap for ventilation and a TyvekR pouch containing sorbitol was placed at the bottom of the tray before storage. Surface moisture content of mushrooms decreased in the presence of sorbitol. Mushrooms packaged with 10g sorbitol had a constant surface moisture content and those packaged with 15g sorbitol had the best overall color. Principal component analysis of Vis-NIR spectra revealed that surface moisture content affected the scattering of incident light and mushroom color. Lowering of the in-package relative humidity did not affect the maturation rate of mushrooms, but reduced bacterial growth, suggesting that improvement in color was probably due to reduced bacterial activity.

    Notes: The effect of O2 concentration, at 2–6% CO2 on shelf-life of fresh mushrooms in modified atmosphere packages (MAP) was studied. For conventional mushrooms optimum in-package O2 was 6% to reduce cap development. Mushrooms irrigated with 0.3% calcium chloride during cropping had a higher rate of maturation and optimum O2 was 2% during storage. At this low O2 concentration, however, storage may be potentially hazardous and is not recommended. Surface moisture of mushrooms significantly increased during storage and may have obscured any beneficial effect of MAP on mushroom color. Precise control of relative humidity in the package is recommended.

    Notes: Induced breeding of Heteropneustes fossilis (Bloch.) was studied in laboratory aquaria. Quantitative and qualitative data on pre-mating, mating, and post-mating behaviour and related parameters were recorded. Among the pairs examined, 83% spawned successfully. Mating behaviour started between 6 and 15 h after administration of pituitary hormones. The number of enfoldings varied from 25 to 350. Over 93% of the released eggs hatched within 21–24 h at temperature 24–31°C.

    Notes: In view of the need for a multidisciplinary approach to study the effect of aging on the oral cavity, “The Veterans Administration Longitudinal and Cross-Sectional Study of Oral Health in Healthy Veterans” or the Dental Longitudinal Study (DLS) was initiated in 1968, as an associated research program of the larger “VA Normative Aging Study.”. The current report presents a review of the literature of masticatory performance and relevant experimental data from the DLS on dentition and age-related changes in performance levels. These data sets demonstrate that in volunteers with natural dentition, there was a slight, but significant, decrease in masticatory performance while dentitions with prostheses showed little change. It appears that when appreciable declines in masticatory function occur, significant alterations also occur in certain perceptual and sensory measures associated with food hedonics and consumption behavior. Generally, these perceptual changes reflect an alteration in taste and texture preferences; phenomena which persist even after appropriate restorative therapy with either removable partial or complete dentures. Information collected in some of our studies suggests that dentate-impaired persons, with and without a dental prosthesis, may self-impose certain dietary restrictions which, in time, can compromise their nutritional status and ultimately place them at health risk. These longitudinal results are attempting to provide a comprehensive description of oral phenomena and represent a unique study of the oral aging process.

    Notes: Since the etiology of vitiligo is still unknown, we searched for some abnormal biochemical parameters, if any, in subjects with vitiligo. Higher urinary excretion of indole metabolites in vitiliginous patients have been noted, in association with higher dioxygenase, superoxide dismutase, and tyrosine aminotransferase activity in their serum. Similar results have also been found in an animal model, Bufo melanostictus, during induced tyrosinase inhibition. Treatment with psoralen can reverse the parameters, except tyrosine aminotransferase, to a normal level. Although psoralens are not the magic bullet for the therapy of vitiligo, they are still being used as a chemotherapeutic agent against vitiligo on a major scale to date. Tryptophan was found to participate in the pathway of melanogenesis, as a precursor as well as a positive regulator of tyrosinase. Its behavior in this regard is much more similar to the conventional substrates tyrosine and dopa (dihydroxyphenylalanine). In consideration of combined participation of tyrosine and tryptophan in the synthesis of melanin and its breakdown, the possible influence of different enzymatic reactions, like mono-oxygenase, dioxygenase, and deamination, has been suggested.

    Notes: The effect of heating film-forming solutions on physical and molecular properties of cast wheat gluten (WG) films was determined. Glycerol-plasticized films were cast from alkaline (pH 10), heat-treated (55,75, or 95°C for 10 min) solutions of WG in aqueous ethanol. Protein solubility (PS) of films in water decreased (P〈0.05) with increasing temperature. Gel permeation chromatograms showed reduced extractability of protein fractions other than ω-gliadins in WG films. This reduced extractability was due to disulfide (S-S) bond formation. SDS-PAGE patterns of native WG and WG film samples suggested increased cross-linking through covalent S-S bonds in films from solutions heated at 75 or 95°C. Water resistance in potential packaging applications of WG edible films could be modified by adjusting heat-treating temperature.
