    Abstract: The mechanism of macrophage migration-inhibition was studied with alveolar macrophages obtained from BCG-sensitized guinea pigs. Sensitivity to migration-inhibition was conferred upon macrophages from normal guinea pigs or upon trypsinized macrophages from sensitized guinea pigs by incubation with either serum or heat eluates prepared from macrophages of BCG-sensitized animals. The migration of passively sensitized macrophages was inhibited only in the presence of specific antigen. Mixtures of PPD and immune serum or macrophage eluates were cytophilic for macrophages but were not demonstrably cytotoxic for mouse L cells. In contrast, supernatants from cultures of antigen-stimulated lymphocytes derived from sensitized guinea pigs were not strongly cytophilic for macrophages but were cytotoxic for L cells. Supernatants from similar cultures of macrophages were less active in inhibiting the migration of normal macrophages. Immunoglobulin was detected in the Sephadex G 200 fractions of macrophage eluates and immune sera which were active in the migration-inhibition test. On the basis of these results it is suggested that macrophage migration-inhibition may involve cytophilic Ab as well as a cytotoxic factor produced by lymphocytes.

    Abstract: Highly purified lymph node and spleen lymphocytes and lung macrophages obtained from normal and tuberculin sensitive guinea pigs were cultured in vitro with tuberculin purified protein derivative (PPD). After 24 hr of incubation, the cell-free culture supernatants were tested for a) mouse L cell cytotoxicity, b) inhibition of macrophage migration, and c) capacity to elicit skin reaction in the skin of non-sensitized guinea pigs. Purified lymphocytes from tuberculin-sensitive guinea pig cultured with PPD yielded culture supernatants which inhibited the growth of L cells. In contrast, purified macrophages from both normal and sensitive animals cultured with PPD released a factor(s) which was cytotoxic for L cells. Macrophage and lymphocyte cytotoxins could not be distinguished by gel filtration, antibody neutralization, heat sensitivity or by the biologic assays employed.

    Abstract: A study has been made of the time course and dose-response curves of lysozyme, acid phosphatase and cathepsin in rabbit alveolar macrophages following the injection of bacillus Calmette-Guérin (BCG) by the intratracheal route. Significant elevation in the levels of these acid hydrolases was observed within 3 to 7 days following the injection of BCG. The increase in enzyme levels was observed to be a dose-related response.
