Zusammenfassung Die Bestimmung der SH-Gruppen in Proteinen bereitet häufig Schwierigkeiten, die in der Proteinstruktur („maskierte” und langsam reagierende SH-Gruppen), in der mangelnden Spezifität der Reagentien oder in äußeren Einflüssen begründet liegen. Die möglichen Gründe für die unterschiedliche Reaktionsfähigkeit der Protein-SH-Gruppen und deren Anderung durch Denaturierung werden diskutiert (Sterische Hinderung, Nachbargruppeneffekte, hydrophobe Bindungen, Thiazolinhypothese, Isothiohamstoffreste, Disulfidspaltung, säurelabile Sulfide). Der Grad der „Maskierung” von Protein-SH-Gruppen äußert sich gegenüber verschiedenen Reagentien unterschiedlich. Obwohl nach Denaturierung häufig alle SH-Gruppen zugänglich werden, ist die Denaturierung nicht generell notwendig oder empfehlenswert. Die Spezifität von AgNO3 für SH-Gruppen ist auf Grund der Erfahrungen mit niedermolekularen SH-Verbindungen angezweifelt worden (Komplexbildung). Die Übertragung auf Proteine ist jedoch nicht gerechtfertigt. Untersuchungen an Proteinen mit definiertem SH- und SS-Gehalt zeigten im Gegenteil, däß unter den Bedingungen der amperometrischen Titration (Trispuffer pH 7,4) Ag+-Ionen als spezifisches SH-Reagens angesehen werden können.

Summary In general it is difficult to measure SH groups in proteins. The problems are due to the protein structure (hidden or slowly reacting SH groups) or to the low specifity of the reagents or the environmental influences like oxidation. The reason of the different reactivity of SH groups in proteins is discussed: Steric hindrance, combined functional groups, hydrophobic environment, thiazolin hypothesis, isothiuronium residues, splitting of disulphide groups and acid-labile sulphides. The extent of the masking of SH groups in proteins varies with different reagents used. The denaturation which makes usually all SH groups accessible to reagents is not always necessary and recommendable. Doubts have been thrown on the specifity of AgNO3 for SH groups because of some experiences with SH compounds of low molecular weight (formation of complexes). But it is not justified to transfer the conditions of small SH compounds to proteins. Studies on proteins with defined SH and SS content showed in the contrary that silver ions under the conditions of the amperometric titration (tris buffer pH 7.4) may be used as a specific SH reagent.
