    Abstract: Interferon regulatory factors (IRFs) have been confirmed as vital transcription factors for the immune system, which play a certain role in anti-infection defense, immune regulation, hematopoietic cell development, and maturation of the immune system. In this study, the cDNA of Interferon regulatory factor 5 (IRF5) was cloned from black-spotted frog, Pelophylax nigromaculatus , and termed PnIRF5. The results indicated, the full-length cDNA of PnIRF5 was 2090 bp, and a putative protein of 504 amino acids was encoded. The results of protein domain prediction suggested that PnIRF5 exhibited a DNA-binding domain (DBD), a middle region (MR), an IRF association domain (IAD), a virus activated domain (VAD), as well as two nuclear localization signals (NLSs). Phylogenetic analysis clustered PnIRF5 into the amphibian IRF5 subgroup in vertebrate IRF5 group. The results of qPCR show that PnIRF5 mRNA was expressed in all examined tissues, with higher levels were identified in the kidney, intestine, and lung. The gene expression of PnIRF5 was analyzed in kidney, spleen, and liver of black-spotted frog after challenged with Elizabethkingia miricola , and the maximum expression level of PnIRF5 in liver, spleen, and kidney were 1.80, 2.33 and 2.88 times that at 0 days post challenged, respectively. As indicated by the results, the PnIRF5 protein may take on critical significance in protecting black-spotted frog from E. miricola infection, and they may play a certain role in the immune response of black-spotted frog against bacteria.

    Abstract: The effects of dietary arachidonic acid (ARA) levels on growth performance, whole-body proximate composition, digestive enzyme activities, and gut morphology were studied in juvenile golden pompano Trachinotus ovatus. Six diets were formulated with six levels of ARA. Fish were fed twice daily to apparent satiation for 56 days (8 weeks). Weight gain (WG), specific growth rate (SGR), protein efficiency ratio (PER), feed conversion ratio (FCR), and viscerosomatic index (VSI) were significantly affected by dietary ARA levels (P 〈 0.05). WG, SGR and PER increased with the increasing levels of ARA (P 〈 0.05), and thereafter slightly declined (P 〉 0.05); they were highest in fish fed the diet with 0.51% ARA and the lowest in fish fed the diet containing 0.15% ARA. Linear regression analysis on SGR indicated that the recommended optimum dietary ARA level for optimal growth of juvenile golden pompano was 0.53%. Whole body protein significantly declined when dietary ARA levels increased from 0.15% to 0.88% (P 〈 0.05), and were lowest in fish fed the diet containing 0.88% ARA. The whole body lipid content showed an opposite trend compared with whole body protein. Pepsin activities showed no significant differences among treatments (P 〉 0.05), while lipase activities of fish were significantly influenced by dietary ARA levels (P 〈 0.05). The number of goblet cells and intestinal villus length increased with increasing levels of ARA from 0.15% to 0.51% (P 〈 0.05), and decreased thereafter. Goblet cells of fish fed diets with 0.36%, 0.51%, 0.71% ARA were higher than in the other groups (P 〈 0.05).

    Abstract: A novel silver cluster ( SD/Ag48a ) co‐protected by p ‐MePhS − and CF 3 COO − ligands with an in‐situ generated unprecedented [Mo 8 O 28 ] 8− template has been synthesized under solvothermal condition. Single‐crystal X‐ray diffraction (SCXRD) and electrospray mass spectrometry (ESI‐MS) were used to determine the atom‐precise structure and solution behavior, respectively. Interestingly, the rare [Mo 8 O 28 ] 8− ion observed in the center of SD/Ag48a consists of 8 edge‐shared {MoO 6 } octahedra or can be deemed as three {Mo 4 O 4 } cubes fused together by sharing faces. The UV‐Vis spetcrum exhibits that a high‐energy (HE) wide absorption centered at ca. 365 nm, which can be assinged to intraligand (IL) π→π* transition absorption, whereas the low‐energy (LE) shoulder peak centered at ca. 500 nm can be ascribed to ligand‐to‐metal‐charge‐transfer (LMCT, S 3p→Ag 5 s) transition.

    Abstract: The effects of polysaccharides, saponins, and water decoction of the Chinese herb Astragalus membranaceus on the immune response of spotted maigre (Nibea albiflora) were investigated. Fish with an average initial weight of 49.6±5.5 g were fed a diet containing 2% extract for four weeks. The lysozyme and phagocytic activities of the fish were determined and compared with those of control fish fed an unsupplemented feed on days 0, 3, 7, 14, 21, and 28. Both activities were significantly higher in groups fed the Astragalus extract than in the control group (p 〈 0.01) but there were no differences between the three Astragalus-fed groups (p 〉 0.05). Most important, the Astragalus-fed groups were significantly protected against Vibrio vulnificus challenge compared to the control group (p 〈 0.01). Our results indicatd that polysaccharides and saponins are main active components of Astragalus extracts and can enhance the non-specific immune response in spotted maigre.

    Abstract: Hypoxia tolerance and adaptive regulation are important for aquatic animals, especially for species with poor mobility, such as most bivalves. Previous studies have confirmed that the blood clam Scapharca broughtonii has strong hypoxia resistance. However, the molecular mechanism supporting its hypoxic tolerance is still largely limited. To further screen the genes and their potential regulation of hypoxia tolerance, the transcriptome changes of S. broughtonii after acute hypoxic stress were explored by RNA sequencing. In this study, the average value of Q30 is 92.89%, indicating that the quality of sequencing is relatively high. The Unigenes obtained were annotated using four databases, namely Interpo, KEGG, Swisspro and TrEMBL. The annotation rates in these four databases were 71.82%, 75.95%, 92.98%, and 79.26%, respectively. And also, there were 649 DEGs in group B (dissolved oxygen (DO) of 2.5 mg/L) compared with group D (DO of 7.5 mg/L), among which 252 were up-regulated, and 397 were down-regulated. There were 965 DEGs in group A (DO of 0.5 mg/L), 2.5 mg/L, and 7.5 mg/L, compared with group B, among which 530 were up-regulated, and 435 were down-regulated. Meanwhile, there were 2,040 DEGs in group A compared with group D, among which 901 were up-regulated, and 1,139 were down-regulated. The main metabolic-related pathways of KEGG enriched in this study included Insulin secretion, Insulin signaling pathway, MAPK signal transduction pathway, and PPAR signaling pathway. These pathways may be critical metabolic pathways to solve energy demand and rebuild energy balance in S. broughtonii under hypoxic conditions. This study preliminarily clarified the response of S. broughtonii to hypoxia stress on the molecular levels, providing a reference for the following study on the response laws of related genes and pathways under environmental stress of S. broughtonii.

    Abstract: A 60-day study was conducted to investigate the effects of dietary lipid levels on growth, whole body composition and digestive enzyme activity of bighead carp (Aristichthys nobilis) with initial average weight (3.28 ± 0.11) g. Six isonitrogenous (40.25% crude protein) diets were formulated to contain graded contents of lipid (4.72%, 7.31%, 11.02%, 14.90%, 17.14%, and 19.37%) using corn oil and soybean oil as main lipid source. The results showed that the maximum weight gain (WG) and specific growth rate (SGR) were recorded at 11.02% dietary lipid level. Significant alterations were demonstrated on whole-body composition by different dietary lipid levels. Crude lipid content of whole body increased in parallel with the increase of dietary lipid levels. Fish fed diet with 11.02% lipid level showed lowest moisture and ash content. The activities of amylase and protease initially increased before decreasing, and the highest activities of amylase and protease were obtained in individuals fed the 11.02% lipid content diet. There were no significant differences in condition factor (CF) and hepatosomatic index (HSI) (P 〉 0.05). Based on the polynomial regression analysis of WG and SGR, this study suggested that the optimum dietary lipid requirement of juvenile bighead carp was estimated to be 7.77%~8.31%.

    Abstract: The spotted sea bass (Lateolabrax maculatus) is an economically valuable cultured fish species in China. In this study, 20 novel polymorphic microsatellite loci of L. maculatus were isolated from genomic data and characterized using 40 wild individuals. The number of alleles and the effective number of alleles ranged from 2 to 12 (average of 5.1000) and from 1.180 to 8.000 (average of 3.3097). The observed and expected heterozygosities ranged from 0.083 to 0.875 (average of 0.4405) and from 0.153 to 0.875 (average of 0.5633), respectively. Deviation from the Hardy-Weinberg equilibrium was observed in 11 loci (P 〈 0.05). Polymorphism information content ranged from 0.141 to 0.862 (average of 0.5265) and most loci were heterozygous. Cross-amplification trials in two cryptic congeneric species, the largemouth bass, Micropterus salmoides, and the barramundi, Lates calcarifer , achieved successful amplification of 16 primers. The microsatellite markers developed in this study could be used for research on genetic breeding of L. maculatus and genetic relationships among tested taxa.

    Abstract: Pond water with algae blooms was collected, and maintained in the dark over a 2-day period without nutritional supplements. The physico-chemical parameters and the density of algae were measured. The concentration of NH 4+-N, DIN and PO4–P in water in darkness significantly increased, and the density of algae decreased significantly. Isolated water was exposed to three bio-remediation water treatments: addition of green algae, Bacillus subtilis and photosynthetic bacteria. After remediation, the concentrations of NH4+ -N, NO3- -N, DIN and PO4- -P in the three treatment groups were significantly lower than the control group. No difference was observed in the concentration of NO2–N between the control group and the photosynthetic bacteria group. Results from all treatments showed water quality deterioration along with increase in nitrogen and phosphorus concentrations. Bio-remediation methods can be effectively adapted in shrimp culture after algae decay, and one of the most promising biological remediation strategies is the use of green algae.

    Abstract: Antibacterial properties of curcumin from turmeric (Curcuma longa) and enrofloxacin against Aeromonas hydrophila were assayed. The minimum inhibitory concentration (MIC) values of curcumin and enrofloxacin against A. hydrophila were found to be 100ug/ml and 9.375ug/ml, respectively. To realize the mechanisms of action of curcumin against A. hydrophila, we researched the antibacterial activity and bacterial membrane permeability of A. hydrophila cells treated with curcumin or enrofloxacin. All results elucidated that curcumin increased membrane permeabilization and caused leakage of intracellular contents, while its role was not as good as enrofloxacin. Moreover, a synergistic effect was shown between curcumin and enrofloxacin. The present study suggests that curcumin extracted from turmeric has the potential to be used as an antimicrobial for the control of A. hydrophila.

    Abstract: Spotted seabass ( Lateolabrax maculatus ) is a popular food fish in Asian countries. Not only does the fish tasted well, but it is also widely utilized as nutraceuticals. However, the knowledge about the composition of the fish is lack. In this work, the proximate composition of spotted seabass flesh was determined. The content of proximate composition (moisture, proteins, fat, and ash) of spotted seabass flesh was fell within the range of other reported fish species, while it was a high-quality food fish with low oil-very high protein. The ratio of essential amino acids (EAAs) to nonessential amino acids (NEAAs) was 0.74, and the amount of EAAs accounted for approximately 42.34% of the total amino acids. Unsaturated fatty acids composed the majority of the fatty acid components, with linoleic acid was the predominate one. The content of docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA) were abundant in spotted seabass flesh. The proximate composition of the dorsal, ventral, lateral, and tail flesh was similar, except for the ventral flesh exhibited a higher content of fatty acids and lower content of moisture. Our research will be helpful for the general population to know the nutritional traits of spotted seabass.
